A 12-week feeding trial with juvenile red drum(Sciaenops ocellatus) fed high-soybean meal(SBM) diets was conducted to investigate a putative biomarker of nutritional imbalance, N-formimino-L-glutamate(FIgLU). three fishmeal-free, 60% SBM pelleted diets(named B12, Fol, and Met, respectively) were tested to evaluate the effects on growth performance and tissue metabolite profiles of supplementation of vitamin B12(0.012 mg/kg), folate(10 mg/kg), methionine(1 g/kg) respectively, above basal supplementation levels. A fourth SBM-based diet(named B12/Fol/Met) was formulated with a combination of B12, folate, and methionine to attain the above-mentioned target concentrations. A fifth 60% SBM diet(named FWS) with methionine supplementation(1 g/kg above basal supplementation levels), enriched with taurine, lysine and threonine as well as minerals, was also tested. this diet contained formulation targets and additives which have allowed for replacing fishmeal with plant proteins in rainbow trout feeds. Control diets included a fishmeal-based diet(named FM), an unsupplemented basal 60% SBM diet(named SBM60), and a “natural” diet(named N) made up of equal parts of fish(cigar minnows), squid and shrimp as a positive reference for growth performance. Formulated feeds contained approximately 37% total crude protein, approximately 14% total crude lipid and were energetically balanced. Standard growth performance metrics were measured, and tissues(liver, muscle) were collected at week 12 to evaluate diet-induced metabolic changes using nuclear magnetic resonance(NMR)-based *** results show that the FWS diet outperformed all other SBM diets and the FM diet under all performance metrics(PgLU was not detected in fish fed the N diet but was detected in those fed the SBM diets and the FM diet. Fibrous dysplasia(FD)is a rare,disabling skeletal disease for which there are no established *** evidence supports inhibiting the osteoclastogenic factor receptor activator of nuclear kappa-B ligand(RANKL)as a potential treatment *** this study,we investigated the mechanisms underlying RANKL inhibition in FD tissue and its likely indirect effects on osteoprogenitors by evaluating human FD tissue pre-and post-treatment in a phase 2 clinical trial of denosumab(NCt03571191)and in murine in vivo and ex vivo preclinical *** analysis of human and mouse tissue demonstrated increased osteogenic maturation,reduced cellularity,and reduced expression of the pathogenic gαs variant in FD lesions after RANKL *** sequencing of human and mouse tissue supported these *** interaction between osteoclasts and mutant osteoprogenitors was further assessed in an ex vivo lesion model,which indicated that the proliferation of abnormal FD osteoprogenitors was dependent on *** results from this study demonstrated that,in addition to its expected antiosteoclastic effect,denosumab reduces FD lesion activity by decreasing FD cell proliferation and increasing osteogenic maturation,leading to increased bone formation within *** findings highlight the unappreciated role of cellular crosstalk between osteoclasts and preosteoblasts/osteoblasts as a driver of FD pathology and demonstrate a novel mechanism of action of denosumab in the treatment of bone disease.

After spinal cord injury,there is an extensive infiltration of immune cells,which exacerbates the injury and leads to further neural ***,a major aim of current research involves targeting the immune response as a treatment for spinal cord *** much research has been performed analyzing the complex inflammatory process following spinal cord injury,there remain major discrepancies within previous literature regarding the timeline of local cytokine *** objectives of this study were to establish an overview of the timeline of cytokine regulation for 2 weeks after spinal cord injury,identify sexual dimorphisms in terms of cytokine levels,and determine local cytokines that significantly change based on the severity of spinal cord *** were inflicted with either a mild contusion,moderate contusion,severe contusion,or complete transection,7 mm of spinal cord centered on the injury was harvested at varying times post-injury,and tissue homogenates were analyzed with a Cytokine/Chemokine 27-Plex *** demonstrated pro-inflammatory cytokines including tumor necrosis factorα,interleukin-1β,and interleukin-6 were all upregulated after spinal cord injury,but returned to uninjured levels within approximately 24 hours post-injury,while chemokines including monocyte chemoattractant protein-1 remained upregulated for days *** contrast,several anti-inflammatory cytokines and growth factors including interleukin-10 and vascular endothelial growth factor were downregulated by 7 days *** spinal cord injury,tissue inhibitor of metalloproteinase-1,which specifically affects astrocytes involved in glial scar development,increased more than all other cytokines tested,reaching 26.9-fold higher than uninjured *** a mild injury,11 cytokines demonstrated sexual dimorphisms;however,after a severe contusion only leptin levels were different between female and male *** conclusion,pro-inflammatory cytokines in

the ability to hit a target with precision and from a great distance has been reserved for the world’s superpowers. However, this resource is increasingly being threatened as drones with this long-range and precision capability are becoming more accessible to those who don’t have this strategic ability. this article starts with an analysis of the Iranian HESA Shahed 136 drone to discuss the latest innovations in low-cost long-range precision weapons, specifically the use of kamikaze drones and loitering munitions. this is an exploratory study that starts by discussing the notion of a kamikaze drone and then analyses the design options for the Shahed 136, to reflect on the future of this new type of weapon and its implications for the economic and political relationship between weapon and cost. the conclusion is that the HESA Shahed 136 revolutionizes the concept of precise long-range strikes, a function that until now was reserved for expensive and technologically demanding tactical missiles and aircraft, and which can now be carried out with cheap drones. this creates an arms race not only in producing the most technological and precise weaponry but also the least expensive.

BACKgROUND total hip arthroplasty(tHA)is a common procedure for end stage *** learning curve for tHA is complex and *** of the most difficult skills to master is acetabular *** wish to identify if experience in arthroplasty leads to preservation of more bone *** to investigate if increasing surgeon experience will predict an ever decreasing acetabular cup *** A retrospective case series of four attending orthopaedic surgeons was *** uncemented elective total hip arthroplasties since appointment were selected for *** size of acetabular cup used was noted and logistic regression was used to identify if a trend to smaller cups *** A total of 1614 subjects were included with a mean age of 64 *** cups were on average 0.18mm smaller per year(95%confidence interval-0.25 to-0.11,Pgeon trends showed cup sizes to decrease 0.27 mm/year for surgeon A,0.02 mm/year for surgeon B,0.15 mm/year for surgeon C and 0.29 mm/year for surgeon *** of the four surgeons had a more pronounced trend to smaller cups for male subjects than their female *** We found increasing surgeon experience to be associated with an ever-decreasing acetabular cup *** acetabular cup size may act as a surrogate marker of surgical proficiency by virtue of decreased acetabular reaming.
