Synthesis and pharmacological properties of naturally occurring prenylated and pyranochalcones as potent anti-inflammatory agents

An efficient approach has been developed for the synthesis of naturally occurring prenylated chalcones viz. kanzonol C （1）, stipulin （2）, crotaorixin （3）, medicagenin （4）, licoagrochalcone A （5） and abyssinone D （6） along with the pyranochalcones paratocarpin C （7）, anthyllisone （8） and 3-O-methylabyssinone A （9）. The key step of the synthesis is a Claisen-Schmidt condensation. Subsequently, their anti-inflammatory effects were investigated in lipopolysaccharides （LPSs）-induced RAW-264.7 macrophages. Of the synthesized chalcones, compounds 5 （IC50= 10.41 μmol[L）, 6 （IC50= 9.65 μmol/L） and 8 （IC50= 15.34 μmol/L） show remarkable activity with no cytotoxicity. Compound 9 （IC50 = 4.5 μmol/L） exhibits maximum （83.6%） nitric oxide （NO） inhibition, but shows slight cytotoxicity. The results reveal that the chalcones bearing the prenyl group at 3- and/or 5-position on ring A （acetophenone moiety）, i.e., 1-4 and 7 show weak, or no inhibition activity, whereas chalcones having the prenyl group only on ring B （aldehyde part）, i.e., 5, 6 and 8 show significant activity on the production of inflammatory mediated NO with no cytotoxicity.

The use of artificial nest boxes by Siberian flying squirrels (Pteromys volans) in South Korea

In this study, we examined the use of artificial nest boxes by Siberian flying squirrels (Pteromys volans) in three coniferous and mixed forests in Gangwon Province, South Korea. Six hundred and twelve boxes with different sized entry holes (ranging from 3 to 7 cm in diameter) were placed in the forests between 2004 and 2009. Pteromys volans used nine boxes in the coniferous forests and two boxes in the mixed forests. The squirrels only used boxes with entrance holes measuring 3.5, 4, and 5 cm in diameter, showing a strong and moderate preference for boxes with 5 and 4-cm holes, respectively, and a strong avoidance for boxes with 3- and 7-cm holes. Therefore, we suggest placing artificial nest boxes with entrance holes 5 cm in diameter to encourage breeding activity. Most nests made in the artificial boxes were composed of fibrous materials from woody vines. We recommend placing artificial nest boxes with holes of 5-cm diameter in coniferous forests, which support dense populations of P. volans, to survey whether this approach would positively affect the breeding habits and population maintenance of this species.

Design of a novel 50kW fast charger for electric vehicles

A novel 50 kW fast charger was proposed for electric vehicles. The proposed fast charger is divided into two main sections an AC-DC converter performing a PFC function and a DC-DC converter performing a charging function. A transformer including leakage inductances was used in the AC-DC converter in order to obtain isolation and inductance. A series-connection topology was used in the DC-DC converter between the DC-bus and outlet. This topology enables high power conversion efficiency up to 95% for the DC-DC converter. In order to reduce the impact of the 50 kW charging on the AC grid, the proposed fast charger system includes a buffering battery unit between the two main power conversion units. This leads to reductions in the power installation costs of power companies and to improvements in the power quality were verified through simulations and experimental results. on the AC grid. Effects of ethanol on insulin-like growth factor-Ⅰ system in primary cultured rat hepatocytes: Implications of JNK1/2 and alcoholdehydrogenase

AIM: To evaluate the effects of ethanol on the insulin- like growth factor-Ⅰ (IGF-Ⅰ) system involved in c-Jun N-terminal kinase (JNK1/2) and alcoholdehydrogenase (ADH) activity in primary cultured rat hepatocytes. METHODS: Hepatocytes isolated from male Sprague-Dawley rats were incubated with various concentrations of ethanol for different durations of time. The cells were pretreated with SP600125 (10 μmol/L) and 4-MP (200 μmol/L), and then treated with ethanol (200 mmol/L). We then measured IGF-Ⅰ secretion, IGF-Ⅰ mRNA expression, cell viability and JNK1/2 activity by radioimmunoassay, RT-PCR, MTT assay and Western blot, respectively (n = 6). RESULTS: Ethanol induced the activity of phospho (p)-JNK1/2, reaching a maximum at 60 min and then decreasing at 180 min. The effects of ethanol on the IGF-Ⅰ system were increased at 60 min (secretion: 7.11 ± 0.59 ng/mg protein vs 4.91 ± 0.51 ng/mg, mRNA expression: 150.2% ± 10.2% vs 101.5% ± 11.3%, P = 0.045) and then decreased at 180 min (secretion: 3.89 ± 0.25 ng/mg vs 5.4 ± 0.54 ng/mg protein; mRNA expression: 41.5% ± 10.4% vs 84.7% ± 12.1%, P = 0.04), however cell viability was decreased in a dose- and time-dependent manner. SP600125 blocked the ethanol-induced changes (at 60 min). Additionally, 4-methylpyrazole prevented the ethanol-induced decreases in the IGF-Ⅰ system, cell viability and p-JNK1/2 activity (at 180 min). CONCLUSION: This study suggests that ethanol- induced p-JNK1/2 activation is associated with the IGF-Ⅰ system and cell viability in hepatocytes. Furthermore, alcohol dehydrogenase is involved in the relationship between ethanol-induced inactivation of p-JNK1/2 and the changes of the IGF-Ⅰ system and cell viability.

Improvement of Shape Recognition Performance of Sendzimir Mill Control Systems Using Echo State Neural Networks

High rigidity twenty-high Sendzimir mills （ZRMs） are widely used for rolling stainless steels, silicon sheets, etc. A ZRM uses a small diameter work roll to produce massive rolling forces. Since a work roll with a small diameter can be bent easily, strips often have complex shapes with mixed quarter and deep edge waves in the shape of plates. In order to solve this problem, fuzzy neural network controls are generally used for shape： recognition in ZRM control systems. Among various neural network types, the multi-layer perceptron （MLP） is typically used in current ZRMs. However, an MLP causes the loss of a large amount of shape recognition data. To improve the shape recognition per- formance of ZRM control systems, echo state networks （ESNs） are proposed to be used. Through simulation re- sults, it is found that shape recognition performance could be improved using the proposed ESN method.

Objective:To develop the rapid and efficient quantitative detection tool for nervous necrosis virus isolated from sevenband grouper Hyporhodus ***:The viral genes of the NNV(SGYeosu08) isolated from sevenband grouper were phylogenetically *** addition,novel quantitative PCR primers based on the genomic sequence of SGYeosu08 isolate were designed and compared it with the conventional bio-assay method(TCID_(50)) using in vitro and in vivo ***:The phylogenetic analysis of viral genes demonstrated the relationship of SGYeosu08 with members of red-spotted grouper nervous necrosis virus(RGNNV).The qNNV_Rl primer set(R1_F and R1_R) and the qNNV_R2 primer set(R2_F and R2_R) revealed 93%primer efficiency(regression:y=-0.2861 x + 9.9401,R^2= 0.9976)and the revealed 108%primer efficiency(regression:y=-0.3172 x + 10.0611,R^2= 0.9982),*** comparison with viral infectivity calculated by TCID_(50) method showed similar kinetic pattern at in vitro and NNV challenged fish(in vivo) ***:Result show that this method is rapid and efficient to diagnose NNV infection compare to traditional bioassay method(TCID_(50)).

Characteristics of radiation and convection heat transfer in indirect near-infrared-ray heating chamber

Numerical study was performed to evaluate the characteristics of combined heat transfer of radiation, conduction and convection in indirect near infrared ray （N/R） heating chamber. The effects of important design parameters such as the shape of heat absorbing cylinder and heat releasing fin on the pressure drop and heat transfer coefficient were analyzed with different Reynolds numbers. The Reynolds numbers were varied from 103 to 3x106, which was defined based on the hydraulic diameter of the heat absorbing cylinder. Analyses were performed to obtain the inner and outer flow and the temperature distributions in the heat absorbing cylinder and the rates of radiation heat transfer and convection heat transfer. As the Reynolds number increases, the convection heat transfer rate is increased while the radiation heat transfer rate is decreased. The average convection heat transfer rate follows a power rule of the Reynolds number. Addition of three-dimensional heat releasing fin to the outside of the heat absorbing cylinder enhances the convection heat transfer.
