Midazolam in rabbits terminates dysrhythmias caused by intracerebroventricular ropivacaine

The current study was designed to investigate the mechanisms by which ropivacaine may act within the central nervous system （CNS） to produce cardiotoxicity. Eighty New Zealand rabbits were divided into four groups randomly. In Group 1,20 rabbits received intracerebroventricular （icv） saline, and then received icv ropivacaine 30 min later. In Group 2, 20 rabbits received icv ropivacaine. Whenever dysrhythmias continued for more than 5 min, 0.1 ml saline was administered into the left cerebral ventricle. Ten minutes later, 0.1 ml midazolam was given into the left lateral ventricle. In Group 3, 20 rabbits received icv ropivacaine, and once the dysrhythmias developed, the inspired isoflurane concentration was increased from 0.75% to 1.50%. In Group 4, 20 animals received an intravenous （iv） phenylephrine infusion until dysrhythmias occurred. In Group 1, the rabbits did not develop dysrhythmias in response to icv saline, whereas dysrhythmias did develop in these animals after icv ropivacaine. In Group 2, icv saline had no effect on the dysrhythmias; however, icv midazolam terminated cardiac dysrhythmias. In Group 3, an increase in the concentration of the inspired isoflurane had no effect on dysrhythmias. In Group 4, icv midazolam had no effect on dysrhythmias in response to iv phenylephdne. Ropivacaine administered directly into the CNS is capable of producing cardiac dysrhythmias; midazolam terminated dysrhythmias presumably by potentiation of y-aminobutyric acid （GABA） receptor activity. Our results suggest that ropivacaine produces some of its cardiotoxicity not only by the direct cardiotoxicity of the drug, but also by the CNS effects of ropivacaine.

Investigation of nursing students’ knowledge of and attitudes about problem-based learning

Purpose:To investigate nursing students’knowledge of and attitudes about problem-based learning(PBL).Methods:A total of 1200 students were surveyed at eight nursing colleges in Hunan ***:In all,1037 valid questionnaires were returned,for an effective return rate of 86.4%.Some 54.4%of the students learned that PBL was a pedagogical method from teachers,and 27.8%of the students had participated in PBL *** all of students(97.6%)were interested in PBL,and 66.7%of survey participants believed that students who were not good at solving problems would have difficulty in PBL ***:Nursing educators should guide students to adapt to new learning approaches,and encourage students to participate in the teaching reform to promote students’autonomous learning ability,innovation ability,and comprehensive ability.

Insights into the pigment and non-pigment phenolic profile of polyphenol extracts of jujube peel and their antioxidant and lipid-lowering activities

This study aimed to provide a comprehensive overview of the pigment and non-pigment phenolic profile of polyphenol extracts of jujube peel and their ***,the pigment and non-pigment phenolic composition as well as antioxidant activities of free and bound polyphenol extracts in jujube peel were compared among twelve varieties *** the antioxidant and lipid-lowering activities of free and bound polyphenol extracts of‘Junzao’jujube peel were investigated ***,23 phenolic compounds were identified,in which flavonols were the main pigments and all varieties showed similar qualitative patterns but different quantitative *** antioxidant activities significantly correlated with the content of total phenolics and total ***,both free and bound polyphenol extracts of‘Junzao’jujube peel exhibited DPPH,ABTS free radical scavenging capacity,and oxygen radical absorbance capacity in a dose-dependent ***,both of them improved the resistance of Caenorhabditis elegans under H_(2)O_(2) oxidative stress and heat stress at 37℃(20-100μg/mL).What’s more,compared to free polyphenol extract,bound polyphenol extract decreased the level of body lipid accumulation and triglyceride more *** results suggest that jujube peel is an exploitable source of natural flavonoid pigments and phenolic ingredients with strong antioxidant and lipid-lowering properties.

Toward long-life Zn anode using highly polar electrolyte additives

Unstable Zn interface caused by rampant dendrite growth and parasitic side reactions always hinders the practical application of aqueous zinc metal batteries（AZMBs）,Herein,tyrosine（Tyr） with high molecular polarity was introduced into aqueous electrolyte to modulate the interfacial electrochemistry of Zn *** AZMBs,the positively charged side of Tyr can be well adsorbed on the surface of Zn anode to form a water-poor layer,and the exposed carboxylate side can be easily coordinated with Zn2+,favoring inducing uniform plating of Zn2+and inhibiting the occurrence of water-induced side *** in turn enable the achievement of highly stable Zn ***,the Zn anodes achieve outstanding cyclic stability(3000 h at 2 mA cm-2,2 mA h cm-2and 1300 h at 5 mA cm-2,5 mA h cm-2),high average Coulombic efficiency（99.4% over 3200 cycles）,and high depth of discharge（80% for 500 h）.Besides,the assembled Zn‖NaV3O8·1.5H2O full cells deliver remarkable capacity retention and ultra-long lifetime(61.8% over 6650 cycles at 5 A g-1) and enhanced rate capability(169 mA h g-1at 5 A g-1).The work may promote the design and deep understanding of electrolyte additives with high molecular polarity for high-performance AZMBs.

Exchange bias(EB)is a fundamental phenomenon in widespread information ***,a comprehensive understanding of its microscopic origin remains a great *** key issue in the debate is the role of frustration and disorder in the EB mechanism,which motivates the exploration of the EB effect in spin glass(SG)***,in the SG state of Crdoped Hund's metal CsFe_(2)As_(2),we discover a giant EB effect with a maximum bias field of -2 T,which is almost two orders of magnitude larger than that of traditional alloy *** results indicate that the giant EB effect should originate from the exchange interactions at the natural boundaries between the tunable ferromagnetic-like(FM)regions around Cr dopants and the SG matrix,via which the FM spins are strongly pinned by the frozen spins in the SG *** addition,the temperaturedependent and cooling-field-dependent EB behaviors could be interpreted well by the SG model with frustrated FM/SG boundaries,which provides an intuitive and explicit understanding of the impact of glassy parameters on the EB *** these results suggest that the correlated metals are promising directions for exploring the EB effect in the SG state.

肝纤维化被认为是各种慢性肝病导致的肝细胞弥漫性损伤的一种病理过程,其特征是细胞外基质(extracellular matrix,ECM)的积累。肝星状细胞(HSC)的活化是被认为是肝纤维化的重要一步,并导致肝纤维化获得多种功能,包括收缩,趋化性,和矩阵重塑。HSC活化是肝纤维化发生发展进程中最重要的环节。通常,抑制细胞增殖,诱导细胞凋亡和衰老是阻断HSC活化的潜在治疗方法。研究发现microRNA(miRNA)在多种纤维化疾病过程中的具有调控作用。MiRNA能调控多种肝纤维化的相关因子的表达,反过来,肝纤维化又能导致多种miRNA表达发生改变。之前,本课题组已证实miR-145与肝纤维化发生与发展密切相关,但是miR-145在肝星状细胞衰老过程中的作用鲜有报道,这需要进一步的研究。本实验探讨了miR-145在肝纤维化发生过程中表达情况,并进一步研究miR-145对活化HSC衰老影响以及其中的分子机制。研究内容如下:(1)miR-145,ZEB2和p53在肝纤维化过程中的表达检测在体内,采用腹腔注射四氯化碳(carbon tetrachloride,CCl)6周建立C57小鼠肝纤维化模型,取肝纤维化组织进行HE,Masson和天狼星红染色,同时检测α-平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)和I型胶原(Collagen type1,Col.I)的蛋白表达对纤维化模型进行鉴定。此外,采用qPCR和Western blot技术检测肝纤维化组织和原代HSCs中miR-145,ZEB2和p53 mRNA和蛋白表达,采用免疫荧光双染技术观察ZEB2和p53是否表达于活化HSCs中。体外我们使用TGF-β1(10ng/ml)浓度对大鼠肝星状细胞HSC-T6和人肝星状细胞LX-2刺激24h,然后检测ZEB2,miR-145和p53的表达变化。结果发现在CCl诱导的肝纤维化小鼠的肝组织和原代HSC中以及体外TGF-β1处理的HSC中,miR-145和p53的表达均是降低的,而ZEB2表达水平升高。(2)miR-145对TGF-β1诱导的活化LX-2细胞衰老的影响在LX-2中,采用miR-145 mimics和inhibitor建立过表达和沉默miR-145细胞模型,同时采用TGF-β1(10ng/ml,24h)进行刺激,紧接着采用流式细胞术,β-半乳糖苷酶染色检测细胞衰老情况以及Western blot检测相关衰老蛋白和Col.I和α-SMA的蛋白表达情况。结果提示:过表达miR-145能促进活化LX-2的衰老,同时抑制Col.I和α-SMA的蛋白表达;沉默miR-145,使TGF-β1诱导的LX-2衰老减少,同时Col.I和α-SMA的蛋白表达水平进一步升高。(3)ZEB2对TGF-β1诱导的活化LX-2细胞衰老的影响在LX-2细胞中,采用ZEB2小干扰RNA(ZEB2 siRNA)和ZEB2质粒(ZEB2plasmid)建立沉默ZEB2和过表达ZEB2细胞模型,同时采用TGF-β1(10ng/ml,24h)进行刺激,采用流式细胞术,β-半乳糖苷酶染色检测细胞衰老情况以及Western blot检测衰老相关蛋白和Col.I和α-SMA的蛋白表达情况。结果提示:沉默ZEB2能促进活化LX-2的衰老,同时抑制Col.I和α-SMA的蛋白表达;过表达ZEB2,使TGF-β1诱导的LX-2衰老减少,同时Col.I和α-SMA的蛋白表达水平进一步升高。(4)p53对TGF-β1诱导的活化LX-2细胞衰老的影响在LX-2细胞中,采用p53小干扰RNA(p53 siRNA)和p53质粒(p53 plasmid)建立沉默p53和过表达p53细胞模型,同时采用TGF-β1(10ng/ml,24h)进行刺激,采用流式细胞术,β-半乳糖苷酶染色检测细胞衰老情况以及Western blot检测衰老相关蛋白和Col.I和α-SMA的蛋白表达情况。结果提示:过表达p53能促进活化LX-2的衰老,同时抑制Col.I和α-SMA的蛋白表达;沉默p53,使TGF-β1诱导的LX-2衰老减少,同时Col.I和α-SMA的蛋白表达水平进一步升高。(5)miR-145与ZEB2的靶向性研究生物信息学软件分析结果显示,miR-145同ZEB2之间可能具有一定的靶向关系。在LX-2细胞中,我们分别采用miR-145 inhibitor和mimics建立沉默与过表达的细胞模型。紧接着采用qPCR与Western blot检测ZEB2 mRNA与蛋白的表达变化。结果提示:过表达的miR-145可以使ZEB2 mRNA与蛋白水平表达降低,相反的,沉默miR-145可以使ZEB2 mRNA和蛋白水平升高。此外,进一步,本课题组采用双荧光素酶报告基因实验确认两者的
