Association of Caveolin-1 polymorphisms with colorectal cancer susceptibility in Taiwan

AIM: To investigate the association of Caveolin-1 (Cav-1) polymorphisms with colorectal cancer (CRC) risk in a central Taiwan Residents population. METHODS: Three hundred and sixty-two patients with colorectal cancer and the same number of recruited age-and gender-matched healthy controls were genotyped. And only those matches with all single nucleotide poly-morphisms data (case/control = 362/362) were selected for final analyzing. RESULTS: There were significant differences between CRC and control groups in the distributions of their genotypes (P = 1.6 × 10 -12 and 3.0 × 10 -4 ) and allelic frequencies (P = 2.3 × 10 -13 and 4.0 × 10 -5 ) in the Cav-1 G14713A (rs3807987) and T29107A (rs7804372) poly-morphisms respectively. As for the haplotype analysis,those who had GG/AT or GG/AA at Cav-1 G14713A/ T29107A showed a 0.68-fold (95% CI: 0.48-0.98) de- creased risk of CRC compared to those with GG/TT,while those of any other combinations were of increased risk. There were joint effects of Cav-1 G14713A and T29107A genotype with smoking status on individual CRC susceptibility. CONCLUSION: This is the first report providing evidence of Cav-1 being involved in CRC and it may be novel useful genomic markers for early detection of CRC.

In this work, nitrogen-doped cobalt nanoparticlesinitrogen-doped plate-like ordered mesoporous carbons (N/Co/OMCs) were used as noble-metal free electrocatalysts with high catalytic efficiency. Compared with OMCs with long channel length, due to more entrances for catalytic target accessibility and a short pathway for rapid diffusion, the utilization efficiency of cobalt nanoparticles inside the plate-like OMCs with short pore length is well improved, which can take full advantage of porous structure in electrocatalysis and increase the utilization of catalysts. The active sites in N/Co/OMCs for oxygen reduction reaction (ORR) are highly exposed to oxygen molecule, which results in a high activity for ORR. By combination of the catalytic properties of nitrogen dopant, incorporation of Co nanoparticles, and structural properties of OMCs, the N/Co/plate-like OMCs are highly active noble-metal free catalysts for ORR in alkaline solution. Measurement of electrical charges carried by airborne bacteria laboratory-generated using a single-pass bubbling aerosolizer

Widely used bioaerosol generators like Collison nebulizer probably produce electrostatically charged par- ticles, but the electrical charges carried by laboratory-generated airborne microorganisms using bubbling aerosolizers are poorly understood. In this study, we measured the fraction of neutral particles and num- ber of elementary charges per particle as a function of the aerodynamic diameter of airborne bacteria （Escherichia coli and Enterococcus hirae）. Bioaerosols were produced by a liquid sparging aerosolizer-type bubbling generator, with particle sizes ranging from roughly 0.6 to 2 p^m. The experimental setup included an electrostatic precipitator and real-time devices including an electrometer, aerodynamic particle sizer, and electrical low-pressure impactor. Experimental results obtained for various operating conditions showed that aerosols produced with a higher bubbling airflow contained a larger proportion of neutral particles （from around 30% to 50%） and that bacteria carried a greater average absolute number of elementary charges （from around -10 to -60 elementary units） than those under lower airflow. Under the investigated conditions, a neutralization step is unnecessary because it may have a negative effect on the viability of sensitive microorganisms. Our results suggest that the neutral fraction can be used down- stream of an electrostatic precipitator, and that this setup may have advantages over bipolar neutralizers.

Pancreatic cancer(PC) is the most aggressive type of common cancers, and in 2014, nearly 40000 patients died from the disease in the United States. Pancreatic ductal adenocarcinoma, which accounts for the majority of PC cases, is characterized by an intense stromal desmoplastic reaction surrounding the cancer cells. Cancer-associated fibroblasts(CAFs) are the main effector cells in the desmoplastic reaction, and pancreatic stellate cells are the most important source of CAFs. However, other important components of the PC stroma are inflammatory cells and endothelial cells. The aim of this review is to describe the complex interplay between PC cells and the cellular and noncellular components of the tumour stroma. Published data have indicated that the desmoplastic stroma protects PC cells against chemotherapy and radiation therapy and that it might promote the proliferation and migration of PC cells. However, in animal studies, experimental depletion of the desmoplastic stroma and CAFs has led to more aggressive cancers. Hence, the precise role of the tumour stroma in PC remains to be elucidated. However, it is likely that a contextdependent therapeutic modification, rather than pure depletion, of the PC stroma holds potential for the development of new treatment strategies for PC patients.

AIM To determine whether it is possible to identify different immune phenotypic subpopulations of cancer-associated fibroblasts (CAFs) in pancreatic cancer (PC). METHODS We defined four different stromal compartments in surgical specimens with PC: The juxtatumoural, peripheral, lobular and septal stroma. Tissue microarrays were produced containing all pre-defined PC compartments, and the expression of 37 fibroblast (FB) and 8 extracellular matrix (ECM) markers was evaluated by immunohistochemistry, immunofluorescence (IF), double-IF, and/or in situ hybridization. The compartment-specific mean labelling score was determined for each marker using a four-tiered scoring system. DOG1 gene expression was examined byquantitative reverse transcription PCR (qPCR). RESULTS CD10, CD271, cytoglobin, DOG1, miR-21, nestin, and tenascin C exhibited significant differences in expression profiles between the juxtatumoural and peripheral compartments. The expression of CD10, cytoglobin, DOG1, nestin, and miR-21 was moderate/strong in juxtatumoural CAFs (j-CAFs) and barely perceptible/weak in peripheral CAFs (p-CAFs). The upregulation of DOG1 gene expression in PC compared to normal pancreas was verified by qPCR. Tenascin C expression was strong in the juxtatumoural ECM and barely perceptible/weak in the peripheral ECM. CD271 expression was barely perceptible in j-CAFs but moderate in the other compartments. Galectin-1 was stronger expressed in j-CAFs vs septal fibroblasts, PDGFRβ, tissue transglutaminase 2, and hyaluronic acid were stronger expressed in lobular fibroblasts vs p-CAFs, and plectin-1 was stronger expressed in j-CAFs vs l-FBs. Liquid biopsy is a highly promising method for non-invasive detection of tumor-associated nucleic acid fragments in body fluids but is challenged by the low abundance of nucleic acids of clinical interest and their sequence homology with the vast background of nucleic acids from healthy ***,programmable endonucleases such as clustered regularly interspaced short palindromic repeats(CRISPR)associated protein(Cas)and prokaryotic Argonautes have been successfully used to remove background nucleic acids and enrich mutant allele fractions,enabling their detection with deep next generation sequencing(NGS).However,the enrichment level achievable with these assays is limited by futile binding events and off-target *** overcome these shortcomings,we conceived a new assay(Programmable Enzyme-Assisted Selective Exponential Amplification,PASEA)that combines the cleavage of wild type alleles with concurrent polymerase *** PASEA increases the numbers of both wild type and mutant alleles,the numbers of mutant alleles increase at much greater rates,allowing PASEA to achieve an unprecedented level of selective enrichment of targeted *** combining CRISPR-Cas9 based cleavage with recombinase polymerase amplification,we converted samples with0.01%somatic mutant allele fractions(MAFs)to products with 70%MAFs in a single step within 20 min,enabling inexpensive,rapid genotyping with such as Sanger ***,PASEA's extraordinary efficiency facilitates sensitive real-time detection of somatic mutant alleles at the point of care with custom designed Exo-RPA ***-time PASEA'performance was proved equivalent to clinical amplification refractory mutation system(ARMS)-PCR and NGS when testing over hundred cancer patients'*** strategy has the potential to reduce the cost and time of cancer screening and genotyping,and to enable targeted therapies in resource-limited settings.

It is proved in this paper that if G is a simple connected r-uniform hypergraph with G ≥ 2, then G has an edge e such that G-e-V1(e) is also a simple connected r-uniform hypergraph. This reduction is naturally called a combined Graham reduction. Under the simple reductions of single edge removals and single edge contractions, the minor minimal connected simple r-uniform hypergraphs are also determined.

The Clitocybaceae is a recently established ***,the infrafamilial divisions and relationships within the family are vague due to limited sampling and genes employed for phylogenetic *** mushrooms of the family contain the neurotoxic muscarine,which has caused many severe and even deadly poisonings ***,the taxonomic distribution and evolution of the toxin within the family is largely *** this study,phylogenetic analyses based on nucleotide sequences of ITS and of six molecular loci(ITS,LSU,TEF1,RPB1,RPB2 and ATP6),plus a phylogenomic analysis based on 485 single-copy orthologous genes,were performed to reconstruct the framework of *** analysis was used to estimate the divergence times within the ***,biochemical analysis for muscarine was conducted of 32 representative *** on these analyses,an updated classification of Clitocybaceae into six genera(Clitocybe,Collybia,Dendrocollybia,Lepista,Pseudolyophyllum,and Singerocybe)is *** genus Collybia is emended to accommodate four subgenera(Collybia,Crassicybe,Leucocalocybe,and Macrosporocybe).Seventeen new Chinese species and 15 new combinations are *** to the genera of Clitocybaceae and the subgenera of Collybia,as well as to the known species of Clitocybe and Collybia *** in China,are *** addition,muscarine was detected in 18 species,and these muscarine-containing species formed a major monophyletic clade within Collybia ***,our phylogenetic,phylogenomic,chemotaxonomic and molecular dating results indicate that the Clitocybaceae is a natural group estimated to have arisen some 60 million years ago,and in this family,muscarine has evolved only once circa 20 million years ago without later losses.
