Color-coded perfluorocarbon nanodroplets for multiplexed ultrasound and photoacoustic imaging

Laser-activated perfluorocarb on n anodroplets are an emerging class of phase-cha nge, dual-c ontrast age nts that can be utilized in ultraso und and photoacoustic imaging. Through the ability to differe ntiate subpopulations of nano droplets via laser activatio n at differe nt wavelengths of n ear-infrared light, optically-triggered color-coded perfluorocarb on nano droplets prese nt themselves as an attractive tool for multiplexed ultrasound and photoacoustic imaging. In particular, laser-activated droplets can be used to provide quantitative spatiotemporal information regarding distinct biological targets, allowing for their potential use in a wide range of diagnostic and therapeutic applications. In the work prese nted, laser-activated color-coded perfluorocarb on nan odroplets are syn thesized to selectively resp ond to laser irradiati on at corresp on ding wavele ngths. The dyn amic ultraso und and photoacoustic signals produced by laser-activated perfluorocarbon nano droplets are evaluated in situ prior to implementation in a murine model. In vivo, these particles are used to distinguish unique particle trafficking mechanisms and are show n to provide ultraso und and photoacoustic contrast for up to 72 hours within lymphatics. Overall, the con ducted studies show that laser-activated color-coded perfluorocarbo n nano droplets are a promising agent for multiplexed ultraso und and photoacoustic imaging.

Predation risk affects egg mass but not egg steroid hormone concentrations in yellow-legged gulls

Predators have both direct, consumptive effects on their prey and non-lethal effects on physiology and behavior, including reproductive decisions, with cascading effects on prey ecology and evol tion. Here, we experime ntally tested such non-lethal effects of exposure to in creased predati on risk on clutch size, egg mass, and the concentration of yolk steroid hormones in the yellow-legged gull Larus michahellis. We simulated increased predation risk by displaying stuffed predators (adult fox Vulpes vulpes, and adult buzzard Buteo buteo) to breeding adults before egg laying. The concentration of corticosterone, which has bee n show n to in crease un der exposure to mater nal predati on risk in other species, and of testosterone did not differ between eggs from mothers exposed to the predators and eggs from control mothers (i.e., eggs exposed to a novel object of similar size and position to the stuffed predators). The concentration of the two hormones negatively covaried. Clutch size did not vary according to experimental treatment, whereas egg mass was markedly larger in clutches from n ests exposed to predators than in clutches from control n ests. By in creasing egg mass, mothers may reduce the risk of cooling of the eggs when incubation is impeded by predators, boost energy reserves, reduce post-natal detectability caused by food solicitation, and/ or enha nee developme nt at hatchi ng, thus increasing the cha nces of offspri ng survival. In general, our results are inconsistent with most of the few previous studies on similar non-lethal predator effects and suggest that such effects may vary among species according to ecological conditions, social behavior, and developmental mode.

Lack of association between UGT1A7,UGT1A9,ARP,SPINK1 and CFTR gene polymorphisms and pancreatic cancer in Italian patients

AIM： To investigate simultaneously UGT1A7, UGT1A9, ARP, SPINK and CFTR genes to verify whether genetic polymorphisms predispose to the development of pancreatic cancer （PC）.METHODS： Genomic DNA of 61 pancreatic cancer patients and 105 healthy controls （HC） were analyzed. UGT1A7 genotyping was determined by PCR-RFLP analysis. Specific PCR and sequencing were used to analyze genetic variants of UGT1A9, ARP, SPINK1 and CFTR genes.RESULTS： Four different alleles （^＊1：WT; ＂2： N129K and R131K; ＂3： N129K, R131K, and W208R; and ＂4： W208R） in UGT1A7 and three different alleles （^＊1：WT;^＊4： Y242X; and ^＊5： D256N） in UGT1A9 were detected. All UGT1A polymorphisms were observed at similar frequency in PC patients and HC. Seven different alleles in ARP were found in PC patients and HC at similar frequency. The SPINK1 mutations N34S and P55S occurred in five PC patients with a prevalence （4.1%） not significantly different from that observed （2.0%） in HC. The only CFTR AFS08 mutation was recognized in three PC patients with a prevalence （4.9%） similar to HC.CONCLUSION： UGT1A7, UGT1A9, ARP, SPINK1 and CFTR gene polymorphisms are not associated with PC in Italian patients.

Performance of interspecific grapevine varieties in north-east Italy

A renewed interest in interspecific varieties has recently emerged, due mainly to producers and consumers more aware of organic farming and impact of phytochemicals in the environment. The assessment of 19 European Vitis hybrids was investigated in an area mostly dedicated to viticulture, the North-EasternItaly. Major agronomic traits, yield, quality characteristics and disease resistance were evaluated during a three-year period (2004 to 2006). Wine sensory analyses were performed and compared with international Vitis vinifera varieties. Even though no genotypes resulted adequate for market release, the results obtained confirm the potential importance of hybrids in an “eco-friendly” viticulture and identify the genotypes interesting for further investigation and breeding: GF 138-3 and ga 48-12 showed good agronomic performance, resistance to more grape diseases and high quality wine.

Performance of Cosmetic Ingredients Evaluated by Their Membrane Protection Efficiency

Plant extracts (PEs) are frequently used as ingredients in skin and hair care products to improve protection against the excess of sun exposure. PEs are usually selected based on their antioxidant capacity for reducing the damage in lipids and proteins, which are key molecules in the maintenance of skin homeostasis and hair integrity. Interestingly, biomolecules that do not have any antioxidant capacity can have a protective effect by physically adsorbing and inhibiting leakage of membranes. Our aims are: 1) to develop an experimental protocol that quantifies the level of membrane damage/protection;2) to recommend an approach to compare the membrane protection efficiency of different PEs;3) to briefly describe the mechanisms of membrane protection. Evaluation of membrane damage/protection is based on a phospholipid liposome system that signals the membrane rupture caused by photo-induced lipid oxidation. Carboxyfluorescein (CF) emission is monitored as a function of time (120 min) in a controlled experimental setup, in which a certain level of membrane damage is induced by Dimethyl-Methylene Blue (DMMB) photosensitized oxidation during red-light irradiation. Percentage levels of membrane damage/protection are calculated by comparing the CF fluorescence level of DMMB-liposomes in the presence of varying concentrations of membrane-protective ingredients with that of surfactant-dissolved DMMB-liposomes. Membrane protectants reduce the level of CF fluorescence during irradiation of DMMB-liposomes. gallic acid (ga) and trehalose (Tre) were used as standard compounds to compute the levels of protection by the antioxidant and physical adsorption mechanisms, respectively. The percentage of membrane protection was shown to be linearly proportional to the concentration of the standard protectants. Age and petrogenesis of the newly discovered Early Permian granite in the Kumtor gold field， Kyrgyz Tien-Shan

Permian intrusions are widespread in the Middle and Southern Tien Shan， with fewer occurrences in the Northern Tien-Shan. Notably， many of these intrusions are spatially associated with a variety of ore deposits， indicating a significant link between magmatic activity and mineralization processes in these areas. We studied granite samples recently recovered from drilling in the Kumtor gold field to evaluate their potential relationships with gold mineralization. We report major and trace element geochemistry， zircon U-Pb age and Hf isotope data for this so-called Kumtor granite. The Kumtor granite is metaluminous to peraluminous and belongs to the high-K and calc-alkaline series with I-type geochemical characteristics. The relatively high K2O and Na2O concentrations and low high field strength elements （HFSE） and heavy rare earth elements （HREE）， the presence of biotite within these I-type granites， together with their low zircon saturation temperatures （731–779 °C）， suggest that they were likely derived from a hydrous source formed by dehydration melting of mica-bearing， medium- to high-K metabasaltic rocks. Our zircon U-Pb dating results indicate that the Kumtor granite intruded at 293 ± 1.7 Ma， which is consistent with the age range of other Middle Tien-Shan granitoids. The zircon Hf isotopic composition is εHf（t）= –7.56 to –5.05， indicating an ancient （1.39 to 1.52 ga） crustal origin. Petrographical， geochemical and geochronological data indicate that the Kumtor granite is similar to leuco-granites of the Terekty complex. Our results indicate that the Kumtor granite was emplaced in the early Permian in a post-collision setting and may have temporal and genetic relationships with gold mineralization.

MASSIVE （mapping seismic vulnerability and risk of cities） is a GIS-based earthquake preparedness system that was developed under the European Union Civil Protection Mechanism project （ga No. 070401/2009/540429/SUB/A4）, in order to provide civil protection authorities with accurate, and easily transferable tools for generating up-to-date maps of seismic hazard, seismic vulnerability and seismic risk of buildings, at the scale of the single building block. In addition, MASSIVE developed and ran state-of-the-art models to assess the risk for population evacuation in dense urban agglomerations given an earthquake event. The MASSIVE methodology was designed, implemented and validated considering two European pilot sites, heavily struck by recent earthquakes, which are the western part of the Larger Metropolitan Area of Athens （GR）, and the city of L＇ Aquila in the Abruzzo Region （IT）. The validation of the results using past earthquake records shows that the performance of MASSIVE is prosperous, achieving a correlation between the modeled and the on-site measured Pgas （peak ground accelerations） higher than 0.75, while the correlation between the on-site reported building damages and the ones predicted by the MASSIVE system has been of the order of 0.80.
