Background:The cellular tumor protein p53(TP53)is a tumor suppressor gene that is frequently mutated in human *** various cancer types,the very aggressive high-grade serous ovarian carcinoma(HGSOC)exhibits the high-est prevalence of TP53 mutations,present in>96%of *** intensive efforts to reactivate p53,no clinical drug has been approved to rescue p53 *** this study,our primary objective was to administer in vitro-transcribed(IVT)wild-type(WT)p53-mRNA to HGSOC cell lines,primary cells,and ortho-topic mouse models,with the aim of exploring its impact on inhibiting tumor growth and dissemination,both in vitro and in ***:To restore the activity of p53,WT p53 was exogenously expressed in HGSOC cell lines using a mammalian vector ***,IVT WT p53 mRNA was delivered into different HGSOC model systems(primary cells and patient-derived organoids)using liposomes and studied for proliferation,cell cycle progression,apoptosis,colony formation,and chromosomal *** alterations induced by p53 mRNA were analyzed using RNA sequencing in OVCAR-8 and primary HGSOC cells,followed by ingenuity path-way *** vivo effects on tumor growth and metastasis were studied using orthotopic xenografts and metastatic intraperitoneal mouse ***:Reactivation of the TP53 tumor suppressor gene was explored in differ-ent HGSOC model systems using newly designed IVT mRNA-based *** introduction of WT p53 mRNA triggered dose-dependent apoptosis,cell cycle arrest,and potent long-lasting inhibition of HGSOC cell *** analysis of OVCAR-8 cells upon mRNA-based p53 reactivation revealed significant alterations in gene expression related to p53 signaling,such as apoptosis,cell cycle regulation,and DNA *** p53 function concurrently reduces chromosomal instability within the HGSOC cells,under-scoring its crucial contribution in safeguarding genomic integrity by moderating the baselin

目的旨在翻译Chase护理管理者胜任力测评工具(the Chase Nurse Manager Competency Instrument,CNMCI)为意大利语,并对其进行信度和效度检验。方法对英文版CNMCI进行翻译,并通过横断面研究验证其信效度。采用预翻译(建立等效)、初始翻译、预测试、审查和管理5个步骤的问卷翻译方法,将英文版工具翻译为意大利语版。于2022年8月到2023年6月,选取意大利3个地区31所医疗机构349名护理管理者进行调查。采用内容效度、结构效度评估工具的效度;采用Cronbach'sα系数、重测信度评估工具的信度。结果意大利语版CNMCI,包括“知识理解”和“实施和/或使用能力”两个分量表,每个分量表包括53个条目。工具的条目内容效度指数(I-CVI)为0.83~1.00,两个分量表的内容效度指数(S-CVI)均为0.97。经验证性因子分析显示该工具各项拟合指数表现良好。工具中“知识理解”分量表的Cronbach'sα系数为0.978,“实施和/或使用能力”分量表的Cronbach'sα系数为0.976。各个维度之间的相关系数为0.494~0.908重测信度为0.820,表明工具的一致性较好。结论意大利语版CNMCI在测量护理管理者胜任力方面显示出良好的信度和效度,并支持该工具的多维度结构。此外,研究结果支持了这些因素的层次性质,进一步支持了该工具的有效性。

The electrochemical corrosion mechanisms of Mg alloys were extensively studied in previous investigations of different chemical com-positions,modified surface states and various electrolyte ***,recent research focused on the active state of Mg dissolution,leading to unresolved effects of secondary phases adjacent to a stableα-solid solution passive *** present study investigates the fundamental electrochemical corrosion mechanisms of three different Laves phases with varying phase morphologies and phase fractions in the passive state of Mg-Al-Ca *** microstructure was characterized by(transmission-)electron microscopy and synchrotron-based transmission X-ray *** electrochemical corrosion resistance was determined with a standard three-electrode setup and advanced in-situ flow cell measurements.A new electrochemical activity sequence(C15>C36>α-Mg>C14)was obtained,as a result of a stable passive layer formation on theα-solid ***,nm-scale Mg-rich precipitates were identified within the Laves phases,which tend to inhibit the corrosion kinetics.

We studied the effect of titanium dioxide(TiO_(2))nanoparticles(NPs)on dielectric behavior of Naþion-conducting salt-complexed polymer nanocomposite system formed from a binary polymer blend of poly(ethylene oxide)(PEO)and polyvinyl pyrrolidone(PVP),with the addition of both sodium metaperiodate(NaIO_(4))at concentration 10 wt.%and TiO_(2) NPs of size10 nm,at concentrations 1,2,3,4 and 5 wt.%.Free standing nanocomposite PEO/PVP/NaIO_(4)/TiO_(2) films(150m)were characterized at room-temperature by analyzing their complex electrical impedance and dielectric spectra in the range 1 Hz–1 *** the concentration of 3 wt.%of TiO_(2) NPs,both ion conductivity and dielectric permittivity of the PEO/PVP/NaIO_(4)/TiO_(2) ion-conducting dielectrics reach an enhancement by more than one order of magnitude as compared to nanoadditive-free case.

目的基于《国际功能、残疾和健康分类》(ICF)框架,系统评价运动疗法对慢性非特异性颈痛(CNSNP)患者身体结构、功能、活动和参与等方面的干预效果。方法构建PICO架构,系统检索CBM、万方、维普、中国知网、Cochrane Library、Embase、PubMed和Web of Science等中、英文数据库,纳入有关运动疗法干预CNSNP患者的随机对照试验,时间范围为建库至2024年3月。采用Cochrane偏倚风险评估工具和物理治疗证据数据库(PEDro)量表对纳入文献进行质量评价,采用GRADE对结局指标进行证据质量评价,采用RevMan 5.3进行数据合并和分析,采用Stata 18.0进行偏倚风险评估。结果最终纳入11篇文献,共668例患者。PEDro量表评分5~8分。运动疗法类型包括肌力训练、稳定性训练、本体感觉训练、瑜伽和普拉提等;对照组包括空白对照、物理因子治疗和健康教育。运动疗法可显著增加患者颅椎角(SMD=0.84, 95%CI 0.42~1.26, P <0.001),降低疼痛视觉模拟量表评分(SMD=-2.05,95%CI-2.58~-1.52, P <0.001),提高压力疼痛阈值(MD=112.27, 95%CI 75.03~149.50, P <0.001),提高颈椎前屈(SMD=1.24, 95%CI 0.34~2.15, P=0.007)、侧屈(SMD=1.52, 95%CI 0.40~2.65, P=0.008)关节活动度,提高颈深屈肌耐力(SMD=1.02, 95%CI 0.10~1.94, P=0.03),改善颈椎关节位置觉(SMD=-1.00,95%CI-1.47~-0.53, P <0.001),但对于改善后屈(SMD=0.85, 95%CI-1.04~2.75, P=0.38)、旋转(SMD=1.65, 95%CI-0.35~3.65, P=0.11)关节活动度的效果不显著;运动疗法还能降低患者颈椎功能障碍指数评分(MD=-11.88, 95%CI-16.09~-7.68, P <0.001),对健康调查简表评分效果不显著(MD=19.04, 95%CI-3.00~41.08, P=0.09)。结论运动疗法可以改善CNSNP患者的异常姿势、疼痛、前屈和侧屈运动、颈屈肌耐力和关节位置觉,改善功能障碍,但对提高颈部后屈和旋转运动,改善生活质量方面仍需进一步的研究。

Use of gold nanoparticles(GNPs)in medicine is an emerging field of translational research with vast clinical implications and exciting therapeutic ***,the safety of using GNPs in human subjects is an important question that remains *** study reviews over 20 clinical trials focused on GNP safety and aims to summarize all the clinical studies,completed and ongoing,to identify whether GNPs are safe to use in humans as a therapeutic *** these studies,GNPs were implemented as drug delivery devices,for photothermal therapy,and utilized for their intrinsic therapeutic effects by various routes of *** studies revealed no major safety concerns with the use of GNPs;however,the number of trials and total patient number remains ***-dose,multicenter blinded trials are required to deepen our understanding of the use of GNPs in clinical settings to facilitate translation of this novel,multifaceted therapeutic *** clinical trials will require collaboration between clinicians,scientists,and biotechnology companies.

Analysis of Changes of Extreme Temperature during June to August Season over Tanzania

Natural and human systems are exposed and vulnerable to climate extremes, which contributes to the repercussions of climate variability and the probability of disasters. The impacts of both natural and human-caused climate variability are reflected in the reported changes in climate extremes. Particularly at the local community levels in the majority of the regions, there is currently a dearth of information regarding the distribution, dynamics, and trends of excessive temperatures among the majority of Tanzanians. Over the years 1982-2022, this study examined trends in Tanzania’s extreme temperature over the June to August season. Based on the distinction between absolute and percentile extreme temperatures, a total of eight ETCCDI climate indices were chosen. Mann-Kendall test was used to assess the presence of trends in extreme climatic indices and the Sen’s Slope was applied to compute the extent of the trends in temperature extremes. The study showed that in most regions, there is significant increase of warm days and nights while the significant decrease of cold days and nights was evident to most areas. Moreover, nighttime warming surpasses daytime warming in the study area. The study suggests that anthropogenic influences may contribute to the warming trend observed in extreme daily minimum and maximum temperatures globally, with Tanzania potentially affected, as indicated in the current research. The overall results of this study reflect patterns observed in various regions worldwide, where warm days and nights are on the rise while cold days and nights are diminishing.
