Design and Simulation of a Novel Pneumotronic System Aimed to the Investigation of Vascular Phenomena Induced by Limb Compression

Intermittent Pneumatic Compression (IPC) devices can be used to analyze the mechanisms underlying several vascular phenomena, such as hyperaemia. Commercial devices have limited dynamics and do not allow the delivery of customizable compressive pressure patterns, making the analysis of such phenomena difficult, which may require the application of long stimulations with low amplitude as well as fast compressions with higher pressure level. To overcome these issues, a novel pneumotronic device aimed to the investigation of the physiological effects induced by limb compressions is conceived and presented in this work. The design requirements of the system, capable of delivering customizable compressive patterns in the range 0 mmHg-200 mmHg, are outlined. The final prototype architecture is described, and a mathematical model of the entire system, also including the interaction between the device and the limb tissues, is proposed. The performance of the device was evaluated in several conditions by means of simulations, whose results were compared to the data collected from experimental trials in order to validate the model. The outcomes of both experimentation and simulation trials proved the effectiveness of the solution proposed. A possible employment of this device for the investigation of the rapid compression-induced hyperaemia is presented. Other potential applications concern the wide range of intermittent-pneumatic compression treatments.

Color-coded perfluorocarbon nanodroplets for multiplexed ultrasound and photoacoustic imaging

Laser-activated perfluorocarb on n anodroplets are an emerging class of phase-cha nge, dual-c ontrast age nts that can be utilized in ultraso und and photoacoustic imaging. Through the ability to differe ntiate subpopulations of nano droplets via laser activatio n at differe nt wavelengths of n ear-infrared light, optically-triggered color-coded perfluorocarb on nano droplets prese nt themselves as an attractive tool for multiplexed ultrasound and photoacoustic imaging. In particular, laser-activated droplets can be used to provide quantitative spatiotemporal information regarding distinct biological targets, allowing for their potential use in a wide range of diagnostic and therapeutic applications. In the work prese nted, laser-activated color-coded perfluorocarb on nan odroplets are syn thesized to selectively resp ond to laser irradiati on at corresp on ding wavele ngths. The dyn amic ultraso und and photoacoustic signals produced by laser-activated perfluorocarbon nano droplets are evaluated in situ prior to implementation in a murine model. In vivo, these particles are used to distinguish unique particle trafficking mechanisms and are show n to provide ultraso und and photoacoustic contrast for up to 72 hours within lymphatics. Overall, the con ducted studies show that laser-activated color-coded perfluorocarbo n nano droplets are a promising agent for multiplexed ultraso und and photoacoustic imaging.

Perinatal variation and covariation of oxidative status and telomere length in yellow-legged gull chicks

The perinatal period is critical to survival and performance of many *** birds,rapid postnatal growth and sudden exposure to aerial oxygen around hatching markedly affect the chick redox status,with potentially negative consequences on physiology mediated by oxidative *** addition,telomere length(TL)undergoes reduction during birds’early life,partly depending on oxidative ***,relatively few studies have focused specifically on the changes in oxidative status and TL that occur immediately after *** this study of the yellow-legged gull Larus michahellis,we found that chicks undergo a marked increase in plasma total antioxidant capacity and a marked decrease in the concentration of pro-oxidant molecules during the first days after *** addition,TL in erythrocytes decreased by 1 standard deviation over the 4days *** mass and tarsus length covaried with total antioxidant capacity and concentration of pro-oxidants in a complex way,that partly depended on sex and laying order,suggesting that oxidative status can affect ***,TL positively covaried with the concentration of pro-oxidant molecules,possibly because retention of high concentrations of pro-oxidant molecules results from mechanisms of prevention of their negative effects,including reduction in ***,this study shows that chicks undergo marked variation in oxidative status,which predicts growth and subsequent TL,prompting for more studies of the perinatal changes in the critical posthatching stages.

Safer intestinal invagination for a solid pancre-atico-jejunal anastomosis in presence of a soft texture pancreatic remnant and non-dilated duct

Pancreatico-jejunal anastomosis after pancreato- duodenectomy still represents the Achilles＇ heel of the proce- dure： the failure of this anastomosis is relatively common and it is the main cause of post-operative morbidity and mortality. Studies have described different reconstruction strategies for the control of the development of post-operative pancreatic fistula, but the strategy to obtain a safer pancreatico-jejunal anastomosis is still far from satisfaction. We report a novel variation of the invagination technique based on preliminary clinical experience in 8 patients who underwent pancreatico- jejunal anastomosis after pancreatoduodenectomy in our hepatobiliopancreatic center from 2008 to 2014. The varia- tion could obtain a safer intestinal invagination for a solid pancreatico-jejunal anastomosis even in the presence of soft pancreatic remnant.

Effects of submergence in water on seed germination and vigor of the Copaifera lucens(Fabaceae) seedlings

We analyzed the tolerance of Copaifera lucens seeds to sub- mersion in water to assess the use of this species for direct seeding in riparian forest restoration programs. Seeds were submerged in water for 2, 4, 8, 16 and 32 days or not submerged （control = 0 days of submer- gence）. For the control and at the end of each period of submersion, germination and seedling vigor tests were carried out. For germination tests, seeds were sown in plastic pots containing sand and kept in labora- tory conditions. The percentage of seed germination, the germination rate and the average germination time were analyzed. For seedlings, total biomass, leaf area, leaf mass per area and leaf area ratio were analyzed. Submersion time drastically affected the dissolved oxygen content and seed germination. Between 4 and 8 days of submersion there was a de- crease from 83.8% to 15.6% in the germination percentage. No seed germination occurred after 16 days of submersion. Although there was a significant decrease in the percentage of seed germination between 4 and 8 days of submersion, seedling vigor was not affected. Seeds of this species were partially tolerant to submersion in water, suggesting that C. lucens is a promising species for direct seeding in riparian forest restora- tion projects.

Despite a decrease in incidence over past decades,gastric cancer remains a major global health problem. In the more recent period, survival has shown only minor improvement, despite significant advances in diagnostic techniques, surgical and chemotherapeutic approaches, the development of novel therapeutic agents and treatment by multidisciplinary teams. Because multiple genetic mutations, epigenetic alterations, and aberrant molecular signalling pathways are involved in the development of gastric cancers, recent research has attempted to determine the molecular heterogeneity responsible for the processes of carcinogenesis, spread and metastasis. Currently, some novel agents targeting a part of these dysfunctional molecular signalling pathways have already been integrated into the standard treatment of gastric cancer, whereas others remain in phases of investigation within clinical trials. It is essential to identify the unique molecular patterns of tumours and specific biomarkers to develop treatments targeted to the individual tumour behaviour. This review analyses the global impact of gastric cancer, as well as the role of Helicobacter pylori infection and the efficacy of bacterial eradication in preventing gastric cancer development. Furthermore, the paper discusses the currently available targeted treatments and future directions of research using promising novel classes of molecular agents for advanced tumours.

Chemokine and chemotactic signals in dendritic cell migration

Dendritic cells (DCs) are professional antigen-presenting cells responsible for the activation of specific T-cell responses and for thedevelopment of immune tolerance. Immature DCs reside in peripheral tissues and specialize in antigen capture, whereas matureDCs reside mostly in the secondary lymphoid organs where they act as antigen-presenting cells. The correct localization of DCs isstrictly regulated by a large variety of chemotactic and nonchemotactic signals that include bacterial products, DAMPs (dangerassociated molecular patterns), complement proteins, lipids, and chemokines. These signals function both individually and inconcert, generating a complex regulatory network. This network is regulated at multiple levels through different strategies, such assynergistic interactions, proteolytic processing, and the actions of atypical chemokine receptors. Understanding this complexscenario will help to clarify the role of DCs in different pathological conditions, such as autoimmune diseases and cancers and willuncover new molecular targets for therapeutic interventions.

Epilepsy in Prader-Willi syndrome:clinical,diagnostic and treatment aspects

Background:Epilepsy associated with Prader-Willi syndrome(PWS)represents an early and important complication,often not clearly reported and described in the ***,there are controversial data about the clinical characteristics of epilepsy and electroencephalographic(EEG)abnormalities found in these *** sources:Based on recent original publications,we have reviewed the different types of seizures and EEG findings in PWS patients,the response to antiepileptic treatment,and the prognosis of ***:The frequency of epilepsy in PWS patients ranges from 4%to 26%.The types of seizure include generalized tonic-clonic seizures,complex partial seizures,atypical absence,staring spells,and myoclonic,tonic and hemiclonic seizures,but the most frequent type is focal *** epilepticus has never been *** abnormalities are not typical but variable in different ***,generalized and focal discharges are the most frequently reported *** is no evidence of relationship between the course of epilepsy and frequency,morphology and spread of EEG ***,epilepsy in PWS patients is usually responsive to antiepileptic monotherapy with rapid seizure control and a good ***:The frequency of epilepsy is higher in PWS patients than in general populations and this complication can be a challenge for the clinicians of these *** studies are needed to confirm the good long-term prognosis.
