Follicle stimulating hormone β (FSHβ) of Japanese flounder (Paralichthys olivaceus) plays a key role in the regulation of gonadal *** study aimed to investigate molecular genetic characteristics of the FSHβ gene and elucidate the effects of single nucleotide polymorphisms (SNPs) of FSHβ on reproductive traits in Japanese *** used polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) and sequencing of the FSHβ gene in 60 *** identified only an SNP (T/C) in the coding region of exon3 of FSHβ.The SNP (T/C) did not lead to amino acid changes at the position 340 bp of FSHβ *** analysis showed that the SNP was significantly associated with testosterone (T) level and gonadosomatic index (GSI) (P<0.05).Individuals with genotype TC of the SNP had significantly higher serum T levels and GSI (P<0.05) than that of genotype ***, FSHβ gene could be a useful molecular marker in selection for prominent reproductive trait in Japanese Flounder.

Viscosity testing method of ultra-low temperature drilling fluids for polar glacier drilling

According to the working environment of the polar glacier core drilling, a separatory funnel capillary viscometer which can be used for testing the viscosity of liquids under ultra-low temperature conditions was de- signed. This viscometer has a simple structure and it is easy to operate, which can meet the testing requirements of different temperature conditions. The viscosity of the dimethyl silicone oil KF-96L-2.0cs was measured under different temperatures using this designed viseometer, and it is found that the viscometer coefficient K changes linearly with temperature. This testing method has relatively high test accuracy and its relative error is less than 4%, which can be used to test the viscosity of the different liquids in ultra-low temperature conditions.

Improvements of drilling fluid processing for cable-suspended electromechanical drills

Deep ice drilling in Polar Regions plays a key role in paleo-climate, microbiological and glaciological researches. For deep ice and subglacial coring the special purpose-built electromechanical cable-suspended drills with bottom-hole circulation are designed and explored. Used at present drilling fluid circulation systems are specified by very high fluid wastes during processing. Measurements in deep drilling projects in Antarctic and Greenland ice sheets showed that 25%-40% of the drilling fluid, pumped into the hole, is retrieved to the surface with cable and drill, and up to 45% of retrieved fluid goes to waste. This leads not only to increasing of the fluid consumption and cost of the project in general but also to severe contamination of air, surface and near-surface snow-firn layers at the drilling site. In order to reduce wastes and environmental impact, it is proposed to modify drilling fluid processing system by avoiding of evaporation and leakages of fluid as far as practicable. Drilling fluid is prepared and pumped into the hole according to predetermined program on semiautomatic mode, and volume and density of blended and pumped into the hole drilling fluid are measured continuously. Components of the drilling fluid are stored and mixed in the special closed containers equipped by pressure relief valves. Separation of drilling fluid from ice cuttings is carried out by two steps: firstly, fluid is separated with hydro extractor and then with thermal separator. To reduce the amount of removed drilling fluid with cable special squeezing collar is installed on the borehole mouth. All these arrangements are able to decrease drilling fluid wastes in many times.

[Objectives]This study was conducted to explore the relationship between morphological traits and body mass trait of Lutraria sieboldii.[Methods]110 were randomly selected from 120 2^(nd)-instar *** collected from the Tieshangang area of Beihai,Guangxi,and 132 were randomly selected from 150 shellfish at the instar of *** morphological traits were measured:shell length(SL),shell height(SH),shell width(SW),anterior length(AL),posterior length(PL),nose length(NL)in closed shell state,and maximum open shell width(OS)between two shells in closed shell state,and the body mass trait BM was also *** methods such as path analysis and multiple regression were used for data analysis,and the effects of these seven morphological traits on the body mass trait were studied,*** correlation between the tested seven quantitative traits and one body mass trait was all positive,all reaching an extremely significant level(P<0.01).[Results]The body mass trait of the shellfish at the instar of 2 had the highest correlation coefficient with shell length(0.922),that is,shell length had the greatest direct impact on the body mass trait;the path coefficient was 0.700;and the final multiple regression equation established was BM=-124.882+1.189 SL+1.551 SH+1.035 SW+0.119 NL,and the total determination coefficient(R^(2))on body mass was *** body mass trait of the shellfish at the instar of 0.6 had the highest correlation coefficient with shell length(0.859),that is,shell length had the greatest direct impact on the body mass trait;the path coefficient was 0.494;and the final multiple regression equation established was BM=-1.917+0.111 SL+0.021 NL+0.078 SW+0.032 OS,and the total determination coefficient(R~2)on body mass was *** multivariate regression variance analysis showed that the regression between the morphological traits and body mass trait of the *** at the instars of 2 and 0.6 reached an extremely significant level(P<0.01)

Cathelicidin是目前发现的一个最大抗菌肽家族,具有多重生物学功能。旨在探索和利用鱼类Cathelicidin抗菌机制和潜在的生物学功能。应用RACE-PCR技术获得日本鳗鲡I型Cathelicidin(Aj Cath I)基因的c DNA全长序列。Aj Cath I的c DNA全长为842 bp,开放阅读框为570 bp,编码189个氨基酸。氨基酸序列分析表明,Aj Cath I靠近C端有4个保守的半胱氨酸序列,抗菌成熟肽与香鱼(Plecoglossus altivelis)抗菌成熟肽相似性最高,同源性为65.57%。进化分析表明,Aj Cath I与其它鱼类亲缘关系较近,处于同一个分支上。将Aj Cath I基因克隆至p ET-28a载体,转化Escherichia coli BL21(DE3)宿主菌进行表达。结果表明,Aj Cath I以包涵体形式表达,经鎳柱纯化、Western blot验证和透析复性,获得高纯度的重组蛋白。

Cost-benefit analysis and breakeven point were used to analyze the production cost and economic benefit of tilapia pond monoculture and *** the aspects of tilapia production and economic benefit,this paper summarized the current production status and the characteristics of tilapia industry development and analyzed its developing *** results indicated that feed cost,pond rates and labor cost are the main production costs,and the production cost of polyculture is significantly lower than that of *** comparative analysis,it is proposed that the tilapia industry should be reasonably guided to the intensive,standardized and pollution-free ***,it is necessary to improve the coverage of improved varieties,strengthen disease prevention and control and promote the healthy farming model of *** also should strengthen the training of tilapia production techniques and the quality and safety testing of tilapia products,and accelerate the development of tilapia industrialization,so as to promote the sustainable and healthy development of tilapia industry.

织锦巴非蛤形态性状对体质量的影响

【目的】研究织锦巴非蛤(Paphia textile)野生群体形态性状对体质量性状的影响,为该贝类的人工选育提供参考。【方法】以广西北海铁山港海区采集的121只织锦巴非蛤为研究对象,分别测定其壳长(SL)、壳高(SH)、壳宽(SW)及体质量(BM)性状。以壳形态性状(SL、SH、SW)为自变量、体质量性状(BM)为因变量进行相关性分析、通径分析、决定程度分析和多元回归分析。【结果】参数统计分析结果表明,体质量变异系数最高。相关性分析结果表明,各性状间的相关性均达到极显著水平,体质量与壳宽的相关系数最大(0.882),壳长与壳宽的相关系数最小(0.596)。通径分析结果表明,壳宽对体质量的直接影响最大、通径系数为0.573,壳高通过壳宽对体质量的间接影响最大、通径系数为0.423;决定程度分析与通径分析结果变化趋势基本一致。运用多元回归分析方法建立了估计体质量的最优回归方程:BM=-32.258+0.309SL+0.356SH+1.456SW(R~2=0.874)。【结论】以体质量为选育目标开展织锦巴非蛤人工选育时,在形态性状上应首先考虑壳宽性状,同时加强壳高的协同选择。

为探究大黄鱼MARCH5A的免疫作用,实验采用反转录PCR确认了该基因的c DNA序列。该序列全长1045 bp,包含1个长度为867 bp的开放阅读框,编码288个氨基酸;序列分析显示该蛋白含1个RINGv和4个跨膜结构域。进化树分析表明大黄鱼有11个MARCH家族蛋白(与哺乳动物相同),但鱼类存在多个亚型,MARCH5A为鱼类特有蛋白,其蛋白理化性质和三维结构均与MARCH5B存在一定的差异。荧光定量PCR分析显示,MARCH5A在健康大黄鱼的多个组织中均有表达,在血液和心脏中表达量最高,其次为鳃和脑,而在肾脏和皮肤中表达量最少。刺激隐核虫感染大黄鱼后,MARCH5A在皮肤中早期表达量显著上调,第2天为对照组的9.65倍,后期下调。在鳃、脾脏和头肾中的前期表达量也有所增加,后期下降。结果表明大黄鱼MARCH5A在抗刺激隐核虫免疫应答过程中可能发挥重要作用。
