Platelets are small, versatile blood cells that are critical for hemostasis/thrombosis. Local platelet accumulation is a known contributor to proinflammation in various disease states. However, the anti-inflammatory/immunosuppressive potential of platelets has been poorly explored. Here, we uncovered, unexpectedly, desialylated platelets (dPLTs) down-regulated immune responses against both platelet-associated and -independent antigen challenges. Utilizing multispectral photoacoustic tomography, we tracked dPLT trafficking to gut vasculature and an exclusive Kupffer cell-mediated dPLT clearance in the liver, a process that we identified to be synergistically dependent on platelet glycoprotein Ibα and hepatic Ashwell–Morell receptor. Mechanistically, Kupffer cell clearance of dPLT potentiated a systemic immunosuppressive state with increased anti-inflammatory cytokines and circulating Cd4+ regulatory T cells, abolishable by Kupffer cell depletion. Last, in a clinically relevant model of hemophilia A, presensitization with dPLT attenuated anti-factor VIII antibody production after factor VIII ( infusion. As platelet desialylation commonly occurs in daily-aged and activated platelets, these findings open new avenues toward understanding immune homeostasis and potentiate the therapeutic potential of dPLT and engineered dPLT transfusions in controlling autoimmune and alloimmune diseases.

The present study focuses on the formulation of new composite consisting of plaster and raffia vinifera particle (RVP) with the purpose to reducing energy consumption. The aim of this study is to test this new compound as an insulating eco-material in building in a tropical climate. The composites samples were developed by mixing plaster with raffia vinifera particles (RVP) using three different sizes (1.6 mm, 2.5 mm and 4 mm). The effects of four different RVP incorporations rates (i.e., 0wt%, 5wt%;10wt%;15wt%) on physical, thermal, mechanicals properties of the composites were investigated. In addition, the use of the raffia vinifera particles and plaster based composite material as building envelopes thermal insulation material is studied by the habitable cell thermal behavior instrumentation. The results indicate that the incorporation of raffia vinifera particle leads to improve the new composite physical, mechanical and thermal properties. And the parametric analysis reveals that the sampling rate and the size of raffia vinifera particles are the most decisive factor to impact these properties, and to decreases in the thermal conductivity which leads to an improvement to the thermal resistance and energy savings. The best improvement of plaster composite was obtained at the raffia vinifera particles size between 2.5 and 4.0 mm loading of 5wt% (C95P5R) with a good ratio of thermo-physical-mechanical properties. Additionally, the habitable cell experimental thermal behavior, with the new raffia vinifera particles and plaster-based composite as thermal insulating material for building walls, gives an average damping of 4°C and 5.8°C in the insulated house interior environment respectively for cold and hot cases compared to the outside environment and the uninsulated house interior environment. The current study highlights that this mixture gives the new composite thermal insulation properties applicable in the eco-construction of habitats in tropical environments

Sex development anomalies represent a group of congenital pathologies in which chromosomal, gonadal and anatomical sex differentiation is atypical. The aim of our study was to use molecular biology techniques to elucidate sex in cases of anomalies of sexual differentiation in Burkina Faso. This cross-sectional study took place from March 2023 to June 2023. Oral and blood samples were collected respectively using sterile swabs and stored on Swab Kits and NUCLEIcardTM (https://***/product-ranges/nucleic-card/) were used to determine gonosome profiles. Extraction was carried out using the dNA Swap solution Kit and the dNA IQ System Kit, and a PowerPlex? 21 kit (Promega) for amplification. The Applied Biosystems 9700 thermal cycler was used for PCR followed by 36 cm capillary electrophoresis in the Applied Biosystems 3130 prism sequencer. Sequence files were analyzed using GeneMapper IdX v. 3.2 software. Seven (07) patients were registered during the study period. There were 4 cases of XX dSd or 46, XX dSd and 3 cases of XY dSd or 46, XY dSd. The median age of our patients was 16 years. Civilian sex was male in 4 cases and female in 3. The most frequent reason for consultation was micropenis in 3 cases, followed by primary amenorrhea and sex ambiguity. There were 03 cases of discrepancy between genetic sex and civil sex. The accessibility of molecular diagnosis is little known to clinicians. XX dSds or 46, XX dSds were the most frequent (4/7) in our study. The problem facing this situation is early diagnosis to help prevent complications in Burkina Faso.

The COVId-19 pandemic caused by SARS-CoV-2 virus is an ongoing global health *** cases of COVId-19 and the rare cases of COVId-19 vaccine-induced-thrombotic-thrombocytopenia(VITT)are both associated with thrombosis and thrombocytopenia;however,the underlying mechanisms remain inadequately *** infection and vaccination utilize the spike protein receptor-binding domain(RBd)of *** found that intravenous injection of recombinant RBd caused significant platelet clearance in *** investigation revealed the RBd could bind platelets,cause platelet activation,and potentiate platelet aggregation,which was exacerbated in the delta and Kappa *** RBd–platelet interaction was partially dependent on theβ3 integrin as binding was significantly reduced inβ3−/−***,RBd binding to human and mouse platelets was significantly reduced with relatedαIIbβ3 antagonists and mutation of the RGd(arginine-glycine-aspartate)integrin binding motif to RGE(arginine-glycine-glutamate).We developed anti-RBd polyclonal and several monoclonal antibodies(mAbs)and identified 4F2 and 4H12 for their potent dual inhibition of RBd-induced platelet activation,aggregation,and clearance in vivo,and SARS-CoV-2 infection and replication in Vero E6 *** data show that the RBd can bind platelets partially thoughαIIbβ3 and induce platelet activation and clearance,which may contribute to thrombosis and thrombocytopenia observed in COVId-19 and *** newly developed mAbs 4F2 and 4H12 have potential not only for diagnosis of SARS-CoV-2 virus antigen but also importantly for therapy against COVId-19.

Epidemiological studies support a general inverse association between the risk of cancer development and Parkinson’s disease(Pd).In recent years however,increasing amount of eclectic evidence points to a positive association between Pd and cancers through different temporal analyses and ethnic *** positive association has been supported by several common genetic mutations in SNCA,PARK2,PARK8,ATM,p53,PTEN,and MC1R resulting in cellular changes such as mitochondrial dysfunction,aberrant protein aggregation,and cell cycle ***,we review the epidemiological and biological advances of the past decade in the association between Pd and cancers to offer insight on the recent and sometimes contradictory findings.

Seagrass ecosystems worldwide have been declining, leading to a decrease in associated fish populations, especially those with low mobility such as syngnathids （pipefish and seahorses）. This two-year pilot study investigated seasonal patterns in density, growth, site fidelity, and population dynamics of Tampa Bay （FL） syngnathid fishes at a site adjacent to two marinas under construction. Using a modified mark-recapture technique, fish were collected periodically from three closely located sites that varied in seagrass species （Thalassia spp., Syringodium spp., and mixed-grass sites） and their distance from open water, but had consistent physical/chemical environmental characteristics. Fish were marked, photographed for body size and gender measurements, and released the same day at the capture site. Of the 5695 individuals surveyed, 49 individuals were recaptured, indicating a large, flexible population. Population density peaks were observed in July of both years, with low densities in late winter and late summer. Spatially, syngnathid densities were highest closest to the mouth of the bay and lowest near the shoreline. Seven species of syngnathid fishes were observed, and species-specific patterns of seagrass use emerged during the study. However, only two species, Syngnathus scovelli and Hippoeampus zosterae, were observed at high frequencies. For these two species, body size decreased across the study period, but while S. scovelli＇s population density decreased, H. zosterae＇s increased. Across six of the seven species, population size declined over the course of this preliminary study; however, seasonal shifts were impossible to distinguish from potential anthropogenic effects of construction [Current Zoology 56 （1）： 118-133, 2010].

The emergence of antiretroviral resistance mutations represents a major threat to the achievement of national and global goals for the elimination of HIV-1 infection. The global strategy in 2019 in Cte d'Ivoire is a new national policy for the management of people living with HIV with the administration of dolutegravir (dTG)-based fixed-dose combination. The aim of our study was to evaluate HIV-1 resistance to antiretrovirals (ARVs) in infected adult subjects in Cte d’Ivoire in the context of a systematic switch to a dTG-based combination. Between February 2022 and October 2023, a cross-sectional survey with random sampling was conducted in 06 services caring for people living with HIV. A total of 139 participants were included in the study. Adults with a viral load ≥ 1000 copies/mL were tested for HIV-1 ARV resistance mutations. Molecular analyses were performed using protocol of ANRS-MIE (National Agency for Research on AIdS and emerging infectious diseases). The interpretation is performed by HIVGRAd (https://***/cms/grade/). The frequencies of HIV-1 resistance to non-nucleotide reverse transcriptase inhibitors (NNRTIs), nucleotide reverse transcriptase inhibitors (NRTIs), integrase inhibitors (IINTs) and protease inhibitors (PIs) were 82%, 73%, 19% and 11% respectively. The main mutations observed in the different classes were K103N (45%), M184V (64%), E157Q (19%) and L10V/M46I/A71V/I54V (6%) respectively. This study reveals the emergence of resistance to dTG-based fixed-dose combinations, favored by high rates of resistance to NRTIs and NNRTIs. This finding underlines the need for enhanced viral load monitoring and HIV-1 genotyping tests to guide the choice of NRTIs for combination therapy. In addition, monitoring for mutations to second-generation NRTIs is essential, given the scale-up of dTG-based regimens currently underway in Cte d’Ivoire.

Switchgrass as a bioenergy crop in the Loess Plateau, China：Potential lignocellulosic feedstock production and environmental conservation

A large portion of the Loess Plateau of China is characterized as “marginal” with serious land degradation and desertification problems. Consequently, two policies, Grain for Green and Western development Action were established by the Chinese government in response to the demand for ecological protection and economic development in the Loess Plateau. These policies are designed to increase forest cover, expand farmlands, and enhance soil and water conservation, while creating sustainable vegetation restoration. Perennial grasses have gained attention as bioenergy feedstocks due to their high biomass yields, low inputs, and greater ecosystem services compared to annual crops. Moreover, perennial grasses limit nutrient runoff and reduce greenhouse gas emissions and soil losses while sequestering carbon. Additionally, perennial grasses can generate economic returns for local farmers through producing bioenergy feedstock or forage on marginal lands. Here, we suggest a United States model energy crop, switchgrass（Panicum virgatum L.） as a model crop to minimize land degradation and desertification and to generate biomass for energy on the Loess Plateau.
