Objective Air pollution is a leading public health *** study investigated the effect of air quality and pollutants on pulmonary function and inflammation in patients with asthma in *** The study monitored 27 asthma outpatients for a year,collecting data on weather,patient self-management[daily asthma diary,peak expiratory flow(PEF)monitoring,medication usage],spirometry and serum *** explore the potential mechanisms of any effects,asthmatic mice induced by ovalbumin(OVA)were exposed to PM_(2.5).Results Statistical and correlational analyses revealed that air pollutants have both acute and chronic effects on *** exposure showed a correlation between PEF and levels of ozone(O_(3))and nitrogen dioxide(NO_(2)).Chronic exposure indicated that interleukin-5(IL-5)and interleukin-13(IL-13)levels correlated with PM_(2.5)and PM_(10)*** asthmatic mouse models,exposure to PM_(2.5)increased cytokine levels and worsened lung ***,PM_(2.5)exposure inhibited cell proliferation by blocking the NF-κB and ERK phosphorylation *** Ambient air pollutants exacerbate asthma by worsening lung function and enhancing Th2-mediated ***,PM_(2.5)significantly contributes to these adverse *** research is needed to elucidate the mechanisms by which PM_(2.5)impacts asthma.

氧化修饰低密度脂蛋白促进人单核细胞源树突状细胞的成熟及活化

目的　探讨氧化修饰低密度脂蛋白 (oxidized lowdensitylipoprotein ,oxLDL)和低密度脂蛋白 (lowdensi tylipoprotein ,LDL)对人单核细胞源的树突状细胞 (monocytederiveddendriticcells,MDCs)免疫功能的影响。方法　采用免疫磁珠法分离人外周血CD14 + 单核细胞 ,经含rhGM CSF(10 0ng/mL)和rhIL - 4 (2 0ng/mL)的Cell gro培养 ,使其分化为MDCs。分别以PBS和LPS(5 0 0 μg/mL)作阴性和阳性 (成熟 )对照 ,观察经5 0 μg/mL的oxLDL和LDL干预 4 8h后 ,流式细胞术检测MDCs表型 (CD1a、CD4 0、CD86、HLA DR) ,混合T淋巴细胞反应检测MDCs对淋巴细胞增殖的影响 ,FITC Dextran检测MDCs吞噬功能 ,ELISA检测细胞培养上清细胞因子 (IL -12、IL - 10和TNF α)浓度。结果　与PBS相比 ,5 0 μg/mLoxLDL干预的MDCs,不但可明显上调共刺激分子CD86的高表达 ,而且反映MDCs成熟程度的CD1a也表达增强 ,与此同时经oxLDL处理的MDCs吞噬作用却明显减弱 ,进一步的混合T淋巴细胞反应显示经过oxLDL刺激的MDCs促T细胞增殖作用明显增强 (P <0 .0 5 ) ;此外 ,oxLDL可明显促进MDC分泌细胞因子TNF α、IL - 10和IL - 12 (P <0 .0 5 ) ,但明显弱于LPS组 ,而LDL无此作用。结论　oxLDL在促进DCs成熟的同时。

Effectiveness of Histopathological Examination of Ultrasound-guided Puncture Biopsy Samples for Diagnosis of Extrapulmonary Tuberculosis

Objective To evaluate the diagnostic value of histopathological examination of ultrasound-guided puncture biopsy samples in extrapulmonary tuberculosis(EPTB).Methods This study was conducted at the shanghai Public Health Clinical Center.A total of 115patients underwent ultrasound-guided puncture biopsy,followed by MGIT 960 culture(culture),smear,Gene Xpert MTB/RIF(Xpert),and histopathological *** assays were performed to evaluate their effectiveness in diagnosing EPTB in comparison to two different diagnostic criteria:liquid culture and composite reference standard(CRS).Results When CRS was used as the reference standard,the sensitivity and specificity of culture,smear,Xpert,and histopathological examination were(44.83%,89.29%),(51.72%,89.29%),(70.11%,96.43%),and(85.06%,82.14%),*** on liquid culture tests,the sensitivity and specificity of smear,Xpert,and pathological examination were(66.67%,72.60%),(83.33%,63.01%),and(92.86%,45.21%),*** examination showed the highest sensitivity but lowest ***,we found that the combination of Xpert and histopathological examination showed a sensitivity of 90.80%and a specificity of 89.29%.Conclusion Ultrasound-guided puncture sampling is safe and effective for the diagnosis of *** with culture,smear,and Xpert,histopathological examination showed higher sensitivity but lower *** combination of histopathology with Xpert showed the best performance characteristics.

基于低频电场变化探测阵列三维闪电定位系统和S波段双偏振天气雷达数据, 对海南岛2021年6月24日一次雹暴过程的闪电活动和云内动力与微物理特征进行了详细的分析.结果显示: 该雹暴闪电活动以云闪为主, 占总闪的85.38%.降雹前发生的地闪以正地闪为主, 在降雹前25 min内, 正地闪频数占总地闪频数的60%, 高于一般雷暴正地闪活动比例.在雹暴成熟阶段, 上升气流出现两次增强, 而闪电频数的增加均滞后于上升气流增强的时段, 同时, 闪电频数的增加也可能与对流单体的合并有关.结合模糊逻辑判断法对云内降水粒子的识别, 计算了总闪与冰相粒子的相关系数, 其中霰和冰晶粒子与总闪相关性较高, 相关系数分别为0.76和0.83.根据闪电辐射源的空间分布特征, 推测该雹暴降雹前为反三极性电荷结构, 正电荷区位于约7~9.5 km(~-10 ℃至-25 ℃).

The growth in biomedical data resources has raised potential privacy concerns and risks of genetic information leakage. For instance, exome sequencing aids clinical decisions by comparing data through web services, but it requires significant trust between users and providers. To alleviate privacy concerns, the most commonly used strategy is to anonymize sensitive data. Unfortunately, studies have shown that anonymization is insufficient to protect against reidentification attacks. Recently, privacy-preserving technologies have been applied to preserve application utility while protecting the privacy of biomedical data. We present the PICOTEES framework, a privacy-preserving online service of phenotype exploration for genetic-diagnostic variants (https://***:3000/). PICOTEES enables privacy-preserving queries of the phenotype spectrum for a single variant by utilizing trusted execution environment technology, which can protect the privacy of the user's query information, backend models, and data, as well as the final results. We demonstrate the utility and performance of PICOTEES by exploring a bioinformatics dataset. The dataset is from a cohort containing 20,909 genetic testing patients with 3,152,508 variants from the Children's Hospital of fudan university in china. dominated by the Chinese Han population (>99.9%). Our query results yield a large number of unreported diagnostic variants and previously reported pathogenicity.
