Trans-10,cis-12, not cis-9,trans-11, conjugated linoleic acid decreases ErbB3 expression in HT-29 human colon cancer cells

AIM： To examine whether trans-10,cis-12 CLA （t10C12） or cis-9,trans-11 CLA （c9t11） inhibits heregulin （HRG）-β- stimulated cell growth and HRG-β-ErbB3 signaling in HT-29 cells. METHODS： We cultured HT-29 cells in the absence or presence of the CLA isomers and/or the ErbB3 ligand HRG-β. MTT assay, [^3H]thymidine incorporation, Annexin V staining, RT-PCR, Western blotting, immunoprecipitation, and in vitro kinase assay were performed. RESULTS： HRG-β increased cell growth, but did not prevent DNA t10c12-induced growth inhibition. T10C12 inhibited DNA synthesis and induced apoptosis of HT-29 cells, whereas c9t11 had no effect. T10c12 decreased the levels of ErbB1, ErbB2, and ErbB3 proteins and transcripts in a dose-dependent manner, whereas c9t11 had no effect. Immunoprecipitation/ Western blot studies revealed that t10c12 inhibited HRG- β-stimulated phosphorylation of ErbB3, recruitment of the p85 subunit of phosphoinositide 3-kinase （PI3K） to ErbB3, ErbB3-associated PI3K activities, and phosphorylation of Akt. However, c9t11 had no effect on phospho Aid： levels. Neither t10c12 nor c9t11 had any effect on HRG-β-induced phosphorylation of ERK-1/2. CONCLUSION： These results indicate that the inhibition of HT-29 cell growth by t10c12 may be induced via its modulation of ErbB3 signaling leading to inhibition of Akt activation.

BACKGROUND The classification of uterine sarcomas is based on distinctive morphological and immunophenotypic characteristics,increasingly supported by molecular genetic *** on neurotrophic tyrosine receptor kinase(NTRK)gene fusionpositive uterine sarcoma,potentially aggressive and morphologically similar to fibrosarcoma,are limited due to its recent ***-TRK immunohistochemistry(IHC)analysis serves as an effective screening tool with high sensitivity and specificity for NTRK-fusion *** SUMMARY We report a case of a malignant mesenchymal tumor originating from the uterine cervix,which was pan-TRK IHC-positive but lacked NTRK gene fusions,accompanied by a brief literature review.A 55-year-old woman presented to the emergency department with abdominal pain and distension,exhibiting significant ascites and multiple solid pelvic *** examination revealed a tumor encompassing the uterine cervix,extending to the vagina and uterine corpus.A punch biopsy of the cervix indicated NTRK sarcoma with positive immunochemical pan-TRK ***,subsequent next generation sequencing revealed no NTRK gene fusion,leading to a diagnosis of poorly differentiated,advanced-stage *** The clinical significance of NTRK gene fusion lies in potential treatment with TRK inhibitors for positive *** such rare tumors is crucial due to the potential applicability of tropomyosin receptor kinase inhibitor treatment.

Entacapone,a catechol-O-methyltransferase inhibitor,can strengthen the therapeutic effects of levodopa on the treatment of Parkinson’s ***,few studies are reported on whether entacapone can affect hippocampal neurogenesis in *** investigate the effects of entacapone,a modulator of dopamine,on proliferating cells and immature neurons in the mouse hippocampal dentate gyrus,60 mice(7 weeks old)were randomly divided into a vehicle-treated group and the groups treated with 10,50,or 200 mg/kg *** results showed that 50 and 200 mg/kg entacapone increased the exploration time for novel object *** staining results revealed that after entacapone treatment,the numbers of Ki67-positive proliferating cells,doublecortin-positive immature neurons,and phosphorylated cAMP response element-binding protein(pCREB)-positive cells were significantly *** blot analysis results revealed that treatment with tyrosine kinase receptor B(TrkB)receptor antagonist significantly decreased the exploration time for novel object recognition and inhibited the expression of phosphorylated TrkB and brain-derived neurotrophic factor(BDNF).Entacapone treatment antagonized the effects of TrkB receptor *** results suggest that entacapone treatment promoted hippocampal neurogenesis and improved memory function through activating the BDNF-TrkB-pCREB *** study was approved by the Institutional Animal Care and Use Committee of Seoul National University(approval ***-130730-1)on February 24,2014.

Recently,ambient vibration test(AVT)is widely used tu estimate dynamic characteristics of large civil struc- *** characteristics ean be affected by various envirnnmental factors such as humidity,intensity of wind,and *** these environmental conditions,tire mass of vehicles may change the measured valnes when traffic-in- duced vibration is used as a source of AVT tor *** effect of vehicle mass on dynamic characteristics is investigated through traffic-induced vibration tests on three bridges;(1)three-span suspension bridge(128m+404m+128m),(2) five-span continuous steel box girder bridge(59m+3@ 95m+59m),(3)simply supported plate girder bridge(46m). Acceleration histories of each measurement location under normal traffic are recorded for 30 minutes at *** recor- ded histories are divided into individual vibrations and are combined into two groups aceording to the level of vibration;one by heavy vehicles such as trucks and buses and the other by light vehicles such as passenger *** processing of the two groups of signals shows that,for the middle and long-span bridges,the difference can be hardly detected,but,for the short span bridges whose mass is relatively small,the measured natural frequencies can change up to 5.4%.

Effect of asymmetrical channel chirping on field intensity patterns of supermodes and modal discrimination in index guided laser arrays

The effect of channel-width chirping on near- and far-field intensity patterns of the six supermodes was investigated. The supermode discrimination was evaluated in various channel-chirped index guided laser arrays. The results show that the linearly channel-chirped laser array has very good supermode discrimination which is better than that of a uniform laser array, the V channel-chirped laser array has the smallest radiation angle of the fundamental supermode among the calculated arrays, and the asymmetrically V channel-chirped array has a very small radiation angle of the fundamental supermode, which is smaller than that of the uniform array and also allows for very good fundamental supermode discrimination against the higher-order supermodes, which is better than that of the V channel-chirped laser array.

Adaptive Data Transmission Method According to Wireless State in Long Range Wide Area Networks

The Internet of Things(IoT)has enabled various intelligent services,and IoT service range has been steadily extended through long range wide area communication technologies,which enable very long distance wireless data ***-nodes are connected to a gateway with a single *** consume very low-power,using very low data rate to deliver *** long transmission time is consequently needed for each data packet transmission in long range wide area networks,data transmission should be efficiently ***,this paper proposes a multicast uplink data transmission mechanism particularly for bad network *** delay will be increased if only retransmissions are used under bad network ***,employing multicast techniques in bad network conditions can significantly increase packet delivery ***,retransmission can be reduced and hence transmission efficiency ***,the proposed method adopts multicast uplink after network condition *** predict network conditions,the proposed method uses a deep neural network *** proposed method performance was verified by comparison with uplink unicast transmission only,confirming significantly improved performance.

Recommendation application for video head impulse test based on fuzzy logic control

Vestibulo-ocular reflex(VOR) is an important biological reflex that controls eye movement to ensure clear vision while the head is in ***,VOR measurement is commonly done with a video head impulse test based on a velocity gain algorithm or a position gain algorithm,in which velocity gain is a VOR calculation on head and eye velocity,whereas position gain is calculated from head and eye *** aim of this work is first to compare the two algorithms' performance and to detect covert catch-up saccade,then to propose a stand-alone recommendation application for the patient's *** the first experiment,for ipsilesional and contralesional sides,the calculated position gain(0.94±0.17) is higher than velocity gain(0.84±0.19).Moreover,gain asymmetry of both lesion and intact sides using velocity gain is mostly higher than that from using position gain(four out of five subjects).Consequently,for subjects who have unilateral vestibular neuritis diagnosed from clinical symptoms and a vestibular function test,vestibular weakness is depicted by velocity gain much better than by position *** catch-up saccade and position gain then are used as inputs for recommendation applications.

A Data Download Method from RSUs Using Fog Computing in Connected Vehicles

Communication is important for providing intelligent services in connected *** must be able to communicate with different places and exchange information while *** service operation,connected vehicles frequently attempt to download large amounts of *** can request data downloading to a road side unit(RSU),which provides infrastructure for connected *** RSU is a data bottleneck in a transportation system because data traffic is concentrated on the ***,it is not appropriate for a connected vehicle to always attempt a high speed download from the *** the mobile network between a connected vehicle and an RSU has poor connection quality,the efficiency and speed of the data download from the RSU is *** problem affects the quality of the user ***,it is important for a connected vehicle to connect to an RSU with consideration of the network conditions in order to try to maximize download *** proposed method maximizes download speed from an RSU using a machine learning *** collect and learn from network data,fog computing is used.A fog server is integrated with the RSU to perform *** the algorithm recognizes that conditions are not good for mass data download,it will not attempt to download at high ***,the proposed method can improve the efficiency of high speed *** conclusion was validated using extensive computer simulations.
