By differential screening of pollen cDNA library of Zea mays, we obtained nine clones. The full length of one of these clones, designed zm401, was obtained by 5’ RACE. Within the full-length cDNA sequence, a clear open reading frame (ORF) was undectable by omega 2.0 and DNAMAN software. The longest potential ORF was 269 nucleotides long (791-1060), coding 89 AA, and it had a poly (A) tail. This indicate that zm401 was probably one of a growing number of non-coding mRNA-like RNA transcripts that exert their cellular functions directly as RNA. RT-PCR and Northern blot analyses showed the zm401 was transcribed from tetrad stage of microspore development and increased in concentration up to mature pollen. This suggests that zm401 likely functions in pollen growth and/or development. For analysis of the function of zm401 was ectopically expressed in tobacco and overexpressed in maize under the control of a pollen specific promoter zm13. Interestingly, all transgenic tobacco and maize plants showed various levels of sterility, ranging from abortive flower development to male sterility. Further analyses on anther development indicated multiple abnormalities of anther development at late stages. These abnormalities include lagged degradation of the tapetum and connective tissue, failed deposition of fibrous bands in endothecial cells, and aborted pollen grain development. These strongly suggest that zm401 has essential roles in anther development and its ability to perturb the anther development in heterogeneous dicotyledonous tobacco and maize indicates that zm401 functions in a molecular mechanism possibly conserved in both monocots and dicots.
