Construction of human Fab library and screening of a single-domain antibody of amyloid-beta 42 oligomers

Screening humanized antibodies from a human Fab phage display library is an effective and quick method to obtain beta-amyloid oligomers. Thus, the present study prepared amyloid-beta 42 oli- gomers and constructed a have human Fab phage display library based on blood samples from six healthy people. After three rounds of biopanning in vitro, a human single-domain antibody that spe- cifically recognized amyloid-beta 42 oligomers was identified. Western blot and enzyme-linked im- munosorbent assay demonstrated this antibody bound specifically to human amyloid-beta 42 tetramer and nonamer, but not the monomer or high molecular weight oligomers. This study suc- cessfully constructed a human phage display library and screened a single-domain antibody that specifically recognized amyloid-beta 42 oligomers.

Polymeric semiconductors spin-coated onto the photoresist grating form discontinuously distributed flocci,which do not fill grating grooves with nanoscale ***,the polymer layer neither forms a continuous gain channel nor creates a waveguide that enhances the distributed feedback(DFB)mechanism provided by the grating *** is verified by the microscopic and spectroscopic investigations on the topographic surface of the polymer-covered grating as it is etched layer by layer using oxygen *** gives more insights into the mechanisms involved in optically pumped polymer lasers and provides new guidance for constructing DFB lasers.

The time resolution of a radiography system for high-Z materials shall be at least 1 ns, hence the need of a time measurement system with a resolution of about 100 ps. In this paper, a Time Measurement Evaluation Board (TMEB) is developed to meet the need. It is based on the time-to-digital converter of ACAM TDC-GP2. Test results show an overall time resolution of 81 ps in detecting cosmic-rays with a plastic scintillator.

VME system of the Resistive Plate Chamber (RPC) electronics for the Daya Bay Reactor Neutrino Experiment is described in this paper. A 9U VME RPC trigger module (RTM) is designed to process coincidence signals coming from front end cards (FECs), to generate local triggers and send them to FECs to select the hit data from RPC detector, to report trigger information to a master trigger system and receive cross triggers from the master trigger system. Another 9U VME readout module is designed to collect data from all FECs, to send out configurations to FECs, and to transmit collected hit data to the data acquisition system via VME bus. Test results prove that the VME system is capable of treating a maximum data rate (2.2 MB·s-1 ), without data loss.

Synthesizing and editing dynamic flames with color temperature

Generating computer animated flames is a difficult and computationally expensive problem. Dynamic textures provide an effective means for extrapolating and synthesizing dynamic flames, but aggravates color distortion due to the high correlation of RGB components. A novel method for dynamic flame texture synthesis using color temperature is proposed in this paper. Firstly, the colortemperature mapping is calculated by using the Planck's law and two-color pyrometric technique to avoid color distortion. Secondly, a novel dynamic texture model is presented to transform the RGB space into temperature space. Finally,the dynamic flames editing is presented to support physical temperature adjustment. Experimental results illustrate that our approach is effective to synthesize visually plausible dynamic flames without color distortions and to edit dynamic flames with intuitive physical interpretation.

OBJECTIVES To establish a scoring system combining the ACEF score and the quantitative blood flow ratio(QFR) to improve the long-term risk prediction of patients undergoing percutaneous coronary intervention(PCI).METHODS In this population-based cohort study, a total of 46 features, including patient clinical and coronary lesion characteristics, were assessed for analysis through machine learning models. The ACEF-QFR scoring system was developed using 1263consecutive cases of CAD patients after PCI in PANDA Ⅲ trial database. The newly developed score was then validated on the other remaining 542 patients in the *** In both the Random Forest Model and the Deep Surv Model, age, renal function(creatinine), cardiac function(LVEF)and post-PCI coronary physiological index(QFR) were identified and confirmed to be significant predictive factors for 2-year adverse cardiac events. The ACEF-QFR score was constructed based on the developmental dataset and computed as age(years)/EF(%) + 1(if creatinine ≥ 2.0 mg/d L) + 1(if post-PCI QFR ≤ 0.92). The performance of the ACEF-QFR scoring system was preliminarily evaluated in the developmental dataset, and then further explored in the validation dataset. The ACEF-QFR score showed superior discrimination(C-statistic = 0.651;95% CI: 0.611-0.691, P < 0.05 versus post-PCI physiological index and other commonly used risk scores) and excellent calibration(Hosmer–Lemeshow χ^(2)= 7.070;P = 0.529) for predicting 2-year patient-oriented composite endpoint(POCE). Objective To investigate whether the very elderly patients with non-ST-segment elevation myocardial infarction (NSTEMI) will benefit from an invasive strategy versus a conservative strategy. Methods 190 consecutive patients aged 80 years or older with NSTEMI were included in the retrospective study from September 2014 to August 2017, of which 69 patients received conservative strategy and 121 patients received invasive strategy. The primary outcome was death. Multivariate Cox regression models were used to assess the statistical association between strategies and mortality. The survival probability was further analyzed. Results The primary outcome occurred in 17.4% patients in the invasive group and in 42.0% patients in the conservative group (P = 0.0002). The readmission rate in the invasive group (14.9%) was higher than that in the conservative group (7.2%). Creatinine level (OR = 1.01, 95% CI: 0.10–1.03, P = 0.05) and use of diuretic (OR = 3.65, 95% CI: 1.56–8.53, P = 0.003) were independent influential factors for invasive strategy. HRs for multivariate Cox regression models were 3.45 (95% CI: 1.77–6.75, P = 0.0003), 3.02 (95% CI: 1.52–6.01, P = 0.0017), 2.93 (95% CI: 1. 46–5.86, P = 0.0024) and 2.47 (95% CI: 1.20–5.07, P = 0.0137). Compared with the patients received invasive strategy, the conservative group had remarkably reduced survival probability with time since treatment (P < 0.001). Conclusions An invasive strategy is superior to a conservative strategy in reducing mortality of patients aged 80 years or older with NSTEMI. Our results suggest that an invasive strategy is more suitable for the very elderly patients with NSTEMI in China.

Objectives To evaluate the very long-term safety and effectiveness of drug-eluting stents （DES） compared to bare-metal stents （BMS） for patients with large coronary vessels. Methods From April 2004 to October 2006, 2407 consecutive patients undergoing de novo lesion percutaneous coronary intervention with reference vessel diameter greater than or equal to 3.5 mm at Fu Wai Hospital in Beijing, China, were prospectively enrolled into this study. We obtained 9-year clinical outcomes including death, myocardial infarction （MI）, thrombosis, target lesion revascularization （TLR）, target vessel revascularization （TVR）, and major adverse cardiac events （MACE, the composite of death, MI, and TVR）. We performed Cox＇s proportional-hazards models to assess relative risks of all the outcome measures after propensity match. Results After propensity scoring, 514 DES-treated patients were matched to 514 BMS-treated patients. The patients treated with BMS were associated with higher risk ofTLR （HR： 2.55, 95%CI： 1.520-4.277, P = 0.0004） and TVR （HR： 1.889, 95%CI： 1.185-3.011, P = 0.0075）, but the rates of death/MI and MACE were not statistically different. All Academic Research Consortium definition stent thrombosis at 9-year were comparable in the two groups. Conclusions During long-term follow-up through nine years, use of DES in patients with large coronary arteries was still associated with significant reductions in the risks of TLR and TVR.
