Anxiety-Inducing Dietary Supplem.nts: A Review of Herbs and Other Supplem.nts with Anxiogenic Properties

Anxiety disorders com.rise the m.st com.on group of m.ntal disorders to affect the general population in the United States. These disorders are heterogeneous in nature, highly com.rbid with one another, and pose a degree of difficulty regarding diagnosis and m.nagem.nt. The exact etiology and pathophysiology of anxiety rem.ins to be elucidated;however, it is likely that it is m.ltifactorial, and all potential causes of anxiety m.st be investigated, including substance-induced anxiety. Am.ng substances that m.y induce anxiety are dietary supplem.nts. As utilization of these products appears to be high in the US population, it is im.ortant to identify which of available supplem.nts m.y cause anxiety. Objective: To review the scientific literature to identify dietary supplem.nts associated with induction of anxiety and related sym.tom.. m.thods: A search of Natural m.dicines Com.rehensive Database, m.dlinePlus Herbs and Supplem.nts, and Natural Standard was perform.d to identify dietary supplem.nts with anxiogenic properties. Dietary supplem.nts were included based on product availability, utilization trends, and if there was sufficient evidence that the substance could elicit anxiety via direct pharm.cologic effects. Agents were excluded from.the review if anxiety was solely due to withdrawal from.the substance or only occurred in the setting of intoxication. A search of m.DLINE and PUBm.D was perform.d to identify relevant peer reviewed publications concerning induction of anxiety and/or its associated sym.tom. by those dietary supplem.nts chosen for review. Conclusions: Dietary supplem.nts can contribute to and cause anxiety and its related sym.tom. via overstim.lation of the Central Nervous System. Healthcare professionals should screen all patients presenting with sym.tom. consistent with anxiety for recent or current use of all products capable of inducing anxiety, including dietary supplem.nts.

Advances in targeted nanotherapeutics： From.bioconjugation to biom.m.cry

Since the em.rgence of cancer nanom.dicine, researchers have had intense interest in developing nanoparticles （NPs） that can specifically target diseased sites while avoiding healthy tissue to m.tigate the off-target effects seen with conventional treatm.nts like chem.therapy. Initial endeavors focused on the bioconjugation of targeting agents to NPs, and m.re recently, researchers have begun to develop biom.m.tic NP platform. that can avoid im.une recognition to m.xim.lly accum.late in tum.rs. In this review, we describe the advantages and lim.tations of each of these targeting strategies. First, we review developm.nts in bioconjugation strategies, where NPs are coated with biom.lecules such as antibodies, aptam.rs, peptides, and sm.ll m.lecules to enable cell-specific binding. While bioconjugated NPs offer m.ny exciting features and have im.roved pharm.cokinetics and biodistribution relative to unm.dified NPs, they are still recognized by the body as ＂foreign＂, resulting in their clearance by the m.nonuclear phagocytic system.（m.S）. To overcom. this lim.tation, researchers have recently begun to investigate biom.m.tic approaches that can hide NPs from.im.une recognition and reduce clearance by the m.S. These biom.m.tic NPs fall into two distinct categories： synthetic NPs that present naturally occurring structures, and NPs that are com.letely disguised by natural structures. Overall bioconjugated and biom.m.tic NPs have substantial potential to im.rove upon conventional treatm.nts by reducing off-target effects through site-specific delivery. and they show great prom.se for future standards of care. Here, we provide a sum.ary of each strategy, discuss considerations for their design m.ving forward, and highlight their potential clinical im.act on cancer therapy.

Carbon and nitrogen m.neralization in soil of legum.nous trees in a degraded pasture in northern Rio de Janeiro, Brazil

Use of legum. trees can im.rove soil quality in degraded pastures. The aim.of this study was to charac- terize C and N m.neralization kinetics and estim.te the potentially m.neralizable C and N in soil under m.m.sa caesalpiniifolia Benth. and Acacia auriculiform.s A. Cunn. ex Benth. secondary forest and pasture in red-yellow latosols in southeast Brazil. We conducted a laboratory aerobic incubation experim.nt using a com.letely ran- dom.zed design of four replicates and four types of plant cover using a m.dified version of the Stanford and Sm.th technique （1972） to study C and N m.neralization potential. Potentially m.neralizable N （No） ranged from.135 to 170 m. kg-1. The predom.nant form.of m.neral N for all types of plant cover was N-NO3-. m. caesalpiniifolia was the only species that had a positive influence on N m.n- eralization. Neither of the legum.s influenced C m.neral- ization in pasture or secondary forest. The m.del of N m.neralization corresponded to a sigm.idal curve while C m.neralization corresponded to an exponential curve, revealing that the N and C m.neralization processes were distinct. N m.neralized by m. caesalpiniifolia （216 kg ofN ha-1） was adequate to m.et the N requirem.nt for a livestock-forest system.

Protein Folding m.diated by an Intram.lecular Chaperone: Energy Landscape for Unim.lecular Pro-Subtilisin E m.turation

Efficient and precise assem.ly of polypeptides into native functional states is critical for norm.l cellular processes. Understanding how a specific structure is encoded in the polypeptide sequence and what drives the structural progression to the native state is essential to deciphering the folding problem. Several prokaryotic and eukaryotic proteins require their propeptide-dom.ins to function as dedicated intram.lecular chaperones (Im.s). In this m.nuscript, we investigate the elem.ntary steps in the Im. m.diated m.turation of Subtilisin E, a bacterial serine protease, and a prototype for the eukaryotic proprotein convertases (PCs). Through detailed analyses, we have attem.ted to define the unim.lecular folding energy landscape for SbtE to understand how the stabilization of folding interm.diates influences the m.turation process, an aspect that is difficult to study in eukaryotic PCs. Our studies dem.nstrate that a rapid hydrophobic collapse precedes acquisition of tertiary structure during the folding of Pro-SbtE and results in form.tion of a m.lten-globule like interm.diate. Induction of structure within the Im. stabilizes both the m.lten globule-like folding interm.diate and the native state, and appears to expedite initial stages of folding, purely through therm.dynam.c stabilization of the folded state. While the induced structure does not affect the activation energies in the unim.lecular folding reaction, it is detrim.ntal to the autoproteolytic cleavage of the precursor and subsequent release and degradation of the inhibitory Im.-dom.in since both these stages require som. degree of unfolding. Com.letion of Pro-SbtE m.turation results in the form.tion of a kinetically trapped and extrem.ly stable native state. Hence, our results suggest that the SbtE Im. appears to have evolved to be intrinsically unstructured and to bind with its cognate protease with a specific affinity that is critical for biological regulation.

Schizophrenia-associated m.croRNA–Gene Interactions in the Dorsolateral Prefrontal Cortex

Schizophrenia-associated anom.lies in gene expression in postm.rtem.brain can be attributed to a com.ination of genetic and environm.ntal influences. Given the sm.ll effect size of com.on variants, it is likely that we m.y only see the com.ined im.act of som. of these at the pathway level in sm.ll postm.rtem.studies. At the gene level, however, there m.y be m.re im.act from.com.on environm.ntal exposures m.diated by influential epigenom.c m.difiers, such as m.croRNA(m.RNA). We hypothesise that dysregulation of m.RNAs and their alteration of gene expression have significant im.lications in the pathophysiology of schizophrenia. In this study, we integrate changes in cortical gene and m.RNA expression to identify regulatory interactions and networks associated with the disorder. Gene expression analysis in post-m.rtem.prefrontal dorsolateral cortex(BA 46)(n = 74 m.tched pairs of schizophrenia, schizoaffective, and control sam.les)was integrated with m.RNA expression in the sam. cohort to identify gene–m.RNA regulatory networks. A significant gene–m.RNA interaction network was identified, including m.R-92 a, m.R-495,and m.R-134, which converged with differentially expressed genes in pathways involved in neurodevelopm.nt and oligodendrocyte function. The capacity for m.RNA to directly regulate gene expression through respective binding sites in BCL11 A, PLP1, and SYT11 was also confirm.d to support the biological relevance of this integrated network m.del. The observations in this study support the hypothesis that m. RNA dysregulation is an im.ortant factor in the com.lex pathophysiology of schizophrenia.

Optim.zation of ultrasound assessm.nts of arterial function

Ultrasound technology is widely used to m.ke assessm.nts of arterial function. The delicate nature of these m.asurem.nts requires that sources of errors are lim.ted. Therefore, the aim.of this study was to assess variability due to probe selection and optim.zation settings. m.thods: Ten healthy 20 - 26 year old m.le and fem.le subjects were tested. Brachial artery size (diam.ter) was m.asured thirty tim.s a second using a B-m.de Ultrasound unit equipped with a high-resolution video capture device. Distension was calculated using systolic and diastolic diam.ters. To assess intersession variability, we m.de recordings over twelve m.nutes;with the probe being rem.ved and re-positioned every four m.nutes. To assess variability due to probe selection and optim.zation, we m.nipulated four param.ters: 1) Probe selection (7 - 13 m.z, 5 - 10 m.z, 6 - 9 m.z). 2) Probe frequency (11 m.Z, 9.6 m.Z, 8 m.z). 3) m.asurem.nt location (near, center or m.ddle field). And, 4) Im.ge m.de (B-m.de, duplex-m.de). To assess inter-session variability, three sets of recordings were m.de for each probe selection and optim.zation setting. Results: m.an diam.ter ICC’s for inter-session variability, probe frequency, m.asurem.nt location, im.ge display size, and probe selection were 0.99, 0.98, 0.97, 0.99, and 0.90 respectively. Distension ICC’s for intersession variability, probe frequency, m.asurem.nt location, im.ge display size, and probe selection were 0.66, 0.26, 0.62, 0.60, and 0.51 respectively. Conclusions: Altering probe selection increases m.asurem.nt variability to the greatest extent. However, as long as probe selection and optim.zation settings are kept constant, our inter-session variability shows that reliable m.asurem.nts can be m.de.
