Composition analysis of Compound Shenhua Tablet,a seven-herb Chinese medicine for IgA nephropathy:evaluation of analyte-capacity of the assays

Compound Shenhua Tablet,a medicine comprising seven herbs,is employed in treating IgA nephropathy.This study aimed to meticulously analyze its chemical composition.Based on a list of candidate compounds,identified through extensive literature review pertinent to the tablet’s herbal components,the composition analysis entailed the systematic identification,characterization,and quantification of the constituents.The analyte-capacity of LC/ESI-MS-based and GC/EI-MS-based assays was evaluated.The identified and characterized constituents were quantified to determine their content levels and were ranked based on the constituents’daily doses.A total of 283 constituents,classified into 12 distinct categories,were identified and characterized in the Compound Shenhua Tablet.These constituents exhibited content levels of 1−10982μg·g^(−1),with daily doses of 0.01−395μmol·d^(−1).The predominant constituents,with daily doses of≥10μmol·d^(−1),include nine organic acids(citric acid,quinic acid,chlorogenic acid,cryptochlorogenic acid,gallic acid,neochlorogenic acid,isochlorogenic acid C,isochlorogenic acid B,and linoleic acid),five iridoids(specnuezhenide,nuezhenoside G13,nuezhenidic acid,secoxyloganin,and secologanoside),two monoterpene glycosides(paeoniflorin and albiflorin),a sesquiterpenoid(curzerenone),a triterpenoid(oleanolic acid),and a phenylethanoid(salidroside).Additionally,there were 83,126,and 55 constituents detected in the medicine with daily doses of 1–10,0.1–1,and 0.01–0.1μmol·d^(−1),respectively.The combination of the LC/ESI-MS-based and GC/EI-MS-based assays demonstrated a complementary relationship in their analyte-capacity for detecting the constituents present in the medicine.This comprehensive composition analysis establishes a solid foundation for further pharmacological research on Compound Shenhua Tablet and facilitates the quality evaluation of this complex herbal medicine.

Weak interaction-steered evolution of polyoxovanadate-based metal-organic polyhedra from transformation via interlock to packing

A bottleneck in biomimetic synthesis consists in the full copy of,for example,the hierarchical structure of proteins directed by weak interactions.By contrast with covalent bonds bearing definite orientation and high stability,weak intermolecular forces within a continuous dynamic equilibrium can be hardly tamed for molecular design.In this endeavor,a ligand-dominated strategy that embodies tunable electrostatic repulsion andπ···πstacking was first employed to shape polyoxovanadate-based metal-organic polyhedra(VMOPs).Structural evolution involving transformation,interlock,and discovery of an unprecedented prototype of the Star of David was hence achievable.Not only as a handy tool for the primary structural control over VMOPs,these weak forces allow for an advanced management on the spatial distribution of such manmade macromolecules as well as the associated physicochemical behaviors,representing an ideal model for simulating and interpreting the conformation-function relationship of proteins.

Pancreatic cancer is among the most malignant cancers,and thus early intervention is the key to better survival outcomes.However,no methods have been derived that can reliably identify early precursors of development into malignancy.Therefore,it is urgent to discover early molecular changes during pancreatic tumorigenesis.As aberrant glycosylation is closely associated with cancer progression,numerous efforts have been made to mine glycosylation changes as biomarkers for diagnosis;however,detailed glycoproteomic information,especially site-specific N-glycosylation changes in pancreatic cancer with and without drug treatment,needs to be further explored.Herein,we used comprehensive solid-phase chemoenzymatic glycoproteomics to analyze glycans,glycosites,and intact glycopeptides in pancreatic cancer cells and patient sera.The profiling of N-glycans in cancer cells revealed an increase in the secreted glycoproteins from the primary tumor of MIA PaCa-2 cells,whereas human sera,which contain many secreted glycoproteins,had significant changes of glycans at their specific glycosites.These results indicated the potential role for tumor-specific glycosylation as disease biomarkers.We also found that AMG-510,a small molecule inhibitor against Kirsten rat sarcoma viral oncogene homolog(KRAS)G12C mutation,profoundly reduced the glycosylation level in MIA PaCa-2 cells,suggesting that KRAS plays a role in the cellular glycosylation process,and thus glycosylation inhibition contributes to the anti-tumor effect of AMG-510.

Optical vortex arrays,with their unique wavefront structures,find extensive applications in fields such as optical communications,trapping,imaging,metrology,and quantum.The methods used to generate these vortex beam arrays are crucial for their applications.In this review,we begin with introducing the fundamental concepts of optical vortex beams.Subsequently,we present three methods for generating them,including diffractive optical elements,metasurfaces,and integrated optical devices.We then explore the applications of optical vortex beam arrays in five different domains.Finally,we conclude with a summary and outlook for the research on optical vortex beam arrays.

乡村居家治疗与照护中的疾病不仅是科学的存在,也是宇宙论意义的存在以及在世的存在。就科学的存在而言,疾病是自然界的物质与病者的身体相遇而互构出来的一种界面,现代医学和传统医学在这个界面上实施干预。就宇宙论意义的存在而言,疾病是宇宙神秘力量的显象,需要借助秘术和民俗医疗手段进行救治。就在世的存在而言,疾病创建了以病人为核心的医疗社会,该类医疗社会是各种医学知识观念显象、具身与交流的场域,即科学的存在和宇宙论的存在在其中存在。然而,按照西方医学科学模式开展救治和护理的城镇医院则把上述诸多内容屏蔽在外。

Isolation, Identification, and Quantitative Determination of Saponin in Apostichopus japonicus by HPLC-DAD

Apostichopus japonicus is an important invertebrate that is widely used as a tonic food in Asian countries.The purpose of this study is to purify and identify a class of compound,the saponins,from the body wall of A.japonicus,and to establish a new me-thod to determine the quantity of saponins in the sea cucumber.In this study,the saponins of A.japonicus,cladoloside A(CA),were ob-tained from 80%ethanol extract by column chromatography for the first time and were characterized using the spectral method.The resulting purified saponins were then profiled using ^(1)HNMR,^(13)CNMR,and ESI-MS,which revealed the CA molecular formula of C_(53)H_(82)O_(2) and contained a triterpenoid backbone,a methylglucopyranosyl moiety,a quinovopyranosyl,and two xylopyranosyls.A me-thod for the quantitative determination of CA,comprising microwave-assisted extraction,high-performance liquid chromatography,and diode array detector method,was established.Extraction efficiency was optimized by changing microwave power,extraction sol-vent,volume,time,and temperature.Results showed that under the optimum conditions(extraction time of 10 min,temperature of 45℃,and solvent of 25 mL 70%ethanol under 400 W),the detection limit of CA was 0.0015 mg mL^(−1),and the recoveries of CA from samples at spiking levels of 10,20,and 50μg g^(−1) ranged from 90.1%-104%.The proposed method was successfully applied to ana-lyze the saponins in different tissues of A.japonicus collected in different seasons.The method developed in this study can provide quantitative technical support for the quality control of A.japonicus.

核燃料包壳热导率是核燃料元件传热性能研究和核安全设计的重要参数,SiC复合包壳的双层结构和其内层的SiC/SiC复合材料特性增加了预测包壳导热性能的复杂性。为预测双层SiC复合包壳及内层复合材料热导率,基于Markworth有效介质理论,考虑孔隙率、温度、中子辐照等因素对SiC/SiC复合材料热导率的影响,在Markworth有效介质理论模型的基础上进行修正,计算了第二代、第三代SiC纤维增强的不同类型SiC/SiC复合材料热导率,并建立了双层SiC复合包壳热导率计算模型,初步预测了核电厂稳态运行期间双层SiC复合包壳的热导率。计算结果表明,核电厂稳态运行4个月后,辐照肿胀变形达到饱和,含第二代纤维的包壳热导率为3.9 W/(m·K),含第三代纤维的包壳热导率为4.8 W/(m·K)。

为缓解智能网联公交车出站强制换道时与社会车辆之间的冲突,分析了智能网联公交车与社会车辆行为决策的演化博弈机制。考虑社会车辆为人工驾驶社会车辆和智能网联社会车辆两种场景,从安全效用、时间效用和节能效用角度构建演化博弈效用函数,并以此建立演化博弈的复制动态方程。根据动态博弈系统的雅克比矩阵的行列式和迹,分析不同情况下博弈的均衡点及其稳定性,并探讨动态博弈系统对合作策略与非合作策略差值的灵敏度。结果表明,动态博弈系统可能的演化方向包括“智能网联公交车换道、社会车辆让路”“,智能网联公交车不换道、社会车辆不让路”和“智能网联公交车不换道、社会车辆让路”。从整体最优的角度看,调整社会车辆安全效用和时间效用的相关变量,能够保证更多车辆在智能网联公交车出站强制换道过程中的安全和效率。节能效用对(换道,让路)策略组合有促进作用,对(不换道,不让路)策略组合有抑制作用。

基层治理体系作为国家制度执行能力和社会基本运作秩序的集中体现，已然随着不确定性时代的来临而涌现出多重风险。现阶段，中国城市基层社区治理的困境主要是围绕以街道为代表的国家行政力量与以居委会为代表的社会自治力量在基层场域的潜在矛盾甚至冲突展开的。凝聚力不足、资源整合受阻、注意力调配失调，共同阻碍着当前城市基层社区治理的深入开展。街道与居委会之间设立的“片区”则通过占据街居之间的结构洞位置成为优化街居关系、纾解基层行政与自治张力的重要接点。尽管片区治理的模式目前还存在一些问题，但借助于结构洞的理论视角分析，我们在经验调查中发现，以“片区”为核心的治理实践不仅形成了一套规范有序的行动逻辑，而且为城市基层治理的合作共治提供了一种新思路。
