Cancer occurrence and development are closely related to increased lipid produc-tion and glucose *** are the basic component of the cell membrane and play a significant role in cancer cell processes such as cell-to-cell recognition,signal transduction,and energy supply,which are vital for cancer cell rapid proliferation,invasion,and *** regulatory element-binding transcription factor 1(SREBP1)is a key transcription factor regulating the expression of genes related to cholesterol biosynthesis,lipid homeostasis,and fatty acid *** addition,SREBP1 and its upstream or downstream target genes are implicated in various metabolic diseases,particularly ***,no review of sREBP1 in cancer biology has yet been ***,we summarized the roles and mechanisms of SREBP1 biological processes in cancer cells,including SREBP1 modification,lipid metabolism and reprogramming,glucose and mitochondrial metabolism,immunity,and tumor microenvi-ronment,epithelial-mesenchymal transition,cell cycle,apoptosis,and ***-ally,we discussed the potential role of SREBP1 in cancer prognosis,drug response such as drug sensitivity to chemotherapy and radiotherapy,and the potential drugs targeting SREBP1 and its corresponding pathway,elucidating the potential clinical application based on SREBP1 and its corresponding signal pathway.

Poly(butylene adipate-co-terephthalate)(PBAT), a widely studied biodegradable material, has not effectively addressed the problem of plastic waste. Taking into consideration the cost-effectiveness, upcycling PBAT should take precedence over direct composting degradation. The present work adopts a chain breaking-crosslinking strategy, upcycling PBAT into dual covalent adaptable networks(CANs). During the chainbreaking stage, the ammonolysis between PBAT and polyethyleneimine(PEI) established the primary crosslinked network. Subsequently, styrene maleic anhydride copolymer(SMA) reacted with the hydroxyl group, culminating in the formation of dual covalent adaptable networks. In contrast to PBAT, the PBAT-dual-CANs exhibited a notable Young's modulus of 239 MPa, alongside an inherent resistance to creep and solvents. Owing to catalysis from neighboring carboxyl group and excess hydroxyl groups, the PBAT-dual-CANs exhibited fast stress relaxation. Additionally,they could be recycled through extrusion and hot-press reprocessing, while retaining their biodegradability. This straightforward strategy offers a solution for dealing with plastic waste.

以城市受污染河道的底泥为研究对象,首先掺入不同比例的单独固化剂(水泥)和复合固化剂(水泥+CaO_(2))对其进行固定,检测固化底泥的无侧限抗压强度、静置和降雨冲刷条件下底泥氮磷的释放特征。在此基础上分别挑选两组中固化效果较优的掺量(50%水泥以及50%水泥+10%CaO_(2)),进一步加入不同量的粉煤灰,探究在粉煤灰协同下水泥和CaO_(2)固化底泥的效果。结果表明,单独水泥固化和水泥+CaO_(2)固化均使底泥成块且具有较强的抗压能力,并且明显降低了底泥中溶解态无机磷(DIP)向上覆水的释放,在静置和降雨冲刷条件下均有较好的固化效果。此外,水泥和CaO_(2)改变了底泥中磷的形态,但水泥并未促进非磷灰石无机磷(NAIP)向磷灰石无机磷(AP)的转化,而加入CaO_(2)后促进了NAIP向AP的转化。加入粉煤灰后进一步提高了水泥和CaO_(2)的固化效果,其中固化剂最优配比为水泥∶CaO_(2)∶粉煤灰=5∶1∶2,与对照组相比,在实验后期(13~20 d)底泥固化后,上覆水中氨氮去除率为86.33%,DIP去除率为90.5%。水泥、CaO_(2)和粉煤灰的协同固化处理能够有效抑制底泥内源氮磷的释放,减少底泥对环境的二次污染。

Perovskite solar cell has gained widespread attention as a promising technology for renewable ***, their commercial viability has been hampered by their long-term stability and potential Pb leakage. Herein, we demonstrate a bifunctional passivator of the potassium tartrate(PT) to address both challenges. PT minimizes the Pb leakage in perovskites and also heals cationic vacancy defects, resulting in improved device performance and stability. Benefiting from PT modification, the power conversion efficiency(PCE) is improved to 23.26% and the Pb leakage in unencapsulated films is significantly reduced to 9.79 ppm. Furthermore, the corresponding device exhibits no significant decay in PCE after tracking at the maximum power point(MPP) for 2000 h under illumination(LED source, 100 mW cm^(-2)).

Metastatic pancreatic cancer(mPC)has a dismal ***,we conducted a prospective,multicentre,single-arm,phase II trial evaluating the efficacy and safety of penpulimab and anlotinib in combination with nab-paclitaxel/gemcitabine(PAAG)in patients with first-line mPC(NCT05493995).The primary endpoints included the objective response rate(ORR)and disease control rate(DCR),while secondary endpoints encompassed progression-free survival(PFS),overall survival(OS),and *** 66 patients analysed for efficacy,the best response,indicated by the ORR,was recorded at 50.0%(33/66)(95%CI,37.4–62.6%),with 33 patients achieving partial response(PR).Notably,the DCR was 95.5%(63/66,95%CI,87.3–99.1%).The median PFS(mPFS)and OS(mOS)were 8.8(95%CI,8.1–11.6),and 13.7(95%CI,12.4 to not reached)months,*** 3/4 treatment-related adverse events(TRAEs)were reported in 39.4%of patients(26/66).In prespecified exploratory analysis,patients with altered SWI/SNF complex had a poorer ***,low serum CA724 level,high T-cell recruitment,low Th17 cell recruitment,and high NK CD56dim cell scores at baseline were potential predicative biomarkers for more favourable *** conclusion,PAAG as a first-line therapy demonstrated tolerability with promising clinical efficacy for *** biomolecular findings identified in this study possess the potential to guide the precise clinical application of the triple-combo regimen.

The use of metal-organic frameworks(MOFs)as solid adsorption materials for carbon capture is promising,but achieving efficient and reversible adsorption with a balance of capacity and selectivity for carbon dioxide(CO_(2))over N_(2) remains a *** take full advantage of the strong channel traffic and robustness of MOFs with relatively small pores,it is highly necessary to employ a defect-engineering strategy to construct a broader channel structure that can facilitate the loading of functional motif-rich amino acids(AAs).This strategy can greatly enhance the CO_(2) adsorption performance of *** this study,motif-rich amino acids are loaded into the defective and robust porous frameworks via combined defect-engineering and post-synthetic *** defective Zr/Hf-MOF-808s modified with AAs,especially for the 18 mol%4-nitroisophthalic acid,generated defective products allowing for the loading of L-serine(L-Ser).This modification resulted in a significant improvement in both the adsorption capacity(248%improvement at 298 K,100 kPa)and the selectivity of CO_(2)/N_(2) using the ideal adsorbed solution theory(IAST),with the selectivity increasing to 120.55 and 38.27 at 15 and 100 kPa,respectively,while maintaining good cycling *** functional theory(DFT)simulation,CO_(2) temperature-programmed desorption(CO_(2)-TPD),and in situ Fourier transform infrared spectroscopy(FTIR)were further employed to have a better understanding of the enhanced CO_(2) adsorption ***,unlike the AAs loaded pristine MOF-808s that showed the best CO_(2) adsorption capacity with the loading of short and small glycine(Gly),the broadened channel size in our work enables the loading of functional motif-rich L-serine,which brings more active binding sites,improving CO_(2) adsorption.

Gene upregulation through genome editing is important for plant research and breeding. Targeted insertion of short transcriptional enhancers (STEs) into gene promoters may offer a universal solution akin to transgene-mediated overexpression while avoiding the drawbacks associated with transgenesis. Here, we introduce an "in locus activation"technique in rice that leverages well-characterized STEs for refined, heritable, and multiplexed gene upregulation. To address the scarcity of potent enhancers, we developed a large-scale mining approach and discovered a suite of STEs that are capable of enhancing gene expression in rice protoplasts. The in locus integration of these STEs into eight rice genes resulted in substantial transcriptional upregulation in the edited plants, with up to 869.1-fold increases in their transcript levels. Employing a variety of STEs, we achieved delicate control of gene expression, enabling the fine-tuning of key phenotypic traits such as plant height. Our approach also enabled efficient multiplexed gene upregulation, with up to four genes activated simultaneously, significantly enhancing the nicotinamide mononucleotide metabolic pathway. Importantly, heritability studies from the T0 to T3 generations confirmed the stable and heritable nature of STE-driven gene activation. Collectively, our work demonstrates that coupled with STE mining, leveraging genome editing for in locus activation and gene upregulation holds great promise to be widely adopted in fundamental plant research and crop breeding.

OBJECTIVE:To evaluate the effect of dahuang Mudan Tang(大黄牡丹汤,DHMD)and allopurinol on the treatment of chronic kidney disease staged G1-G3b patients with hyperuricemia and to provide novel insights into the clinical management of chronic kidney disease ***:A total of 80 chronic kidney patients staged G1-G3b with hyperuricemia were randomly grouped to receive single allopurinol treatment(control)and combined treatment with DHMD(treated)for 8 *** kidney function and proteinuria indicators of patients were compared between pre-and *** oxidative stress and inflammation responses were evaluated by corresponding indicators and *** clinical efficiency rate and adverse reaction events were also summarized to assess the therapeutic efficiency and ***:The kidney function and proteinuria of enrolled patients were alleviated after their therapies,behaved as the increasing estimated glomerular filtration rate and decreasing serum creatinine,serum uric acid,urea nitrogen,24 h urine protein *** the other hand,the malondialdehyde level and pro-inflammation cytokines were suppressed by the therapies,and the superoxide dismutase was found to be significantly *** in the treated groups showed a better recovery in kidney function,proteinuria,oxidative stress,and inflammation ***,patients in the treated group showed a higher efficiency rate(95%)and fewer adverse reaction events(5%).CONCLUSIONS:The combination of allopurinol with DHMD significantly promoted the recovery of chronic kidney disease stage G1-G3b patients with hyperuricemia,which can be considered a novel clinical therapeutic strategy.
