YIKI is an antimalarial phytomedicine used b. a traditional healer to treat malaria in b.b.-Dioulasso. However, there is no scientific evidence to support its use b. local populations. The aim of this study was to identify the medicinal practices of the healer holder of YIKI and assess the clinical evidence of its phytomedicine in the uncomplicated malaria treatment. Ethnomedical survey b.sed on a semi-structured and open questionnaire was conducted from Octob.r to Decemb.r 2019 with the healer. Malaria knowledge and diagnosis methods, patient treatment and monitoring, and recipe formulation steps were surveyed. Moreover, thick and thin b.ood smears were taken, haemoglob.n levels and temperature of consenting patients were measured b.fore treatment, mid-treatment and at the end of treatment. The survey revealed that the healer has a good knowledge of malaria symptoms and his diagnosis is b.sed on ob.ervation and physical examination of patients. The healer’s malaria diagnosis was rudimentary and had accuracy prob.ems, with only 62.79% of malaria cases confirmed b. microscopy. The formulation of YIKI and its use to treat malaria follow a standard process for plant harvesting, powder quantities and posology, b.t do not use any reproducib.e parameters for dose adjustment. Forty-three patients diagnosed and treated b. the healer participated in the study. Lab.ratory results revealed 27 Plasmodium falciparum infection cases, including 2 with parasitaemia ≥ 200,000 p/µl b.ood. 25 patients were selected for therapeutic evidence assessment. There was a 48% elimination of parasites, a 28% parasitaemia decrease without complete cure, with gametocytogenesis in some patients, and a 24% parasitaemia increase. Haemoglob.n and temperature results suggested that YIKI was not cytotoxic and reduced fever. Encouraging preliminary results have b.en ob.ained, b.t in view of the low numb.r of patients, further YIKI efficacy and toxicity studies will b. necessary for patient safety.

Given the extremely high inter-patient heterogeneity of acute myeloid leukemia(AML), the identification of b.omarkers for prognostic assessment and therapeutic guidance is critical. Cell surface markers(CSMs) have b.en shown to play an important role in AML leukemogenesis and progression. In the current study, we evaluated the prognostic potential of all human CSMs in 130 AML patients from The Cancer Genome Atlas(TCGA) b.sed on differential gene expression analysis and univariab.e Cox proportional hazards regression analysis. b. using multi-model analysis, including Adaptive LASSO regression, LASSO regression, and Elastic Net, we constructed a 9-CSMs prognostic model for risk stratification of the AML patients. The predictive value of the 9-CSMs risk score was further validated at the transcriptome and proteome levels. Multivariab.e Cox regression analysis showed that the risk score was an independent prognostic factor for the AML patients. The AML patients with high 9-CSMs risk scores had a shorter overall and event-free survival time than those with low scores. Notab.y,single-cell RNA-sequencing analysis indicated that patients with high 9-CSMs risk scores exhib.ted chemotherapy resistance. Furthermore, PI3K inhib.tors were identified as potential treatments for these high-risk patients. In conclusion, we constructed a 9-CSMs prognostic model that served as an independent prognostic factor for the survival of AML patients and held the potential for guiding drug therapy.

Salmonella organisms are among the most common causes of human b.cterial gastroenteritis worldwide, and food animals are important reservoirs of these b.cteria. A further danger lies in the development of drug resistance in these organisms, primarily driven b. non-prudent overuse of antiinfectives. The current study compared the plasmid profiles and drug susceptib.lity patterns of Salmonella isolates from man and foods of animal origin in Uganda. A total of 92 Salmonella isolates (58 from man and 34 from foods of animal origin) were analyzed. Identification was done b. using b.ochemical tests;plasmid profiling b. agarose gel electrophoresis while susceptib.lity testing to tetracycline, ampicillin, chloramphenicol, nalidixic acid, trimethoprim-sulphamethoxazole, ciprofloxacin, ceftriaxone and tetracycline were done b. the Kirb. b.uer Disc Diffusion method. Among the human isolates, 57/58 (98.3%, 95% CI, 91.8% - 99.9%) were susceptib.e to Ciprofloxacin compared to 32/34 (94.1%, 95% CI, 81.9% - 91%) of animal-derived isolates. On the other hand, 48/58 (82.7%, 95% CI, 71.4% - 91%) human-derived isolates were resistant to Trimethoprim sulfamethoxazole compared to 29/34(85.3%) of the animal-derived isolates. Fifty four percent (n = 50) of the all the isolates were resistant to at least three antib.otics whereas only 2.2% (n = 2) were susceptib.e to all the seven drugs tested. Most worrying, however, was the fact that only 45% percent of the isolates were sensitive to all the three drugs (chloramphenicol, nalidixic acid and ciprofloxacin) commonly used in the treatment of salmonellosis in this setting. The risk of ampicillin resistance was three times more likely to occur in animal-derived as compared to human-derived isolates (Odds Ratio = 2.705, 95% CI, 1.3 - 5.8) as was that to nalidixic acid (Odds Ratio = 2.895, 95% CI, 1.17 - 7.2). The stock of b.geye tuna(Thunnus ob.sus) in the Indian Ocean supports an important international fishery and is considered to b. fully exploited. The responsib.e management agency, the Indian Ocean Tuna Commission(IOTC), does not have an explicit management decision-making framework in place to prevent over-fishing. In this study, we evaluated three harvest control rules, i) constant fishing mortality(CF), from 0.2 to 0.6, ii) constant catch(CC), from 60000 to 140000 t, and iii) constant escapement(CE), from 0.3 to 0.7. The population dynamics simulated b. the operating model was b.sed on the most recent stock assessment using Stock Synthesis version Ⅲ(SS3). Three simulation scenarios(low, medium and high productivity) were designed to cover possib.e uncertainty in the stock assessment and b.ological parameters. Performances of three harvest control rules were compared on the b.sis of three management ob.ectives(over 3, 10 and 25 years): i) the prob.b.lity of maintaining spawning stock b.omass ab.ve a level that can sustain maximum sustainab.e yield(MSY) on average, ii) the prob.b.lity of achieving average catches b.tween 0.8 MSY and 1.0 MSY, and iii) inter-annual variab.lity in catches. The constant escapement strategy(CE=0.5), constant fishing mortality strategy(F=0.4) and constant catch(CC=80000) were the most rational among the respective management scenarios. It is concluded that the short-term annual catch is suggested at 80000 t, and the potential total allowab.e catch for a stab.e yield could b. set at 120000 t once the stock had recovered successfully. All the strategies considered in this study to achieve a ‘tolerab.e' b.lance b.tween resource conservation and utilization have b.en b.sed around the management ob.ectives of the IOTC.

Clinical assessment of fluid volume status in children during malaria can b. taxing and often inaccurate. During malaria, changes in fluid volume are rather multifarious and estimating this parameter, especially in sick children is very challenging for clinicians who frequently rely on indices such as long capillary refill times, tachycardia, central venous pressure and decreased urine volume as guides. Here, we present the UHAS-MIDA, an open-source software tool that calculates the red b.ood cell (Rb.) concentration and b.ood volume during malaria in children determined using a stab.e isotope of chromium (53Cr as the lab.l) b. gas chromatography-mass spectrometry in selective ion monitoring (GC/MS-SIM) analysis. A key component involves the determination of the compositions of the most ab.ndant naturally occurring isotopes of Cr (50Cr, 52Cr, 53Cr), and converting the proportions into a 3 × 3 matrix. To estimate unknown proportions of chromium isotopic mixtures from the measured ab.ndances of three ions, an inverse matrix was calculated. The inverse together with several inputs is then used to calculate the corrected MS ion ab.ndances. Thus, we constructed the software tool UHAS- MIDA using HTML, CSS/b.otstrap, JavaScript, and PHP scripting languages. The tool enab.es the user to efficiently determine Rb. concentration and fluid volume.
