目的基于血液筛查核酸检测反应性献血者的HbV感染的确认,探讨核酸检测反应性献血者的归队策略。方法联合应用自建的高灵敏度核酸检测体系、血液核酸筛查等多种核酸检测(NAT)方法,并结合血清学检测、献血者随访,对核酸检测反应性(NAT-yield)献血者中的HbV感染进行确认和感染状态识别。依据确认的HbV感染血浆样本,比较不同确认方法、确认指标或指标组合对HbV感染确认的效果。结果2010年11月—2021年2月,在血液筛查检出的876位NAT-yield献血者中共确认HbV感染者511人(ObI 451人,急性早期HbV感染者27人,不能确认感染者33人,无感染者30人,不能确认HbV感染者335人)。采用单检系统对混检系统检出的HbV感染血浆进行复测的检出率为96.6%,明显高于混检系统对单检系统检出的HbV DNA反应性(HbV DNA R)组和鉴别试验无反应性(NDR)组的复测检出率(76.4%和55.7%)(PbV DNA R样本复测检出率无明显差异(P>0.05),但均明显高于NDR样本(PbI献血者既往NAT数据,有46%经历多次NAT检测而未能检出。有59.1%ObI献血者随访检不出HbV DNA。ObI献血者中抗-Hbc+占比为90.2%,单独抗-Hbc+为49.2%,远高于不能确认感染组(PbeAg、抗-Hbe和抗-Hbc IgM在ObI和不能确认感染组中的比例极低且无差异(P>0.05)。结论近60%的NAT-yield献血者可以确认HbV感染。为保证献血者归队的安全性,需要更高灵敏度的HbV DNA确证技术提高HbV感染的确认率。抗-Hbc是NAT-yield献血者ObI风险排查和归队评估最重要的血清学指标。

A study on the effects of Navier slip, in conjunction with other flow parameters, on unsteady flow of reactive variable viscosity third-grade fluid through a porous saturated medium with asymmetric convective boundary conditions is presented. The channel walls are assumed to be subjected to asymmetric convective heat exchange with the ambient, and exothermic chemical reactions take place within the flow system. The heat exchange with the ambient obeys Newton＇s law of cooling. The coupled equations, arising from the law of conservation of momentum and the first law of thermodynamics, then the derived system are non- dimensionalised and solved using a semi-implicit finite difference scheme. The lower wall slip parameter is observed to increase the fluid velocity profiles, whereas the upper wall slip parameter retards them because of backflow at the upper channel wall. Heat pro- duction in the fluid is seen to increase with the slip parameters. The wall shear stress increases with the slip parameters while the wall heat transfer rate is largely unaltered by the lower wall slip parameter but marginally increased by the upper wall slip parameter.

The objective of this work was to evaluate different superficial treatments of sisal fibres employing lignin,and their use as a reinforcement agent in cementitious *** treatments consisted of superficially impregnating sisal fibres(S)with organosolv lignin(LO),organosolv lignin and glutaraldehyde(LOG),Kraft lignin(LK)and Kraft lignin and glutaraldehyde(LKG).The fibre modifications were verified by FTIR-ATR and SEM analyzes,and the presence of lignin on the surface of the fibres was evidenced,confirming the effectiveness of the *** mechanical,thermal(by TGA)and water absorption properties of the fibres before and after the modifications were also *** treatment,the modified fibres presented an expressive reduction of the water absorption and did not show significant changes in the mechanical properties when compared with the natural unmodified sisal fibre(SNAT).It was verified an increase in the thermal stability of the treated fibres which can be attributed to the insertion of lignin on the *** evaluate the performance of the fibres in the cementitious composites,cement plates(CP)were produced with different treated fibres(CP-SLOG,CP-SLO,CP-SLKG,CP-SLK)and fibres without treatment(CP-SNAT).The composites were evaluated concerning to the water absorption,porosity and mechanical *** fractured regions were also investigated by *** composites prepared showed sim.lar values of absorption and porosity *** the mechanical properties,the composites prepared with modified fibres showed a significant increase in the modulus of rupture and modulus of elasticity compared with CP-SNAT,while toughness was sim.lar to all *** the SEM images,it was observed that the modified fibres immersed in the cementitious plates showed no degradation,indicating that the impregnation of lignin acted as a protective agent of the ***,the treatments of the fibres with lignin led to a significant improvement in th

Characterization of Leaf Transcriptome in banksia hookeriana

Abstract banksia is a significant element in vegetation of southwestern Australia, a biodiversity hotspot with global significance. In particular, banksia hookeriana represents a species with significant economic and ecological importance in the region. For better conservation and management, we reported an overview of transcriptome of b. hookeriana using RNA-seq and de novo assembly. We have generated a total of 202.7 million reads （18.91 billion of nucleotides） from four leaf sam- ples in four plants of b. hookeriana, and assembled 59,063 unigenes （average size = 1098 bp） through de novo transcriptomc assembly. Among them, 39,686 unigenes were annotated against the Swiss-Prot, Clusters of Orthologous Groups （COG）, and NCbI non-redundant （NR） protein databases. We showed that there was approximately one single nucleotide polymorphism （SNP） per 5.6-7.1 kb in the transcriptome, and the ratio of transitional to transversional polymorphisms was approximately 1.82. We compared unigenes of b. hookeriana to those of Arabidopsis thaliana and Nelumbo nucifera through sequence homology, Gene Ontology （GO） annotation, and KEGG pathway analyses. The comparative analysis revealed that unigenes of b. hookeriana were closely related to those of N. nucifera. b. hookeriana, N. nucifera, and A. thaliana shared sim.lar GO anno- tations but different distributions in KEGG pathways, indicating that b. hookeriana has adapted to dry-Mediterranean type shrublands via regulating expression of specific genes. In total 1927 potential sim.le sequence repeat （SSR） markers were discovered, which could be used in the genotype and genetic diversity studies of the banksia genus. Our results provide valuable sequence resource for further study in banksia.

Mesenchymal stem cell （MSC）-based treatments have shown promise for improving tendon healing and repair. MSCs have the potential to differentiate into multiple lineages in response to select chemical and physical stimuli, including into tenocytes. Cell elongation and cytoskeletal tension have been shown to be instrumental to the process of MSC differentiation. Previous studies have shown that inhibition of stress fiber formation leads MSCs to default toward an adipogenic lineage, which suggests that stress fibers are required for MSCs to sense the environmental factors that can induce differentiation into tenocytes. As the Rho/ROCK signal transduction pathway plays a critical role in both stress fiber formation and in cell sensation, we examined whether the activation of this pathway was required when inducing MSC tendon differentiation using rope-like silk scaffolds. To accomplish this, we employed a loss-of-function approach by knocking out ROCK, actin and myosin （two other components of the pathway） using the specific inhibitors Y-27632, Latrunculin A and blebbistatin, respectively. We demonstrated that independently disrupting the cytoskeleton and the Rho/ ROCK pathway abolished the expression of tendon differentiation markers and led to a loss of spindle morphology. Together, these studies suggest that the tension that is generated by MSC elongation is essential for MSC teno-differentiation and that the Rho/ROCK pathway is a critical mediator of tendon differentiation on rope-like silk scaffolds.

Skin is widely used as a drug delivery route due to its easy access and the possibility of using relatively painless methods for the administration of bioactive ***,the barrier properties of the skin,along with its multilayer structure,impose severe restrictions on drug transport and ***,bioengineered models aimed at emulating the skin have been developed not only for optimizing the transdermal transport of different drugs and testing the safety and toxicity of substances but also for understanding the biological processes behind skin *** though in vivo research is often preferred to study biological processes involving the skin,in vitro and ex vivo strategies have been gaining increasing relevance in recent ***,there is a noticeably increasing adoption of in vitro and ex vivo methods by internationally accepted ***,microfluidic organ-on-a-chip devices are nowadays emerging as valuable tools for functional and behavioural skin *** in miniaturization,automation and reliability still need to be addressed in order to create skin models that can predict skin behaviour in a robust,high-throughput manner,while being compliant with regulatory issues,standards and *** this review,skin models for transdermal transport,wound repair and cutaneous toxicity will be discussed with a focus on high-throughput *** microfluidic strategies driven by advancements in microfabrication technologies will also be revised as a way to improve the efficiency of existing models,both in terms of complexity and throughput.
