Catechins have been proven to exert antitumor effects in different kinds of cancers.However,the underlying mechanisms have not been completely clarified yet.This study aimed to assess the effects and mechanisms of(-)-epigallocatechin-3-gallate(EGCG)and(-)-epicatechin-3-gallate(ECG)on human melanoma skin A375 cells.Results showed that EGCG and ECG inhibited the proliferation of A375 cells and ECG showed better inhibitory effect.Flow cytometry analysis had shown that EGCG and ECG induced apoptosis and led to cell cycle arrest.EGCG and ECG decreased Bcl-2 expression and upregulated Caspase-3 protein level,indicating the development of apoptosis.Furthermore,EGCG and ECG could decreased mitochondrial membrane potential of A375 cells.In addition,the expression of Beclin-1,LC3 and Sirt3 were downregulated at protein levels,which known to be associated with autophagy.After autophagy was increased by rapamycin,the apoptotic trend was not change,indicating that apoptosis and autophagy are independent.Mechanistically,EGCG and ECG treatments decreased phosphorylated-AMPK(p-AMPK)and increased the ratios of P-PI3K,p-AKT and p-mTOR in melanoma cells.Conclusively,EGCG and ECG induced apoptosis via mitochondrial signaling pathway,downregulated autophagy through modulating the AMPK/mTOR and PI3K/AKT/mTOR signaling pathway.It indicated that EGCG and ECG may be utilized in human melanoma treatment.

Recently,continuous interests and persisting efforts on heteropolyniobate have been dedicated to its preparation and exploration.In this work,a novel Se-centred heteropolyniobate,[H2SeNb12(VO)4O40][Cu(en)2]2{[VNb12(VO)4O40][Cu(en)2]4}0.5·39H2O(en=ethanediamine),was synthesized.The compound was synthesized in aqueous solution by reacting the K7HNb6O19·13H2O with a mixture of vanadate(Ⅴ) and selenite(Ⅳ) source,which consists of two polyanions,i.e.,V-centred one and Se-centred another.The architecture was characterized by infrared spectra,thermogravimetric analysis,X-ray powder diffraction spectroscopy,ultraviolet-visible and X-ray photoelectron spectroscopy.Further,the magnetic property shows the antiferromagnetic interaction of the compound.

Study on tungsten sintering temperature field based on electromagnetic simulation and computational fluid dynamics

In order to study the influence of various sintering variables more intuitively and clearly,electromagnetic field analysis and computational fl uid dynamics are used to analyze the infl uence of gas flow,heating power and other process parameters on the temperature range of medium-frequency sintering in this paper.The results indicate that the uniformity of temperature fi eld is determined by the gas flow rate and heating power.When the heating power is 50 kW,the stable temperature drops about 10 K for every 1 m^(3)·h^(−1)increase of hydrogen flow.The peak value of the maximum temperature diff erence on the tungsten rods gradually increases with the increase of the hydrogen flow rate,and it appears slightly later.The temperature distribution of tungsten rods in furnace is similar with diff erent hydrogen flow rate.At the same time,the closer to the furnace wall,the higher the temperature of the tungsten rod.The temperature diff erence between tungsten rods mainly occurred within 1 h after the beginning of heating.Increasing the heating power can improve the stable temperature,but has little eff ect on the heating time.Increasing heating power can increase the peak value of the maximum temperature diff erence on the tungsten rods,but has little eff ect on the occurrence time of the peak value of the maximum temperature diff erence on the tungsten rods.Increasing the heating power will increase the temperature diff erence of a single tungsten rod,but the temperature diff erence of a single tungsten rod never exceeds 65 K.This study can be used to guide the process optimization design.Combined with numerical simulation calculation,the sintering parameters with low energy consumption and high temperature uniformity can be quickly and eff ectively found,thus improving the microstructure and properties of sintered tungsten products.

Objective This study aimed to analyze the temporal trends and characteristics associated with waist circumference(WC)among elderly Chinese people.Methods We used data from 3,096 adults≥65 years who participated in the China Health and Nutrition Survey(CHNS),an ongoing cohort study,between 1993 and 2015.We used longitudinal quantile regression models to explore the temporal trends and characteristics associated with WC.Results WC increased gradually among the elderly Chinese population during the survey.The WC curves shifted to the right with wider distributions and lower peaks in men and women.All WC percentile curves shifted upward with similar growth rates in the 25th,50th,and 75th percentiles.The WC means increased from 78 cm to 86 cm during the 22 years of our study.WC significantly increased with age and body mass index and decreased with physical activity(PA).These associations were stronger in the higher percentiles than in the lower percentiles.Conclusions WC is rising among Chinese adults≥65 years.Factors affecting WC in elderly people may have different effects on different percentiles of the WC distribution,and PA was the most important protective factor in the higher percentiles of the WC distribution.Thus,different interventional strategies are needed.

P-selectin engagement of P-selectin glycoprotein Iigand-1 （PSGL-1） causes circulating leukocytes to roll on and adhere to the vascular surface, and mediates intracellular calcium flux, a key but unclear event for subsequent arresting firmly at and migrating into the infection or injured tissue. Using a parallel plate flow chamber technique and intracellular calcium ion detector （Fluo-4 AM）, the intracellular calcium flux of firmly adhered neutrophils on immobilized P-selectin in the absence of chemokines at various wall shear stresses was investigated here in real time by fluorescence microscopy. The results demon- strated that P-selectin engagement of PSGL-1 induced the intracellular calcium flux of firmly adhered neutrophils in flow, increasing P-selectin concentration enhanced cellu- lar calcium signaling, and, force triggered, enhanced and quickened the cytoplasmic calcium bursting of neu- trophils on immobilized P-selectin. This P-selectin-induced calcium signaling should come from intracellular calcium release rather than extracellular calcium influx, and be along the mechano-chemical signal pathway involving the cytoskeleton, moesin and Spleen tyrosine kinase （Syk）. These results provide a novel insight into the mechano-chemical regulation mechanism for P-selectininduced calcium signaling of neutrophils in flow.

Quantitative Evaluation Method of Shape Indicator for Cold-rolled Strip

Quantitative level evaluation of cold-rolled strip was significant for the shape control and the product structure optimization. Based on the probability statistics method and the shape pattern recognition method, the shape evaluation model of cold-rolled strip was established to evaluate real-time shape of the online strip. The model was embedded in the shape control system, and the online strip shape state was real-time observed whether to meet the technical indicators, so the shape level could be identified and evaluated. Based on the shape evaluation indicators, the shape component could be well controlled to improve the shape hit rate and the control stability, so that the rolled strip shape could meet the technical requirements. At a 1050 cold strip mill, the shape data of the scene typical specifications strip were online collected and real-time evaluated, and the hit of macro shape in less than 8 I was 68.44%, indicating that the mill shape control had considerable room for improvement, as well as the evaluation method was feasible and practical for quantitative evaluation of the strip shape. So the new evaluation model has important guiding significance to increase value-added and reduce product quality objection.

教育游戏中引入摄像头交互方式,在增加游戏趣味性的同时还能促进玩家的身体运动参与。从现有研究来看,摄像头交互式游戏的设计还缺乏较为完善的设计框架,其教学应用的有效性有待验证。论文针对这两个问题设计了一种基于开放机器视觉库和Flash前端显示的分层式游戏设计框架。该框架借助机器视觉库完成手势识别,并根据交互规则实现交互。论文以儿童英语单词学习游戏为原型系统验证了该框架的可行性,同时利用幼儿园中随机选出的被试者从认知能力和情感态度两方面验证了系统的有效性。

Application of confocal laser Raman spectroscopy on marine sediment microplastics

Marine sediment is the primary sink of microplastics and is an indicator of pollution levels.However,although there are well-developed detection methods,detection is rarely focused on lowmicrometer-sized particles,mainly due to technique limitations.In this study,a simplifi ed process omitting digestion procedures was developed to pretreat microplastics obtained from marine sediment and was coupled with micro-Raman spectroscopy to identify microplastics.Based on the overall analysis of the characteristic peak assignments,a Raman spectral reference library was constructed for 18 types of plastic.In addition,the eff ects of the measurement parameters were systematically described.Field research was then conducted to validate the developed process and investigate microplastic contamination in Huiquan Bay,Qingdao,China.This simplifi ed process could retain the original appearance of microparticles and accomplish the detection of<500μm-sized microplastics in environmental samples.Microplastics in the size range of 10-150μm accounted for 76%of all microplastics,and 56%of the total particles was particles smaller than 50μm.Polypropylene(42%)and polyethylene(20%)were predominant components of the particles.In particular,polypropylene particles smaller than 10μm were identifi ed in marine sediment.This work demonstrates that Raman spectroscopy is not only an eff ective tool for detecting environmental particles but also highly applicable for identifying particles extracted from marine sediment.
