Colonization of germ-free mice with a mixture of three lactobacillus strains enhances the integrity of gut mucosa and ameliorates allergic sensitization

Increasing numbers of clinical trials and animal experiments have shown that probiotic bacteria are promising tools for allergy prevention. Here, we analyzed the immunomodulatory properties of three selected lactobacillus strains and the impact of their mixture on allergic sensitization to Bet v I using a gnotobiotic mouse model. We showed that Lactobacillus （L.） rhamnosus LOCK0900, L. rhamnosus LOCK0908 and L. casei LOCK0919 are recognized via Toll-like receptor 2 （TLR2） and nucleotide-binding oligomerization domain-containing protein 2 （NOD2） receptors and stimulate bone marrow-derived dendritic cells to produce cytokines in species- and strain-dependent manners. Colonization of germ-free （GF） mice with a mixture of all three strains （Lmix） improved the intestinal barrier by strengthening the apical junctional complexes of enterocytes and restoring the structures of microfilaments extending into the terminal web. Mice colonized with Lmix and sensitized to the Bet v I allergen showed significantly lower levels of allergen-specific IgE, IgG 1 and IgG2a and an elevated total IgA level in the sera and intestinal lavages as well as an increased transforming growth factor （TGF）-β level compared with the sensitized GF mice. Splenocytes and mesenteric lymph node cells from the Lmix-colonized mice showed the significant upregulation of TGF-β after in vitro stimulation with Bet v 1. Our results show that Lmix colonization improved the gut epithelial barrier and reduced allergic sensitization to Bet v 1. Furthermore, these findings were accompanied by the increased production of circulating and secretory IgA and the regulatory cytokine TGF-β. Thus, this mixture of three lactobacillus strains shows potential for use in the prevention of increased gut permeability and the onset of allergies in humans,

Integrative techniques using acupuncture,Chinese herbal medicine,diet,and supplements for polycystic ovary syndrome:a case report

Patients with a diagnosis of polycystic ovary syndrome （PCOS） are on the rise. About 4%-12% of women are currently estimated to have this condition. It is hypothesized that PCOS appears in women who have long-standing insulin resistance （1R）, which leads to high androgen and testosterone levels; this ultimately disrupts their menstrual cycles. Some researchers attribute IR to genetic factors, although there have been only minute changes in the human genome in the past 20 000 years. However, even with a stable gene pool, genes can be turned on and offby the environment, food and air quality and toxin exposure.

THE EVALUATION OF CHANGE IN CONCRETE STRENGTH DUE TO FABRIC FORMWORK

Fabric,as a flexible formwork for concrete,gives builders,engineers,and architects the ability to form virtually any *** technique produces a superb concrete surface quality that requires no further touch up or *** polyolefin fabrics are recommended for this *** texture of this fabric allows water from concrete mix to bleed,and therefore reduces the water-cement ratio of the *** to the reduction in the water-cement ratio,a higher compressive strength in fabric-formed concrete is achieved,which is also suggested by earlier *** current research study was conducted to investigate and document the changes in concrete strength and overall quality due to these woven polyolefin *** of fabric formwork will result in a decrease in construction cost,construction waste,and greenhouse gas *** sets of tests were conducted in this research study:a comparison of the compressive strength of fabric-formed versus PVC-formed concrete cylinders,and a comparison of the behaviour of the fabric-formed columns versus cardboard-formed reinforced concrete *** in this research were limited to two types of fabric that included one with coarse and one with a more refined texture,and two types of concrete that included ordinary and flyash *** laboratory results revealed that the effects of fabric formwork on concrete quality in a large member are limited mostly to the surface zone and the core of the concrete remains the same as a conventionally formed *** though fabric-formed cylinder tests showed an average of a 15%increase in compressive strength of the concrete samples,the compressive strength of the reinforced columns did not dramatically change when compared to the companion cardboard formed control *** research confirmed that fabric formwork is a structurally safe alterna-tive for forming reinforced concrete columns.

Leaf beetles Gastrophysa viridula have attracted recently increased research interest from various points of view, since they are：（i） pest insects in rhubarb crops; （ii） potential biocontrol agents of dock plants Rumex spp. in grasslands; and （iii） a model species in ecological studies on insect population dynamics, biochemistry, behavior, biomechanics and biomimetics. The continuous rearing of beetles at standardized conditions may help to unify the fitness state of different individuals, allowing a better comparison of experimental results. The present communication suggests a modular space- and time-saving rearing method ofG. viridula in stackable faunariums under laboratory conditions, which has been successfully established and continuously used over the last 5 years. Several developmental stages were kept in separate boxes, and multiple generations were kept simultaneously, depending on the required number of beetles.

Epigenetic modulations rendering cell-to-cell variability and phenotypic metastability

Tumor cells display phenotypic plasticity and heterogeneity due to genetic and epigenetic variations which limit the predictability of therapeutic interventions. Chromatin modifications can arise stochas- tically but can also be a consequence of environmental influences such as the microenvironment of cancer cells. A better understanding of the impact and dynamics of epigenetic modulation at defined chromosomal sites is required to get access to the underlying mechanisms. We investigated the epige- netic modulations leading to cell-to-cell heterogeneity in a tumor cell line model. To this end, we analyzed expression variance in 80 genetically uniform cell populations having a single-copy reporter randomly integrated in the genorne. Single-cell analysis showed high intraclonal heterogeneity. Epige- netic characterization revealed that expression heterogeneity was accompanied by differential histone marks whereas contribution of DNA methylation could be excluded. Strikingly, some clones revealed a highly dynamic, stochastically altered chromatin state of the transgene cassette which was accompanied with a metastable expression pattern. In contrast, other clones represented a robust chromatin state of the transgene cassette with a stable expression pattern. Together, these results elucidate locus-specific epigenetic modulation in gene expression that contributes to phenotypic heterogeneity of cells and might account for cellular plasticity.

Methodological impact of starch determination on starch content and ileal digestibility of starch in grain legumes for growing pigs

Background： Grain legumes represent a valuable energy source in pig diets due to their high starch content. The present study was conducted to determine the content and apparent ileal digestibility（AID） of starch in different grain legume cultivars for pigs by means of both a polarimetric and enzymatic method for starch ***： Three experiments were conducted with six barrows each which were fitted with ileal T-cannulas. In total, 18 diets including six different cultivars of faba beans（Vicia faba L.） and peas（Pisum sativum L.）, five different cultivars of lupins（Lupinus luteus L., Lupinus angustifolius L.）, and one diet with a soybean meal（SBM） were ***： The starch content of faba beans and peas was greater（P ＆lt; 0.05） when determined polarimetrically than enzymatically（438 vs. 345 g/kg dry matter（DM） in faba beans and 509 vs. 390 g/kg DM in peas, respectively）.Considerable lower starch contents were obtained in lupins and SBM, with 82 and 48 g/kg DM（analyzed polarimetrically） and ＆lt;1.1 and 3 g/kg DM（analyzed enzymatically）, respectively. Mean values for contents of neutral detergent fiber（NDF） and acid detergent fiber（ADF） in grain legumes ranged from 111 and 79 g/kg DM in peas to248 and 207 g/kg DM in lupins, respectively. Contents of condensed tannins in the colored flowered faba bean cultivars ranged from 2.1 to 7.4 g/kg DM. The AID of starch was greater（P ＆lt; 0.05） in pea than in faba bean cultivars,and using the polarimetric starch determination method resulted in greater（P ＆lt; 0.05） digestibility values than using enzymatic starch analysis（84 vs. 80% in faba beans and 86 vs. 83% in peas）. Moreover, AID of starch differed（P ＆lt; 0.05）within pea cultivars and starch digestibility in faba beans decreased linearly（P ＆lt; 0.05） as the content of condensed tannins increased. Effect of unburned carbon content in fly ash on the retention of 12 elements out of coal-combustion flue gas

The aim of this study was to evaluate whether unburned carbon particles present in fly ash can help in the retention of S, Cl, Br, As, Se, Cu, Ni, Zn, Ga, Ge, Rb, and Pb out of flue gas during the coal combustion at fluidised-bed power station where the coal was combusted along with limestone. The competitive influence of 10%-25% CaO in fly ashes on the distribution of studied elements was studied as well to be clear which factor governs behaviour of studied elements. Except of S （with significant association with CaO） and Rb and Pb （with major affinity to A1203） the statistically significant and positive correlation coefficients were calculated for the relations between unburned carbon content and Br （0.959）, Cl （0.957）, Cu （0.916）, Se （0.898）, Ni （0.866）, As （0.861）, Zn （0.742）, Ge （0.717）, and Ga （0.588） content. The results suggest that the unburned carbon is promising material in terms of flue gas cleaning even if contained in highly calcareous fly ashes.
