A genome scan of recent positive selection signatures in three sheep populations

Domesticated sheep have been exposed to artificial selection for the production of fiber, meat, and milk as well as to natural selection. Such selections are likely to have imposed distinctive selection signatures on the sheep genome. Therefore, detecting selection signatures across the genome may help elucidate mechanisms of selection and pinpoint candidate genes of interest for further investigation. Here, detection of selection signatures was conducted in three sheep breeds, Sunite （n=66）, German Mutton （n=159）, and Dorper （n=93）, using the Illumina OvineSNP50 Genotyping BeadChip array. Each animal provided genotype information for 43 273 autosomal single nucleotide polymorphisms （SNPs）. We adopted two complementary haplotype-based statistics of relative extended haplotype homozygosity （REHH） and the cross-popu- lation extended haplotype homozygosity （XP-EHH） tests. In total, 707,755, and 438 genomic regions subjected to positive selection were identified in Sunite, German Mutton, and Dorper sheep, respectively, and 42 of these regions were detected using both REHH and XP-EHH analyses. These genomic regions harbored many important genes, which were enriched in gene ontology terms involved in muscle development, growth, and fat metabolism. Fourteen of these genomic regions overlapped with those identified in our previous genome-wide association studies, further indicating that these genes under positive selection may underlie growth developmental traits. These findings contribute to the identification of candidate genes of interest and aid in understanding the evolutionary and biological mechanisms for controlling complex traits in Chinese and western sheep.

目的:优化芍药苷聚乳酸-羟基乙酸共聚物(PLGA)纳米粒制备工艺,并探讨芍药苷PLGA纳米粒对H2O2诱导损伤的H9c2心肌细胞的保护作用。方法:首先使用复乳-溶剂挥发法制备芍药苷PLGA纳米粒,采用Plackett-Burman设计实验以及Box-Behnken响应面设计实验对其制备工艺进行优选,得出最佳处方,并对按最佳处方制备的芍药苷PLGA纳米粒进行表征分析、4℃储藏稳定性考察以及体外释放考察;最后通过H2O2诱导建立大鼠H9c2心肌细胞氧化损伤模型,考察芍药苷PLGA纳米粒对心肌细胞的保护作用,CCK-8法检测细胞存活率,试剂盒检测心肌细胞乳酸脱氢酶(LDH)漏出量、丙二醛(MDA)、超氧化物歧化酶(SOD)的含量。结果:最佳处方:泊洛沙姆浓度为0.4%,给药量为3.1 mg,PLGA为21.4 mg;所得芍药苷PLGA纳米粒包封率为(45.49±0.29)%,载药量为(4.52±0.05)%,粒径为(115.1±3.61)nm,多分散系数(polydiseperse index,PDI)为(0.122±0.01),Zeta电位为-(23.2±0.19)mV,形态为规则圆形,体外释放12 h释放率为80%;细胞实验结果表明芍药苷PLGA纳米粒能提高心肌细胞存活率,降低LDH和MDA含量,升高SOD含量(P<0.05)。结论:优化所得芍药苷PLGA纳米粒制备工艺可行,所得纳米粒稳定性好,具有缓释作用,体外释放符合Ritger-Peppas动力学模型;芍药苷PLGA纳米粒对H2O2诱导损伤的H9c2心肌细胞具有明显的保护作用。

Genetic parameters for somatic cell score and production traits in the fi rst three lactations of Chinese Holstein cows

The objectives of this study were to estimate genetic parameters of lactation average somatic cell scores （LSCS） and examine genetic associations between LSCS and production traits in the first three lactations of Chinese Holstein cows using single-parity multi-trait animal model and multi-trait repeatability animal model. There were totally 273605 lactation records of Chinese Holstein cows with first calving from 2001 to 2012. Heritability estimates for LSCS ranged from 0.144 to 0.187. Genetic correlations between LSCS and 305 days milk, protein percentage and fat percentage were -0.079, -0.082 and -0.135, respectively. Phenotypic correlation between LSCS and 305 days milk yield was negative （-0.103 to -0.190）. Genetic correlation between 305 days milk and fat percentage or protein percentage was highly negative. Genetic correlation between milk fat percentage and milk protein percentage was highly favorable. Heritabilities of production traits decreased with increase of parity, whereas heritability of LSCS increased with increase of parity.

Molecular Cloning of IGFBP-1 Gene and Developmental Expression of Its mRNA in Different Tissues of Nanjiang Mongolian Gazelles

The objective of the present study was to investigate the developmental expression patterns of Insulin-like growth factor-binding protein 1 (IGFBP-1) gene in different tissues of postnatal Nanjiang Mongolian Gazelles. Samples of heart, liver, spleen, lung, longissimus dorsi, semimembranosus, m. triceps brachii and biceps muscle of thigh were collected from a total of 36 Nanjiang Mongolian Gazelles at the age of 0, 15, 30, 60, 90 and 120 days after birth (3 males and 3 females at each age). The CDS was sequenced and ontogeny of mRNA levels of IGFBP-1 were measured by real-time fluorescence quantitative RT-PCR. The size of IGFBP-1 ORF was 792 bp encoding 263 amino acid residues, and displayed higher nucleotide/amino acid sequence identities with other ruminants compared to non-ruminants. The levels of IGFBP-1 mRNA in liver were highest (P0.05). Three expression patterns of IGFBP-1 mRNA during postnatal growth from birth to day 60 were found: consistently decreasing (liver), fluctuating as increasing then decreasing (heart) or as decreasing then increasing then decreasing (spleen, lung and muscles). The results indicate that the IGFBP-1 gene is highly conserved among species, and liver has the highest expression. It was concluded that IGFBP-1 plays important roles in early postnatal growth and is expressed in a developmental-tissue-dependent manner.

Construction of SMART cDNA library of Sheep Ovary and Identification of Candidate Gene by Homologous Cloning

The cDNA library of an ovary from Small Tail Han sheep before estrus was constructed by switching mechanism at 5＇ end of RNA transcript （SMART） approach. This library had a plaque titer of 1 x 109 pfu mL-1 and a 96% recombinant ratio of which the fragment length of inserted average cDNA sequences was 1.0 kb. Based on bioinformatics analysis of the sequences, we obtained 338 expressed sequence tags （ESTs） from 380 cDNA clones which indicated 191 contigs. These contigs consist of 89 unmatched ESTs, 9 homologous known genes in sheep, and 93 homologous sequences in species of mouse, bovine, and human beings, including 19 sequences expressed in the ovary or follicle and 14 unknown sequences. Several candidate genes associated with sheep reproduction trait such as epidermal growth factor （EGF）, estrogen receptor （ESR）, Inhibin, follicle stimulating hormone receptor （FSHR）, prostaglandin （PG）, and transforming growth factor-β （TGF-β） were identified and the homologous were cloned from this library, which will contribute to compile expression profiles and find the major genes of prolificacy of Small Tail Han sheep.

A Bird’s Eye View of Quantitative Proteome of Tumor Tissues from Lung Cancer Patients by a High Precision Mass Spectrometry Method

Lung cancer produces a high incidence of malignant tumors.There have been many studies on lung cancer using mass spectrometry(MS)technologies.However,most studies have focused on humoral samples.In this work,26 pairs of tissue samples(tumor vs.para-tumor)from patients with lung cancer were analyzed using liquid chromatography-tandem MS(LC-MS/MS)with data-independent acquisition mode.In total,3152 proteins were quantified from tissue samples with high confidence,including 189 up-regulated and 522 down-regulated proteins(tumor vs.para-tumor).In addition,79 and 690 proteins were identified only in para-cancerous samples and cancerous samples,respectively.The results from bio-informatics tools indicated that altered proteins like PEBP1,HRG and LYZ could be ideal reservoirs for screening the potential biomarkers for lung cancer.It is believed these tissue-specific proteomics results will assist in the studies of lung cancer.

Process intensification for the synthesis and purification of battery-grade li_(2)CO_(3)with microfluidics

Battery-grade lithium carbonate(li_(2)CO_(3))with a purity of higher than 99.5 wt%is of great importance as a high value-added lithium salt.However,influences of different reaction systems and process control on product purity remain unclear.Herein,a membrane dispersion microreactor was used to enhance the mass transfer of preparation and purification processes in homogeneous and heterogeneous system.Synthetic systems of Na_(2)CO_(3)–liCl,NH_(4)HCO_(3)–liCl,and NH_(3)·H_(2)O−CO_(2)−liCl,CO_(2)purification based on carbonation and decomposition were adopted.The li_(2)CO_(3)purity was increased by the improvement of mixing performance.The carbonation time was reduced by 62.5%and 58.3%for the NH_(3)·H_(2)O−CO_(2)and CO_(2)purification systems,respectively.In the two ammonia-based systems,li_(2)CO_(3)particles with a purity of 99.7–99.8 wt%were one-step prepared with a size of 3–5μm,which also met the requirement of the battery-grade standard.The purity was further increased to 99.9 wt%by CO_(2)purification and liHCO_(3)decomposition.The investigation could provide a feasible alternative for the controllable preparation of battery-grade li_(2)CO_(3)in one or multiple steps.

Intraspecific Relationship of Sheep Based on Mitochondrial Cytochrome b Gene

[ Objective] To preliminarily explore the intraspecific relationship of sheep based on cytochrome b （ Cyt b） gene of mitochondrial DNA （mtDNA）. [Method] The Cyt b gene sequences in 112 sheep individuals of two local sheep breeds were amplified by PCR. Then the amplified products were digested with EcoR I and analyzed by restriction fragment length polymorphism （RFLP）. [ Result] As many as 56 samples from Tan sheep and 56 samples from Wadi sheep were detected. The results showed that the amplified Cyt b gene in 51 individuals of Tan sheep had one EcoR I restriction site and no EcoR I restriction site in other five individuals, thus the Cyt b gene in Tan sheep showed two restriction morphs; the Cyt b gene in all 56 individuals of Wadi sheep had one EcoR I restriction site and showed one restriction morph. [ Conclusion] The polymorphism of mitochondrial Cyt b gene in Tan sheep and Wadi sheep is poor, and the Cyt b gene in sheep breeds is very conservative. Therefore, using Cyt b gene as gene marker to study the intraspecific relationship of sheep has some limitations.

Data-driven Wasserstein distributionally robust chance-constrained optimization for crude oil scheduling under uncertainty

Crude oil scheduling optimization is an effective method to enhance the economic benefits of oil refining.But uncertainties,including uncertain demands of crude distillation units(Cdus),might make the production plans made by the traditional deterministic optimization models infeasible.A data-driven Wasserstein distributionally robust chance-constrained(WDRCC)optimization approach is proposed in this paper to deal with demand uncertainty in crude oil scheduling.First,a new deterministic crude oil scheduling optimization model is developed as the basis of this approach.The Wasserstein distance is then used to build ambiguity sets from historical data to describe the possible realizations of probability distributions of uncertain demands.A cross-validation method is advanced to choose suitable radii for these ambiguity sets.The deterministic model is reformulated as a WDRCC optimization model for crude oil scheduling to guarantee the demand constraints hold with a desired high probability even in the worst situation in ambiguity sets.The proposed WDRCC model is transferred into an equivalent conditional value-at-risk representation and further derived as a mixed-integer nonlinear programming counterpart.Industrial case studies from a real-world refinery are conducted to show the effectiveness of the proposed method.Out-of-sample tests demonstrate that the solution of the WDRCC model is more robust than those of the deterministic model and the chance-constrained model.

Early diagnosis and management of neonatal sepsis:a perspective

Neonatal sepsis is an illness caused by bloodstream bacteria or fungal infections that affect newborn infants under 28 days of age.It stands as a significant contributor to morbidity and mortality in neonatal intensive care units(NICUs).With the improved survival rates of very low-birth-weight or extremely premature infants,a growing number of these vulnerable infants are receiving intensive care and undergoing invasive procedures,making nosocomial infections a persistent challenge in NICUs.The mortality rate associated with neonatal sepsis varies depending on factors such as birth weight,gestational age,and other co-morbidities.It can also lead to a range of long-term health consequences,including neurodevelopmental disabilities like cerebral palsy,visual or hearing impairments,and cognitive problems.Furthermore,neonatal sepsis is linked to various adverse outcomes,including respiratory complications,bronchopulmonary dysplasia,nutritional and growth issues,and immunological dysfunction[1].due to its atypical clinical presentation and the low rate of specific diagnostic indicators,such as a positive blood culture,accurately diagnosing neonatal sepsis is challenging.Consequently,the appropriateness of antibiotic treatment is often a matter of controversy.
