Establishing and clinically validating a machine learning model for predicting unplanned reoperation risk in colorectal cancer

Functional switching of ATM:sensor of DNA damage in proliferating cells and mediator of Akt survival signal in post-mitotic human neuron-like cells

Ataxia-telangiectasia(A-T) is an autosomal recessive disorder characterized by cerebellar ataxia and oculocutaneous *** gene mutated in this disease,ATM(A-T,mutated),encodes a 370-kda Ser/Thr protein *** not only mediates cellular response to DNA damage but also acts as an activator of Akt in response to ***,despite intensive studies,the mechanism underlying the neuronal degeneration symptoms of human A-T is still poorly *** found that the topoisomerase inhibitors etoposide and camptothecin readily induced apoptosis in undifferentiated proliferating SH-SY5Y cells but could not induce apoptosis in neuronally differentiated SH-SY5Y *** addition,etoposide induced p53 phosphorylation and H2AX foci formation in proliferating SH-SY5Y cells but failed to do so in differentiated SH-SY5Y ***,while inhibition of ATM in undifferentiated SH-SY5Y cells partially protected them from etoposide-induced apoptosis,the same treatment had no effect on cell viability in differentiated SH-SY5Y *** results suggest that DNA damage or defective response to DNA damage is not the cause of neuronal cell death in human *** contrast,we discovered that Akt phosphorylation was inhibited when ATM activity was suppressed in differentiated SH-SY5Y ***,inhibition of ATM induced apoptosis following serum starvation in neuronally differentiated SH-SY5Y cells but could not trigger apoptosis under the same conditions in undifferentiated proliferating SH-SY5Y *** results demonstrate that ATM mediates the Akt signaling and promotes cell survival in neuron-like human SH-SY5Y cells,suggesting that impaired activation of Akt is the reason for neuronal degeneration in human A-T.

Anthocyanin Accumulation Mediated by Blue Light and Cytokinin in Arabidopsis Seedlings

It has been reported that pigmentation In plants Is stimulated by light and cytoklnln （CTK）; however, the signaling pathways and the relationship between light and CTK Involved In the regulation of anthocyanln accumulation remain to be elucidated. We Investigated （i） the role of blue light （BL） and CTK In anthocyanln accumulation ; and （ii） the relationship between BL and CTK In wild type （WT） and by4 mutants of Arabidopsis thaiiana. Two-d-old seedlings grown on medium with or without klnetln （KT） or zeatln （ZT） In darkness were Irradiated using BL at different fluence rates for 3 d before the anthocyanln content was determined using a spectrophotometrlc method. Anthocyanln accumulation was strongly Induced by BL In WT seedlings but not In hy4 seedlings, which demonstrated that CRY1 Is the main photoreceptor for BL. Both KT and ZT enhanced the response of the WT seedlings to BL In a dose-dependent manner, whereas they were not sufficient to promote anthocyanln eccumulatlon In darkness. In addition, data from experiments using the hy4 mutant showed that the CTK effect of BL was also CRYl-dependent. The results from experiments with three different treatment programs showed that the relationship between BL and KT In anthocyanln accumulation of Arabidopsis seedlings seems neither muItlpllcatlve nor additive coactlon, but rather Interaction. BL Is necessary for anthocyanln accumulation, and KT might be Involved In the BL signaling pathway.

Directed energy deposition-arc(DED-Arc)technology has the advantages of simple equipment,low manufacturing cost and high deposition rate,while the use of DED-Arc has problems of microstructure inhomogeneity,position dependence of macroscopic mechanical properties and ***,it is necessary to carry out a subsequent heat treatment to improve its microstructure uniformity,mechanical properties and *** this investigation,the DED-Arc 15-layer NiTi alloy thin-walled parts with the solution treatment at different process parameters were studied to analyze the effects of solution heat treatment on microstructure,phase composition,phase transformation,microhardness,tensile and *** temperature range of solution treatment is 800-1050℃,and the treatment time range is 1-5.5 *** results show that after solution treatment at 800℃/1 h,the content of precipitated phase decreases,the grain is refined,the microhardness increases,and the mechanical properties in the 0°direction are *** strain recovery rate after 10 tensile cycles has increased from 37.13%(as-built)to 49.25%(solid solution treatment).This research provides an effective post treatment method for high-performance DED-Arc NiTi shape memory alloys.

The classical Archimedean approximation of π uses the semiperimeter or area of regular polygons inscribed in or circumscribed about a unit circle in R~2 and it is well-known that by using linear combinations of these basic estimates,modern extrapolation techniques can greatly speed up the approximation ***,when n vertices are randomly selected on the circle,the semiperimeter and area of the corresponding random inscribed and circumscribing polygons are known to converge to π almost surely as n→∞,and by further applying extrapolation processes,faster convergence rates can also be achieved through similar linear combinations of the semiperimeter and area of these random *** this paper,we further develop nonlinear extrapolation methods for approximating π through certain nonlinear functions of the semiperimeter and area of such *** focus on two types of extrapolation estimates of the forms χ_n=S_n~αA_n~β and Y_n(p)=(αS_n~p+βA_n~p)~(1/p) where α+β=1,p≠0,and Sn and An respectively represents the semiperimeter and area of a random n-gon inscribed in the unit circle in R~2,and Xn may be viewed as the limit of Y_n(p) when p→*** deriving probabilistic asymptotic expansions with carefully controlled error estimates for Xn and Y_n(p),we show that the choice α=4/3,β=-1/3 minimizes the approximation error in both cases,and their distributions are also asymptotically normal.

Pressure transient characteristics of non-uniform conductivity fractured wells in viscoelasticity polymer flooding based on oil-water two-phase flow

Polymer flooding in fractured wells has been extensively applied in oilfields to enhance oil *** contrast to water,polymer solution exhibits non-Newtonian and nonlinear behavior such as effects of shear thinning and shear thickening,polymer convection,diffusion,adsorption retention,inaccessible pore volume and reduced effective ***,the flux density and fracture conductivity along the hydraulic fracture are generally non-uniform due to the effects of pressure distribution,formation damage,and proppant *** this paper,we present an oil-water two-phase flow model that captures these complex non-Newtonian and nonlinear behavior,and non-uniform fracture characteristics in fractured polymer *** hydraulic fracture is firstly divided into two parts:high-conductivity fracture near the wellbore and low-conductivity fracture in the far-wellbore section.A hybrid grid system,including perpendicular bisection(PEBI)and Cartesian grid,is applied to discrete the partial differential flow equations,and the local grid refinement method is applied in the near-wellbore region to accurately calculate the pressure distribution and shear rate of polymer *** combination of polymer behavior characterizations and numerical flow simulations are applied,resulting in the calculation for the distribution of water saturation,polymer concentration and reservoir *** with the polymer flooding well with uniform fracture conductivity,this non-uniform fracture conductivity model exhibits the larger pressure difference,and the shorter bilinear flow period due to the decrease of fracture flow ability in the far-wellbore *** field case of the fall-off test demonstrates that the proposed method characterizes fracture characteristics more accurately,and yields fracture half-lengths that better match engineering reality,enabling a quantitative segmented characterization of the near-wellbore section with high fracture conductivity and th
