Role of Salmonella enterica exposure in Chilean Crohn's disease patients

AIM:To study the association between exposure toSalmonella enterica(SE)and Crohn’s disease(CD)and its clinical implications in Chilean ***:Ninety-four unrelated Chilean CD patients from CAREI(Active Cohort Registry of Inflammatory Bowel Disease)presenting to a single inflammatory bowel disease(IBD)unit of a University Hospital were prospectively included in this study.A complete clinical evaluation,including smoking history,was performed at the initial visit,and all the important data of clinical evolution of CD were *** samples from these CD patients and 88 healthy sex-and agematched control subjects were analyzed for exposure to SE and for their NOD2/CARD15 gene status using the presence of anti-Salmonella lipopolysaccharide antibodies[immunoglobulin-G type(IgG)]and polymerase chain reaction(PCR),*** also evaluated exposure to SE in 90 sex-and age-matched patients without CD,but with known smoking status(30 smokers,30 non-smokers,and 30 former smokers).RESULTS:CD patients comprised 54 females and 40males,aged 35.5±15.2 years at diagnosis with a mean follow-up of 9.0±6.8 *** was inflammatory in 59 patients(62.7%),stricturing in 24(25.5%)and penetrating in 15(15.5%).Thirty cases(31.9%)had lesions in the ileum,29(30.8%)had ileocolonic lesions,32(34.0%)had colonic lesions and 23(24.4%)had perianal *** CD patients(17%)were exposed to SE compared to 15(17%)of 88 healthy control subjects(P=0.8).Thirty-one CD patients(32.9%)were smokers,and 7(7.4%)were former smokers at *** the group exposed to SE,10 of 16 patients(62.5%)were active smokers compared to 21 of 78 patients(26.9%)in the unexposed group(P=0.01).On the other hand,10 of 31 smoking patients(32%)were exposed to SE compared to 5 of 56 nonsmoking patients(9%),and one of the seven former smokers(14%)(P=0.01).In the group of 90 patients without CD,but whose smoking status was known,there was no differ-ence in exposure to SE[3 of 30 smokers(10%),5 of30 non-s

Data Analysis of Multiplex Sequencing at SOLiD Platform:A Probabilistic Approach to Characterization and Reliability Increase

New sequencing technologies such as Illumina/Solexa, SOLiD/ABI, and 454/Roche, revolutionized the biological researches. In this context, the SOLiD platform has a particular sequencing type, known as multiplex run, which enables the sequencing of several samples in a single run. It implies in cost reduction and simplifies the analysis of related samples. Meanwhile, this sequencing type requires an additional filtering step to ensure the reliability of the results. Thus, we propose in this paper a probabilistic model which considers the intrinsic characteristics of each sequencing to characterize multiplex runs and filter low-quality data, increasing the data analysis reliability of multiplex sequencing performed on SOLiD. The results show that the proposed model proves to be satisfactory due to: 1) identification of faults in the sequencing process;2) adaptation and development of new protocols for sample preparation;3) the assignment of a degree of confidence to the data generated;and 4) guiding a filtering process, without discarding useful sequences in an arbitrary manner.

Lack of association between UGT1A7,UGT1A9,ARP,SPINK1 and CfTR gene polymorphisms and pancreatic cancer in Italian patients

AIM： To investigate simultaneously UGT1A7, UGT1A9, ARP, SPINK and CfTR genes to verify whether genetic polymorphisms predispose to the development of pancreatic cancer （PC）.METHODS： Genomic DNA of 61 pancreatic cancer patients and 105 healthy controls （HC） were analyzed. UGT1A7 genotyping was determined by PCR-RfLP analysis. Specific PCR and sequencing were used to analyze genetic variants of UGT1A9, ARP, SPINK1 and CfTR genes.RESULTS： four different alleles （^＊1：WT; ＂2： N129K and R131K; ＂3： N129K, R131K, and W208R; and ＂4： W208R） in UGT1A7 and three different alleles （^＊1：WT;^＊4： Y242X; and ^＊5： D256N） in UGT1A9 were detected. All UGT1A polymorphisms were observed at similar frequency in PC patients and HC. Seven different alleles in ARP were found in PC patients and HC at similar frequency. The SPINK1 mutations N34S and P55S occurred in five PC patients with a prevalence （4.1%） not significantly different from that observed （2.0%） in HC. The only CfTR AfS08 mutation was recognized in three PC patients with a prevalence （4.9%） similar to HC.CONCLUSION： UGT1A7, UGT1A9, ARP, SPINK1 and CfTR gene polymorphisms are not associated with PC in Italian patients.

fair Plan 4: Safeguarding the Climate of “This Island Earth”

Earth is the only habitable planet in the solar system and beyond in interstellar space for a distance that would take us at least 80,000 years to traverse at the speed of Voyager 1. Thus our home planet is “This Island Earth”. Here we use our Simple (engineering-type) Climate Model to calculate the change in global-mean near-surface air temperature from 1765 through the third millennium for historical emissions and two scenarios of future emissions of greenhouse gases: (1) a Reference scenario of unabated emissions, and (2) our fair Plan scenario wherein emissions are phased out to zero from 2020 to 2100. The temperature change for the Reference cases increases to 5.2°C (9.4°f) in 2225 and remains there for at least 40 human generations. By design, the temperature change for the fair Plan increases only to 2°C (3.6°f)—the limit adopted by the UN framework Convention on Climate Change “to prevent dangerous anthropogenic interference with the climate system”—in 2082 and thereafter decreases through the remainder of the millennium. Accordingly, we need to adopt the fair Plan to safeguard the climate of “This Island Earth”.

Magnetic resonance imaging:A new tool for diagnosis of acute ischemic colitis?

AIM： To define the evolution of ischemic lesions with 7T magnetic resonance imaging （7T-MRI） in an animal model of acute colonic ischemia. METHODS： Adult Sprague-Dawley rats were divided into two groups. Group I underwent inferior mesenteric artery （IMA） ligation followed by macroscopic observa- tions and histological analysis. In group H, 7T-MRI was performed before and after IMA ligation and followed by histological analysis. RESULTS： Morphological alterations started to develop 1 h after IMA ligation, when pale areas became evident in the splenic flexure mesentery and progressively wors-ened up to 8 h thereafter, when the mesentery was less pale, and the splenic flexure loop appeared very dark. The 7T-MRI results reflected these alterations, showing a hyperintense signal in both the intraperitoneal space and the colonic loop wall 1 h after IMA ligation; the lat- ter progressively increased to demonstrate a reduction in the colonic loop lumen at 6 h. Eight hours after IMA ligation, MRI showed a persistent colonic mural hyper- intensity associated with a reduction in peritoneal free fluid. The 7T-MRI findings were correlated with histolog- ical alterations, varying from an attenuated epithelium with glandular apex lesions at 1 h to coagulative necro- sis and loss of the surface epithelium detected 8 h after IMA ligation. CONCLUSION： MRI may be used as a substitute for invasive procedures in diagnosing and grading acute ischemic colitis, allowing for the early identification of pathological findings.

Surgical Management of Two Simultaneously Occurring Epiphrenic Esophageal Diverticula

Among all diverticula of the esophagus, epiphrenic diverticula occur less frequently than those in the pharyngo-esophageal segment. Two simultaneously occurring diverticula in the epiphrenic esophagus are very rarely reported in the English literature. A 52-year-old woman had a 3-year history of troublesome dysphagia, heartburn, chest pain and cough. Initial investigation included a physical examination and a barium swallow, which showed the presence of two diverticula in the epiphrenic esophagus. Esophagoscopy confirmed the presence only of the lower diverticulum. The patient underwent a left thoracotomy, the esophagus was mobilised from the hiatus to the aortic arch. The necks of the diverticula were localised in the posterior wall and between them there was a bridge of circular muscular fibres, which was divided. Diverticulectomy over an intra-esophageal 54 f Maloney dilator was performed. The upper diverticulum was left intact because it disappeared after the bridge of esophageal muscles has been myotomized. A long myotomy was carried out, and a modified Belsey fundoplication was added. The postoperative course was uneventful, and the patient remains well at 10 years with no recurrence of symptoms.

Brief report: Plasma oxytocin is lower in children with Asperger syndrome and associated with autistic trait attention to detail

Aim: Autism spectrum disorders (ASD) are described as a continuum of severity gradient of autistic symptoms diffusing through particular ASD diagnoses, however the biological correlates among individuals with the different ASD diagnoses slightly or considerably differ. Oxytocin (OT) has been implicated to play an important role in autism etiology. Lower OT levels have been previously found in children with infantile autism, however in a group of high-functioning autistic subjects, no differences have been shown compared to controls. Moreover, whereas the opposite patterns of OT associations with social measures have been found in children with infantile autism compared to healthy children, no associations have been found in individuals with high-functioning autism. We aimed to find out the plasma OT differences between separate group of children with Asperger syndrome (AS) and healthy children and the associations of OT with particular autistic traits in a group of children with AS. Methods: We included 9 children (m = 6, f = 3) with AS at the age 9 to 12 years and 9 age- and gender-matched controls. OT levels were analyzed by ELISA method. Autistic traits in children with AS were evaluated by Autism Spectrum Quotient (AQ), child and adolescent versions. Results: Children with AS had significantly lower plasma OT levels compared to healthy children. We found the significant negative correlation of OT level and AQ Attention to detail area score. Conclusion: In spite of the lower OT level in children with AS, which is also previously found in children with infantile autism, the pattern of OT associations with autistic traits more resembles the pattern in non-autistic population. Our preliminary results support the hypothesis of continuum within the ASD particular diagnoses in the terms of biological correlates.

Viral hepatitis represents a major public health hazard and is associated withsignificant global mortality. Over the last decade, there have been significantdevelopments in the prevention and treatment of viral hepatitis. These changeshave led to a situation whereby global elimination has become a realistic goal,fully endorsed by the World Health Organization (WHO). By 2030, the WHOaims to reduce viral hepatitis mortality by 65% and reduce new infections by 90%by 2030. These are ambitious targets and will only be met through a sustainedprogramme. This will require expertise from hepatologists and virologists and thefields of public health and primary care. In this article, we review the causes ofviral hepatitis, its management through prevention and treatments, and the mostpressing challenges and recent advances.

In this paper we present the science potential of the enhanced X-ray Timing and Polarimetry(eXTP) mission for studies of strongly magnetized objects. We will focus on the physics and astrophysics of strongly magnetized objects, namely magnetars, accreting X-ray pulsars, and rotation powered pulsars. We also discuss the science potential of eXTP for QED studies. Developed by an international Consortium led by the Institute of High Energy Physics of the Chinese Academy of Sciences, the eXTP mission is expected to be launched in the mid 2020s.
