Human-induced pluripotent stem cell-derived neural stem cell exosomes improve blood-brain barrier function after intracerebral hemorrhage by activating astrocytes via PI3K/AKT/MCP-1 axis

Cerebral edema caused by blood-brain barrier injury after intracerebral hemorrhage is an important factor leading to poor ***-induced pluripotent stem cell-derived neural stem cell exosomes(hiPSC-NSC-Exos)have shown potential for brain injury repair in central nervous system *** this study,we explored the impact of hiPSC-NSC-Exos on blood-brain barrier preservation and the underlying *** results indicated that intranasal delivery of hiPSC-NSC-Exos mitigated neurological deficits,enhanced blood-brain barrier integrity,and reduced leukocyte infiltration in a mouse model of intracerebral ***,hiPSC-NSC-Exos decreased immune cell infiltration,activated astrocytes,and decreased the secretion of inflammatory cytokines like monocyte chemoattractant protein-1,macrophage inflammatory protein-1α,and tumor necrosis factor-αpost-intracerebral hemorrhage,thereby improving the inflammatory *** sequencing indicated that hiPSC-NSC-Exo activated the PI3K/AKT signaling pathway in astrocytes and decreased monocyte chemoattractant protein-1 secretion,thereby improving blood-brain barrier *** with the PI3K/AKT inhibitor LY294002 or the monocyte chemoattractant protein-1 neutralizing agent C1142 abolished these *** summary,our findings suggest that hiPSC-NSC-Exos maintains blood-brain barrier integrity,in part by downregulating monocyte chemoattractant protein-1 secretion through activation of the PI3K/AKT signaling pathway in astrocytes.

Schwann cell transplantation is considered one of the most promising cell-based therapy to repair injured spinal cord due to its unique growth-promoting and myelin-forming properties.A the Food and Drug Administration-approved Phase I clinical trial has been conducted to evaluate the safety of transplanted human autologous Schwann cells to treat patients with spinal cord injury.A major challenge for Schwann cell transplantation is that grafted Schwann cells are confined within the lesion cavity,and they do not migrate into the host environment due to the inhibitory barrier formed by injury-induced glial scar,thus limiting axonal reentry into the host spinal *** we introduce a combinatorial strategy by suppressing the inhibitory extracellular environment with injection of lentivirus-mediated transfection of chondroitinase ABC gene at the rostral and caudal borders of the lesion site and simultaneously leveraging the repair capacity of transplanted Schwann cells in adult rats following a mid-thoracic contusive spinal cord *** report that when the glial scar was degraded by chondroitinase ABC at the rostral and caudal lesion borders,Schwann cells migrated for considerable distances in both rostral and caudal *** Schwann cell migration led to enhanced axonal regrowth,including the serotonergic and dopaminergic axons originating from supraspinal regions,and promoted recovery of locomotor and urinary bladder ***,the Schwann cell survival and axonal regrowth persisted up to 6 months after the injury,even when treatment was delayed for 3 months to mimic chronic spinal cord *** findings collectively show promising evidence for a combinatorial strategy with chondroitinase ABC and Schwann cells in promoting remodeling and recovery of function following spinal cord injury.

In this work exergetical performance analysis is carried out based on the second law of thermodynamics for organic flash cycle(OFC) using a two-phase expander instead of throttle expansion in order to recover efficiently finite thermal *** exergy destructions(anergies) at various components of the system are theoretically investigated as well as the exergy *** show that the anergy of heat exchanger or two-phase expander decreases while the anergy of throttle valve increases with increasing flash temperature,and the exergy efficiency has an optimum value with respect to the flash *** the optimal conditions with respect to the flash temperature,exergy efficiency increases with the heating temperature and the component having the largest exergy destruction varies with the flash temperature or heating temperature.

The concept of entransy has been newly proposed in terms of the analogy between heat and electrical conduction and could bc useful in analyzing and optimizing the heat-work conversion systems. This work presents comparative analyses of entransy and exergy for optimizations of heat-work conversion. The work production and heat transfer processes in Carnot cycle system are investigated with the formulations of exergy destruction, entransy loss, work entransy, entransy dissipation, and cfficiencics for both cases of dumping and non-dumping of used source fluid. The effects of source and condensation temperatures on the system performance arc systematically investigated for optimal condition of producing maximum work or work cntransy.

Many animals possess adhesive pads on their feet, which are able tO attach to various substrates while controlling adhesive forces during locomotion. This review article studies the morphology of adhesive devices in animals, and the physical mechanisms of wet adhesion and dry adhesion. The adhesive pads are either ＇smooth＇ or densely covered with special adhesive setae. Smooth pads adhere by wet adhesion, which is facilitated by fluid secreted from the pads, whereas hairy pads can adhere by dry adhesion or wet adhesion. Contact area, distance between pad and substrate, viscosity and surface tension of the liquid filling the gap between pad and substrate are the most important factors which determine the wet adhesion. Dry adhesion was found only in hairy pads, which occurs in geckos and spiders. It was demonstrated that van der Waals interaction is the dominant adhesive force in geckos＇ adhesion. The bio-inspired applications derived from adhesive pads are also reviewed.

While the nonholonomic robots with no-slipping constraints are studied extensively nowadays, the slipping effect is inevitable in many practical applications and should be considered necessarily to achieve autonomous navigation and control purposes especially in outdoor environments. In this paper the robust point stabilization problem of a tracked mobile robot is discussed in the presence of track slipping, which can be treated as model perturbation that violates the pure nonholonomic constraints. The kinematic model of the tracked vehicle is created, in which the slipping is assumed to be a time-varying pa- rameter under certain assumptions of track-soil interaction. By transforming the original system to the special chained form of nonholonomic system, the integrator backstepping procedure with a state-scaling technique is used to construct the controller to stabilize the system at the kinematic level. The global exponential stability of the final system can be guaranteed by Lyapunov theory. Simulation results with different initial states and slipping parameters demonstrate the fast convergence, robustness and insensitivity to the initial state of the proposed method.

Practical real-world scenarios such as the Internet,social networks,and biological networks present the challenges of data scarcity and complex correlations,which limit the applications of artificial *** graph structure is a typical tool used to formulate such correlations,it is incapable of modeling highorder correlations among different objects in systems;thus,the graph structure cannot fully convey the intricate correlations among *** with the aforementioned two challenges,hypergraph computation models high-order correlations among data,knowledge,and rules through hyperedges and leverages these high-order correlations to enhance the ***,hypergraph computation achieves collaborative computation using data and high-order correlations,thereby offering greater modeling *** particular,we introduce three types of hypergraph computation methods:①hypergraph structure modeling,②hypergraph semantic computing,and③efficient hypergraph *** then specify how to adopt hypergraph computation in practice by focusing on specific tasks such as three-dimensional(3D)object recognition,revealing that hypergraph computation can reduce the data requirement by 80%while achieving comparable performance or improve the performance by 52%given the same data,compared with a traditional data-based method.A comprehensive overview of the applications of hypergraph computation in diverse domains,such as intelligent medicine and computer vision,is also ***,we introduce an open-source deep learning library,DeepHypergraph(DHG),which can serve as a tool for the practical usage of hypergraph computation.
