Ginsenoside Rg_(1)Reduces Cardiotoxicity While Increases Cardiotonic Effect of Aconitine in vitro

Objective To explore the synergic mechanism of ginsenoside Rg_(1)(Rg_(1))and aconitine(AC)by acting on normal neonatal rat cardiomyocytes(NRCMs)and pentobarbital sodium(PS)-induced damaged NRCMs.Methods The toxic,non-toxic,and effective doses of AC and the most suitable compatibility concentration of Rg_(1) for both normal and damaged NRCMs exposed for 1 h were filtered out by 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-diphenytetrazoliumromide,respectively.Then,normal NRCMs or impaired NRCMs were treated with chosen concentrations of AC alone or in combination with Rg_(1) for 1 h,and the cellular activity,cellular ultrastructure,apoptosis,leakage of acid phosphatase(ACP)and lactate dehydrogenase(LDH),intracellular sodium ions[Na^(+)],potassium ions[K^(+)]and calcium ions[Ca^(2+)]levels,and Nav1.5,Kv4.2,and RyR_(2) genes expressions in each group were examined.Results For normal NRCMs,3000µmol/L AC significantly inhibited cell viability(P<0.01),promoted cell apoptosis,and damaged cell structures(P<0.05),while other doses of AC lower than 3000µmol/L and the combinations of AC and Rg_(1) had little toxicity on NRCMs.Compared with AC acting on NRCMs alone,the co-treatment of 3000 and 10µmol/L AC with 1µmol/L Rg_(1) significantly decreased the level of intracellular Ca^(2+)(P<0.01 or P<0.05),and the co-treatment of 3000µmol/L AC with 1µmol/L Rg_(1) significantly decreased the level of intracellular Ca^(2+)via regulating Nav1.5,RyR_(2) expression(P<0.01).For damaged NRCMs,1500µmol/L AC aggravated cell damage(P<0.01),and 0.1 and 0.001µmol/L AC showed moderate protective effect.Compared with AC used alone,the co-treatment of Rg_(1) with AC reduced the cell damage,0.1µmol/L AC with 1µmol/L Rg_(1) significantly inhibited the level of intracellular Na+(P<0.05),1500µmol/L AC with 1µmol/L Rg_(1) significantly inhibited the level of intracellular K+(P<0.01)via regulating Nav1.5,Kv4.2,RyR_(2) expressions in impaired NRCMs.Conclusion Rg_(1) inhibited the cardiotoxicity and enhanced the cardiotonic effect of AC via regulating the ion c

土壤氮输入形式显著影响土壤CO_(2)排放.生物炭负载氨氮作为一种新型氮输入形式,既降低了农田化学氮肥投入,又减少了环境治理成本,其对推动国家化肥零增长、农田面源污染防治和实现国家“碳达峰”、“碳中和”目标意义重大.通过室内培养试验,设置不施肥(CK)、单施化学氮肥(CF)、生物炭配施化学氮肥(BF)和生物炭负载氨氮(BN)这4种处理,研究了不同氮输入形式对土壤碳排放、酶活性及微生物群落的影响.结果表明,与CF相比,BF显著提高了累计碳排放量(66.24%),而BN未见显著性差异.值得注意的是,BN累计碳排放量较BF处理显著降低了35.28%.与CF和BF相比,BN处理β-葡萄糖苷酶,过氧化物酶,多酚氧化酶活性显著提高了20.25%和5.20%,36.72%和36.19%,90.36%和61.36%.与CF相比,BF处理提高了微生物群落丰富度和群落多样性,BN处理降低了微生物群落丰富度.与BF相比,BN处理变形菌门相对丰度降低了11.16%,放线菌门和拟杆菌门相对丰度分别提高了8.12%和5.83%.对微生物群落结构影响最大的土壤因子是木糖苷酶活性,绿弯菌门相对丰度与纤维二糖水解酶活性极显著相关;芽单胞菌门相对丰度与β-葡萄糖苷酶活性极显著相关;变形菌门相对丰度与累计碳排放量呈极显著相关.综上所述,生物炭负载氨氮较生物炭配施化学氮肥显著减少了累计碳排放量,其减排效果更优.研究结果将有利于国家“双碳战略”落地、生物天然气产业健康发展、国家绿色种养循环农业体系构建和国家化肥零增长战略实现.

提出一种电动力耦合循环井修复技术,以铬酸盐为典型目标污染物,研究该技术对低渗透含水层中地下水污染的修复效果及去除规律.结果表明:电动力耦合循环井技术相比循环井技术,修复半径扩大为原来的1.25倍,修复盲区面积占比从35.56%降为0,污染物的去除率从76.01%提高到了90.56%,相较于电动力修复技术电动力能耗降低了3个数量级,污染物去除率更高,可提升2~3倍.该技术耦合电动力方法实现了低渗透含水层中带电离子组分的强化定向迁移,解决了循环井在低渗透含水层中传质效率低、修复范围小、存在水力循环盲区等问题,修复效果显著提升.

MicroR-9c-5p and novel-mir50 co-target Akt to regulate Lasioderma serricorne reproduction

High fecundity is a common characteristic of insect pests which increases the difficulty of population control.Serine/threonine kinase Akt is an indispensable component of the insulin signaling pathway.Silencing of LsAkt severely hinders reproduction in Lasioderma serricorne,a stored product insect pest.However,the post-transcriptional pathway of LsAkt in L.serricorne remains unknown.This study identified 2 binding sites of miR-9c-5p and novel-mir50 in the coding sequences of LsAkt.The expression profiles of 2 microRNAs(miRNAs)and LsAkt displayed an opposite pattern during the adult stages.Luciferase reporter assay showed that novel-mir50 and miR-9c-5p could downregulate the expression of LsAkt.Overexpression of miR-9c-5p and novel-mir50 by injection of mimics inhibited the expression of LsAkt and reduced oviposition,decreased egg hatchability,and blocked ovarian development.It also decreased the expression of genes involved in ovarian development(LsVg and LsVgR)and the nutritional signaling pathway(LsTOR,LsS6K,and Ls4EBP),and reduced the phosphorylation of Akt.Conversely,injection of miR-9c-5p and novel-mir50 inhibitors induced the expressions of LsAkt,LsVg,LsVgR,LsTOR,LsS6K,and Ls4EBP,enhanced Akt phosphorylation level,and accelerated ovarian development.Injection of bovine insulin downregulated the expression of miR-9c-5p and novel-mir50 and upregulated the LsAkt expression.It also rescued the reproductive development defects associated with miR-9c-5p/novel-mir50 overexpression,forming a positive regulatory loop of insulin signaling.These results indicate that miR-9c-5p/novel-mir50 regulates the female reproduction of L.serricorne by targeting Akt in response to insulin signaling.The data also demonstrate the effects of the insulin/miRNA/Akt regulatory axis in insect reproduction.

Background Cardiomyocyte transplantation for the therapy of myocardial ischaemia is being paid close attention. However, how the microenvironment controls the differentiation of transplanted bone marrow stromal cells （BMSCs） is unknown. Endothelin-1 （ET-1）, a cytokine, increases during myocardial infarction, but it is not known whether ET-1 is responsible for the fate of transplanted BMSCs. In the present study, we investigated the effects of ET-1 on differentiation and maturation of induced rabbit BMSCs, in vitro, to elucidate the cellular biological mechanisms.

Current understanding of ELF4 deficiency:a novel inborn error of immunity

Background ELF4 deficiency has been recently recognized as a novel disorder within the spectrum of inborn errors of immunity(IEIs),specifically categorized as a“disease of immune dysregulation.”Cases of this condition,reported by our team and others,are very limited worldwide.As such,our current knowledge of this new disease remains preliminary.This review aims to provide a brief overview of the clinical manifestations,pathogenesis,and treatment strategies for this novel IEI.Data sources A comprehensive review was conducted after an extensive literature search in the PubMed/Medline database and websites concerning transcriptional factor ELF4 and reports concerning patients with ELF4 deficiency.Our search strategy was“ELF4 OR ETS-related transcription factor Elf-4 OR EL4-like factor 4 OR myeloid Elf-1-like factor”as of the time of manuscript submission.Results The current signature manifestations of ELF4 deficiency disorder are recurrent and prolonged oral ulcer,abdominal pain,and diarrhea in pediatric males.In some cases,immunodeficiency and autoimmunity can also be prominent.Targeted Sanger sequencing or whole exome sequencing can be used to detect variation in ELF4 gene.Western blotting for ELF4 expression of the patient’s cells can confirm the pathogenic effect of the variant.To fully confirm the pathogenicity of the variant,further functional test is strongly advised.Glucocorticoid and biologics are the mainstream management of ELF4 deficiency disorder.Conclusions Pediatric males presenting with recurring ulcerations in digestive tract epithelium with or without recurrent fever should be suspected of DEX.When atypical presentations are prominent,variations in ELF4 gene should be carefully evaluated functionally due to the complex nature of ELF4 function.Experience of treating DEX includes use of glucocorticoid and biologics and more precise treatment needs more patients to identify and further mechanistic study.

Esophageal cancer ranks among the most prevalent malignant tumors globally,primarily due to its highly aggressive nature and poor survival rates.According to the 2020 global cancer statistics,there were approximately 604000 new cases of esophageal cancer,resulting in 544000 deaths.The 5-year survival rate hovers around a mere 15%-25%.Notably,distinct variations exist in the risk factors associated with the two primary histological types,influencing their worldwide incidence and distribution.Squamous cell carcinoma displays a high incidence in specific regions,such as certain areas in China,where it meets the cost-effect-iveness criteria for widespread endoscopy-based early diagnosis within the local population.Conversely,adenocarcinoma(EAC)represents the most common histological subtype of esophageal cancer in Europe and the United States.The role of early diagnosis in cases of EAC originating from Barrett's esophagus(BE)remains a subject of controversy.The effectiveness of early detection for EAC,particularly those arising from BE,continues to be a debated topic.The variations in how early-stage esophageal carcinoma is treated in different regions are largely due to the differing rates of early-stage cancer diagnoses.In areas with higher incidences,such as China and Japan,early diagnosis is more common,which has led to the advancement of endoscopic methods as definitive treatments.These techniques have demonstrated remarkable efficacy with minimal complications while preserving esophageal functionality.Early screening,prompt diagnosis,and timely treatment are key strategies that can significantly lower both the occurrence and death rates associated with esophageal cancer.

Research on Bast Fiber Extracted from the White Bark of Three Species in the Genus Broussonetia

Against the backdrop of a global paper resource shortage,there is a growing need to identify fast-growing tree species capable of producing long-lasting paper.Three plant species namely Broussonetia kazinoki,Broussonetia papyrifera and hybrid paper mulberry,belong to the Broussonetia genus,were collected from China to study their white bark suitability for pulp and papermaking.Their chemical composition revealed that the holocellulose content in Broussonetia kazinoki and Broussonetia papyrifera was more than 80%.The molecular weight distribution of several holocellulose/α-cellulose is observed by GPC,which allows us to better observe the changes of various components on the molecular weight.The yield,lignin,whiteness,and molecular weight of the pulps obtained by NaOH treatment were determined.Optical microscope was used to characterize the fiber length-width ratio and rigidity.Finally,the improvement of the fiber rigidity method based on the Kratky-Porod chain model makes it more theoretical and further reveals the influencing factors of fiber rigidity.This study demonstrates the high potentiality of these three species for papermaking applications.

Prevalence of hypertension and its associations with body composition across Chinese and American children and adolescents

Background The age of onset of hypertension(HTN)is decreasing,and obesity is a significant risk factor.The prevalence and racial disparities in pediatric HTN and the association between body composition and blood pressure are insufficiently studied.This study aimed to evaluate the prevalence of HTN in Chinese and American children and adolescents and to assess the relationship between various body composition indices and HTN.Methods Seven thousand,five hundred and seventy-three Chinese and 6239 American children and adolescents aged 8–18 years from the 2013–2015 China Child and Adolescent Cardiovascular Health study and the 2011–2018 National Health and Nutrition Examination Surveys were analyzed.Blood pressure and body composition(fat and muscle)were measured by trained staff.The crude prevalence and age-standardized prevalence rate(ASPR)of primary HTN and its subtypes[isolated systolic hypertension(ISH)and isolated diastolic hypertension(IDH)]were calculated based on 2017 American Academy of Pediatrics guidelines.Multivariable-adjusted linear regression coefficients and odds ratios(ORs)were calculated to assess the associations of body composition indicators with HTN,ISH and IDH.Results The ASPR of HTN was 18.5%in China(CN)and 4.6%in the United States(US),whereas the obesity prevalence was 7.4%and 18.6%,and the population attributable risk of HTN caused by overweight and obesity was higher in the US than in CN.Increased fat mass,muscle mass and body fat percentage mass were associated with a higher risk of HTN in both countries.The percent of muscle body mass had a protective effect on HTN and ISH in both countries[HTN(CN:OR=0.83,95%CI=0.78–0.88;US:OR=0.72,95%CI=0.64–0.81);ISH(CN:OR=0.87,95%CI=0.80–0.94;US:OR=0.71,95%CI=0.62–0.81)],and the protective effect was more common among children and adolescents with high levels of physical activity.Conclusions The burden of HTN in Chinese children and adolescents was substantial and much greater than that in the US,and the contribution of obesity to HTN was higher in t

Hypertension-Induced Cerebral Small Vessel Disease Leading to Cognitive Impairment

Objective： Alzheimer＇s disease and vascular dementia are responsible for more than 80% of dementia cases. These two conditions share common risk factors including hypertension. Cerebral small vessel disease （CSVD） is strongly associated with both hypertension and cognitive impairment. In this review, we identify the pathophysiological changes in CSVD that are caused by hypertension and further explore the relationship between CSVD and cognitive impairment. Data Sources： We searched and scanned the PubMed database for recently published literatures up to December 2017. We used the keywords of＂hypertension＂, ＂cerebral small vessel disease＂, ＂＇white matter lesions＂, ＂enlarged perivascular spaces＂, ＂lacunar infarcts＂, ＂cerebral microbleeds＂, and ＂cognitive impairment＂ in the database of PubMed. Study Selection： Articles were obtained and reviewed to analyze the hypertension-induced pathophysiological changes that occur in CSVD and the correlation between CSVD and cognitive impairment. Results： In recent years, studies have demonstrated that hypertension-related changes （e.g., small vascular lesions, inflarnmator3, reactions, hypoperfusion, oxidative stress, damage to autoregulatory processes and the blood-brain barrier, and cerebral amyloid angiopathy） can occur over time in cerebral small vessels, potentially leading to lower cognitive function when blood pressure （BP） control is poor or lacking. Both isolated and co-occurrent CSVD can lead to cognitive deterioration, and this effect may be attributable to a dysfunction in either the cholinergic system or the functionality of cortical and subcortical tracts. Conclusions： We explore the currently available evidence about the hypertensive vasculopathy and inflammatory changes that occur in CSVD. Both are vital prognostic indicators of the development of cognitive impairment. Future studies should be performed to validate the relationship between BP levels and CSVD progression and between the nunabers, volumes, and anatomical locations of CSVD and cognitive impairment.
