Identification of common and distinct origins of human serum and breastmilk IgA1 by mass spectrometry-based clonal profiling

The most abundant immunoglobulin present in the human body is IgA. It has the highest concentrations at the mucosal lining and in biofluids such as milk and is the second most abundant class of antibodies in serum. We assessed the structural diversity and clonal repertoire of IgA1-containing molecular assemblies longitudinally in human serum and milk from three donors using a mass spectrometry-based approach. IgA-containing molecules purified from serum or milk were assessed by the release and subsequent analysis of their Fab fragments. Our data revealed that serum IgA1 consists of two distinct structural populations, namely monomeric IgA1 (∼80%) and dimeric joining (J-) chain coupled IgA1 (∼20%). Also, we confirmed that IgA1 in milk is present solely as secretory (S)IgA, consisting of two (∼50%), three (∼33%) or four (∼17%) IgA1 molecules assembled with a J-chain and secretory component (SC). Interestingly, the serum and milk IgA1-Fab repertoires were distinct between monomeric, and J-chain coupled dimeric IgA1. The serum dimeric J-chain coupled IgA1 repertoire contained several abundant clones also observed in the milk IgA1 repertoire. The latter repertoire had little to no overlap with the serum monomeric IgA1 repertoire. This suggests that human IgA1s have (at least) two distinct origins;one of these produces dimeric J-chain coupled IgA1 molecules, shared in human serum and milk, and another produces monomeric IgA1 ending up exclusively in serum.

Plant responses to variable timing of aboveground clipping and belowground herbivory depend on plant age

Aims Plants use different types of responses such as tolerance and induced defense to mitigate the effects of *** direc-tion and magnitude of both these plant responses can vary with plant ***,most studies have focused on aboveground herbivory,whereas important feeding occurs ***,we tested the hypothesis that plant tolerance and defense following shoot damage or root herbivory depends on plant *** In order to test our hypothesis,we exposed the perennial grass spe-cies Holcus lanatus to defoliation and root nematode inoculation at three growth stages(young,intermediate and old plants),and exam-ined responses of plant traits related to tolerance(regrowth follow-ing defoliation)and defense(leaf and root nitrogen and phenolics).Important Findings Defoliation overall reduced plant shoot and root biomass as well as foliar concentrations of phenolics regardless of plant age at *** contrast,defoliation increased foliar N concentrations,but only when defoliation occurred at intermediate and old plant *** with root-feeding nematodes reduced root N concentra-tions after a prolonged period of growth,but only when nematodes had been inoculated when plants were *** relative shoot re-growth rate of plants increased immediately after defoliation but this was independent of the plant age at which defoliation occurred,*** not stronger in plants that were defoliated at a more advanced age,as ***,relative root growth rates increased shortly after defoliation,but this was only observed for plants defoli-ated when they were *** conclude that plant responses to aboveground and belowground herbivory in traits related to both defense and tolerance are affected by plant age,but do not gener-ally change with plant age.

Background: Acute acoustic trauma (AAT) is an acute hearing impairment caused by intense noiseimpact. The current management strategy for AAT with substantial hearing loss in the Dutch militaryis the combination therapy with corticosteroids and hyperbaric oxygen therapy (HBOT). In a previousstudy, early initiation of the combination therapy was associated with better outcomes. Therefore, weperformed a new analysis to assess the difference in hearing outcome between patients in whomcombination therapy was started within two days, versus after more than two ***: A retrospective analysis was performed on military patients diagnosed with AAT with substantial hearing loss who presented between February 2018 and march 2020. Absolute and relativehearing improvement between first and last audiograms were calculated for all affected frequencies(defined as loss of 20 dB on initial audiogram). We also determined the amount of patients whorecovered to the level of Dutch military requirement, and performed speech discrimination ***: In this analysis, 30 male patients (49 ears) with AAT were included. The median age was 24.5years (IQR 23e29). The median time to initiation of therapy with corticosteroids and HBOT were one andtwo days, respectively. HBOT was started within two days in 31 ears, and after more than two days in 18ears. The mean absolute and relative hearing gains were 18.8 dB (SD 14.6) and 46.8% (SD 31.3) on allaffected frequencies. The 100% discrimination/speech perception level improved from 64.0 dB to 51.7 dB(gain 12.3 dB ± 14.1). There was significantly more improvement in absolute and relative hearingimprovement when HBOT was started in 2 days, compared to >2 ***: Our analysis shows results in favor of early initiation ( 2 days) of the combination treatmentof HBOT and corticosteroids in patients with AAT.

The pygmy marmoset,the smallest of the anthropoid primates,has a broad distribution in Western *** studies using molecular and morphological data have identified two distinct species separated by the Napo and Solimoes-Amazonas ***,reconciling this new biological evidence with current taxonomy,i.e.,two subspecies,Cebuella pygmaea pygmaea(Spix,1823)and Cebuella pygmaea niveiventris(Lönnberg,1940),was problematic given the uncertainty as to whether Spix’s pygmy marmoset(Cebuella pygmaea pygmaea)was collected north or south of the Napo and Solimoes-Amazonas rivers,making it unclear to which of the two newly revealed species the name pygmaea would ***,we present the first molecular data from Spix’s type specimen of Cebuella pygmaea,as well as novel mitochondrial genomes from modern pygmy marmosets sampled near the type locality(Tabatinga)on both sides of the *** these data,we can confirm the correct names of the two species identified,i.e.,*** for animals north of the Napo and Solimoes-Amazonas rivers and *** for animals south of these two *** analyses of the novel genetic data placed into the context of cytochrome b gene sequences from across the range of pygmy marmosets further led us to reevaluate the geographical distribution for the two Cebuella *** dated the split of these two species to 2.54 million years *** discuss additional,more recent,subdivisions within each lineage,as well as potential contact zones between the two species in the headwaters of these rivers.
