The transcription factor ZEB1 promotes chemoresistance in prostate cancer cell lines

One of the factors promoting tumoral progress is the abnormal activation of the epithelial-mesenchymal transition(EMT)program which has been associated with chemoresistance in tumoral cells.The transcription factor zinc finger E-box-binding homeobox 1(ZEB1),a key EMT activator,has recently been related to docetaxel resistance,the main chemotherapeutic used in advanced prostate cancer treatment.The mechanisms involved in this protective effect are still unclear.In a previous work,we demonstrated that ZEB1 expression induced an EMT-like phenotype in prostate cancer cell lines.In this work,we used prostate cancer cell lines 22Rvl and DU145 to study the effect of ZEB1 modulation on docetaxel resistance and its possible mechanisms.The results showed that ZEB1 overexpression conferred to 22Rvl cell resistance to docetaxel while its silencing made DU145 cells more sensitive to it.Analysis of resistance markers showed no presenee of ATP-binding cassette subfamily B member 1(MDR1)and no changes in breast cancer resistance protein(BCRP)or ATP-binding cassette subfamily C member 10(MRP7).However,a correlation between ZEB1,multidrug resistance-associated protein 1(MRP1),and ATP-binding cassette subfamily C member 4(MRP4)expression was observed.MRP4 inhibition,using MK571,resensitized cells with ZEB1 overexpression to docetaxel treatment.In addition,modulation of ZEB1 and subsequent change in MRP4 expression correlated with a lower apoptotic response to docetaxel,characterized by lower B-cell lymphoma 2(Bcl2),high BCL2-associated X protein(Bax),and high active caspase 3 expression.The response to docetaxel in our model seems to be mediated mainly by activation of the apoptotic death program.Our results showed that modulation of MRP4 could be a mediator of ZEBl-related resistance to docetaxel in prostate cancer,making it a possible marker for chemotherapy response in patients who do not express MDR1.

It has been reported that one of the factors that promotes tumoral progression is the abnormal activation of the epithelial-mesenchymal transition program. This process is associated with tumoral cells acquiring invasive and malignant properties and has the transcription factor zinc finger E-box-binding homeobox 1 （ZEB1） as one of its main activators. However, the role of ZEB1 in promoting malignancy in prostate cancer （PC, a） is still unclear. Here, we report that ZEB1 expression correlates with Gleason score in PCa samples and that expression of ZEB1 regulates epithelial-mesenchymal transition and malignant characteristics in PCa cell lines. The results showed that ZEB1 expression is higher in samples of higher malignancy and that overexpression of ZEB1 was able to induce epithelial-mesenchymal transition by upregulating the mesenchymal marker Vimentin and downregulating the epithelial marker E-Cadherin. On the contrary, ZEB 1 silencing repressed Vimentin expression and upregulated E-Cadherin. ZEB1 expression conferred enhanced motility and invasiveness and a higher colony formation capacity to 22Rvl cells whereas DU145 cells with ZEB1 silencing showed a decrease in those same properties. Blueberries are rich in phenolic compounds including anthocyanins which are closely related to biological health functions.The purpose of this study was to investigate the antioxidant activity of blueberry anthocyanins extracted from'Brightwell'rabbiteye blueberries in mice.After one week of adaptation,C57BL/6J healthy male mice were divided into different groups that were administered with 100,400,or 800 mg/kg blueberry anthocyanin extract(BAE),and sacrificed at different time points(0.1,0.5,1,2,4,8,or 12 h).The plasma,eyeball,intestine,liver,and adipose tissues were collected to compare their antioxidant activity,including total antioxidant capacity(T-AOC),superoxide dismutase(SOD)activity and glutathione-peroxidase(GSH-PX/GPX)content,and the oxidative stress marker malondialdehyde(MDA)level.The results showed that blueberry anthocyanins had positive concentration-dependent antioxidant activity in vivo.The greater the concentration of BAE,the higher the T-AOC value,but the lower the MDA level.The enzyme activity of SOD,the content of GSH-PX,and messenger RNA(mRNA)levels of Cu,Zn-SOD,Mn-SOD,and GPX all confirmed that BAE played an antioxidant role after digestion in mice by improving their antioxidant defense.The in vivo antioxidant activity of BAE indicated that blueberry anthocyanins could be developed into functional foods or nutraceuticals with the aim of preventing or treating oxidative stress-related diseases.

Ectophylla alba is a tent-making bat that roosts in mixed-sex clusters comprising adults and offspring.Our goal was to determine the genetic identity of individuals belonging to different roosting groups.We tested the hypothesis of kin selection as a major force structuring group composition.We used 9 microsatellites designed for E.alba to determine the genetic identity and probability of parentage of individuals.We analyzed parentage and kinship using the software ML-Relate,GenAIEx,and Cervus.The obtained relationship probabilities(0.5)revealed a clear maternal relationship between female adults and offspring with allele compatibility,and at least 5 relationships between male adults and pups.We found a low degree of relatedness within roosting groups.Between roosting groups at different sites,the mean probability of a half-sibling relationship ranged from 0.214 to 0.244 and,for full-sibling relationship,from 0.383 to 0.553.Genetically,adult individuals were poorly related within clusters,and kinship as an evolutionary force could not explain group membership.

In the last decades several epidemic emergencies have been affecting the world,influencing the social relationships,the economics and the habits.In particular,starting in the early 080,the Acquired Immunodeficiency Syndrome,AIDS,is representing one of the most worrying sanitary emergency,that has caused up to now more than 25 million of dead patients.The infection is caused by the Human Immunodeficiency Virus,HIV,that may be transmitted by body fluids;therefore with wise behaviours the epidemic spread could rapidly be contained.This sanitary emergency is peculiar for the long incubation time:it can reach even 10 years,a long period in which the individual can unconsciously infect other subjects.The identification of the number of infected unaware people,mandatory to define suitable containment measures,is here obtained by using the extended Kalman filter applied to a noisy model in which,reasonably,only the number of infected diagnosed patients is available.Numerical simulations and real data analysis support the effectiveness of the approach.

Disaster Risk Governance as Assemblage: The Chilean Framework of the 1985 San Antonio Earthquake

The purpose of this article is to analyze disaster risk governance through assemblage theory, identifying how-during the altered political context of a military regime with a centralized disaster risk management as in the case of Chile in 1985-new actors emerge during the disaster response phase as a de/reterritorialization effect that is influenced by their agencies and relationships, disfiguring the edges of the assemblage. Based on this conceptualization, it is possible to investigate the interactions between the different actors, their power relations, and their reconfigurations in the governance exercise. For this purpose, we reviewed the response phase of the 1985 San Antonio earthquake that affected the central zone of Chile, where strategic functions, institutions, and forms of power are concentrated.To describe and visualize the actors during the response phase in the disaster risk governance framework, a map of actors was developed that identifies the existing relationships and their different weights. The central scale proved to be dominant and occupied a political space that was transfigured by its overrepresentation-enforced by allies such as the banking system and business associations-enhancing a neoliberal agenda. The leaps in scale from the central scale to the local scale cancel agency of the last, destabilizing its capacity to deal with the effects of the earthquake and isolating it from the decision-making processes. Consequently,delays in providing aid demonstrate that authoritarian governments do not provide better management in the disaster response phase.

Cancers remain among the most devastating diseases in the human population in spite of considerable advances in limiting their impact on lifespan and healthspan.The multifactorial nature of cancers,as well as the number of tissues and organs that are affected,have exposed a considerable diversity in mechanistic features that are reflected in the wide array of therapeutic strategies that have been adopted.Cachexia is manifested in a number of diseases ranging from cancers to diabetes and ageing.In the context of cancers,a majority of patients experience cachexia and succumb to death due to the indirect effects of tumorigenesis that drain the energy reserves of different organs.Considerable information is available on the pathophysiological features of cancer cachexia,however limited knowledge has been acquired on the resident stem cell populations,and their function in the context of these diseases.Here we review current knowledge on cancer cachexia and focus on how tissues and their resident stem and progenitor cell populations are individually affected.

Treat-to-target in Crohn's disease:Will transmural healing become a therapeutic endpoint?

Crohn's disease(CD) represents a chronic transmural inflammatory condition of the gastrointestinal tract, which usually leads to structural damage and signific-ant disability. Deep remission--defined by both clinical and endoscopic remission, signifying mucosal healing-represents the current endpoint in the treat--to--target strategy, significantly improving patients' long--term outcomes. Transmural healing(TH) could be a more effe-ctive target, but this possibility remains unclear. This narrative review aims to critically review and summarize the available literature relating TH to long--term outcomes, being the first of its kind and to the best of the author's knowledge. A systematic literature search(from incep-tion to March 31 2018) was performed, using multiple databases, and identifying seven full--text manuscripts. In those studies, long--term favorable outcomes(≥ 52 wk) included sustained clinical remission, as well as fewer therapeutic changes, CD--related hospitalizations, and surgeries. Despite heterogeneous design and me-thodological limitations, six of the studies demonstrated that TH or intestinal healing(TH plus mucosal healing) were predictive for the aforementioned favorable out-comes. Therefore, TH may become a reasonable the-rapeutic target and be included in the concept of deep remission. Further prospective, well-designed, multicenter trials aiming to better define the role of TH in personalized therapy for CD and to determine the long-term influence of TH on bowel damage and disability are warranted.
