Mangoes often suffer from low temperature-induced chilling injury (CI) during postharvest cold storage.Therefore,advanced techniques are crucial and in high demand to solve the chilling stress of mango fruit for a higher value.This study addresses chilling stress modulation by investigating the effects of melatonin treatment on CI,proline metabolism,and related gene expressions of ‘Keitt’ mango during cold storage after dipped in 0 (control),0.1 (MT1),and 0.2 mmol L^(-1)(MT2) melatonin solution for 30 min.The results revealed that melatonin treatment in MT1 significantly reduced CI development and increased proline content in mango fruit during cold storage compared to the control.These changes were along with increases in the activity of critical enzymes as well as the expression of encoding genes involved in proline biosynthesis,such as pyrroline-5-carboxylate synthetase (P5CS),pyrroline-5-carboxylate reductase (P5CR),ornithine D-aminotransferase (OAT),P5CS2,P5CR2,and OAT3.Additionally,proline dehydrogenase (PDH) activity and the expression of the PDH3 gene associated with proline dehydrogenation were lower in MT1-treated mangoes than the controlled group.Thus,melatonin treatment has regulated proline metabolism resulting in the accumulation of proline,subsequently contributing to enhancing the chilling tolerance of‘Keitt’mango fruit.

DEAR EDITOR,Spermatogenesis is the process by which male gametes are formed from spermatogonial stem cells(SSCs)and it is essential for the reliable transmission of genetic information between generations.After birth,prospermatogonia(ProSG)give rise to SSCs.While mouse spermatogenesis is relatively well studied,we are only just beginning to unravel this process in larger animals.Here,we analyzed key developmental transitions and differentiation trajectories by profiling neonatal,juvenile,and adult testes through single-cell sequencing(scRNA-seq).We found that SSCs were established at 30 days old,and that CDH1 was a novel cell surface marker for porcine ProSG and undifferentiated spermatogonia.

Background:The incidence of liver injury caused by anti-tuberculous(TB)drugs is very high.However,owing to a lack of sufficient evidence,preventive use of hepatoprotective drugs is not yet recommended.Therefore,we aimed to assess the protective effect of hepatoprotective drugs for anti-TB drug-induced liver injury.Methods:We conducted a literature search in China Biology Medicine disc,China National Knowledge Infras-tructure,WanFang,Chinese Scientific and Technological Journal,PubMed,Cochrane Library,Web of Science,and Embase.We performed meta-analysis using R 4.0 and Review Manager 5.3 software.Results:A total of 18 studies involving 3589 patients from 2 groups were included.Use of hepatoprotective drugs contributed to a lower incidence of liver injury as compared with conventional anti-TB treatment alone(relative risk[RR]=0.39,95%confidence interval[CI]:0.28-0.53,p<0.001).In subgroup analysis,significant protective effects were noted for mild liver injury(RR=0.30,95%CI 0.15-0.58),moderate(or severe)liver injury(RR=0.35,95%CI 0.19-0.65),and liver injury within 2-4 weeks(RR=0.37,95%CI 0.19-0.71).We also found a statistically significant difference in the incidence of drug withdrawal(RR=0.58,95%CI 0.34-0.97,p=0.040).Conclusions:Our results demonstrate that hepatoprotective drugs are effective in preventing liver injury in pa-tients receiving anti-TB treatment,to some extent.

The booming development of rare earth industry and the extensive utilization of its products accompanied by urban development have led to the accelerated accumulation of rare earth elements(REEs)as emerging pollutants in atmospheric environment.In this study,the variation of REEs in PM_(2.5)with urban(a non-mining city)transformation was investigated through five consecutive years of sample collection.The compositional variability and provenance contribution of REEs in PM_(2.5)were characterized,and the REEs exposure risks of children and adults via inhalation,ingestion and dermal absorption were also evaluated.The results showed an increase in the total REEs concentration from 46.46±35.16 mg/kg(2017)to 81.22±38.98 mg/kg(2021)over the five-year period,with Ce and La making the largest contribution.The actual increment of industrial and traffic emission source among the three pollution sources was 1.34 ng/m^(3).Coal combustion source displayed a downward trend.Ingestion was the main exposure pathway for REEs in PM_(2.5)for both children and adults.Ce contributed the most to the total intake of REEs in PM_(2.5)among the population,followed by La and Nd.The exposure risks of REEs in PM_(2.5)in the region were relatively low,but the trend of change was of great concern.It was strongly recommended to strengthen the concern about traffic-related non-exhaust emissions of particulate matter.

Background Formation of protein complexes across synapses is a critical process in neurodevelopment,having direct implications on brain function and animal behavior.Here,we present the understanding,importance,and potential impact of a newly found regulator of such a key interaction.Data sources A systematic search of the literature was conducted on PubMed(Medline),Embase,and Central-Cochrane Database.Results Membrane-associated mucin domain-containing glycosylphosphatidylinositol anchor proteins(MDGAs)were recently discovered to regulate synaptic development and transmission via suppression of neurexins-neuroligins transsynaptic complex formation.MDGAs also regulate axonal migration and outgrowth.In the context of their physiological role,we begin to consider the potential links to the etiology of certain neurodevelopmental disorders.We present the gene expression and protein structure of MDGAs and discuss recent progress in our understanding of the neurobiological role of MDGAs to explore its potential as a therapeutic target.Conclusion MDGAs play a key role in neuron migration,axon guidance and synapse development,as well as in regulating brain excitation and inhibition balance.

Objective:To investigate the mechanistic basis for the attenuation of bone degeneration by edible bird's nest(EBN)in ovariectomized rats.Methods:Forty-two female Sprage-Dawley rats were randomized into 7 groups(6 in each group).The ovariectomized(OVX)and OVX+6%,3%,and 1.5%EBN and OVX+estrogen groups were given standard rat chow alone,standard rat chow+6%,3%,and 1.5%EBN,or standard rat chow+estrogen therapy(0.2mg/kg per day),respectively.The sham-operation group was surgically opened without removing the ovaries.The control group did not have any surgical intervention.After 12 weeks of intervention,blood samples were taken for serum estrogen,osteocalcin,and osteoprotegerin,as well as the measurement of magnesium,calcium abd zinc concentrations.While femurs were removed from the surrounding muscles to measure bone mass density using the X-ray edge detection technique,then collected for histology and estrogen receptor(ER)immunohistochemistry.Results:Ovariectomy altered serum estrogen levels resulting in increased food intake and weight gain,while estrogen and EBN supplementation attenuated these changes.Ovariectomy also reduced bone ER expression and density,and the production of osteopcalcin and osteorotegerin,which are important pro-osteoplastic hormones that promote bone mineraliztion and density.Conversely,estrogen and EBN increased serum estrogen levels leading to increased bone ER expression,pro-osteoplastic hormone production and bone density(all P<0.05).Conclusion:EBN could be used as a safe alternative to hormone replacement therapys for managing menopausal complications like bone degeneration.

BACKGROUND Fascia iliaca compartment block is a technique that blocks three nerves,similar to a 3-in-1 nerve block.This block provides analgesia for patients undergoing lower limb surgery,and is a simple technique that is easy to implement.Here,we report a case of fascia iliaca compartment block in a patient with myocardial infarction who underwent emergency middle thigh amputation.CASE SUMMARY A 78-year-old female patient weighing 38 kg with gangrene and occlusive peripheral atherosclerosis of the right leg underwent an emergency middle thigh amputation.The patient had a history of hypertension,coronary heart disease,cerebral infarction,anterior wall myocardial infarction,and had recently undergone percutaneous coronary intervention consisting of coronary angiography and right coronary artery stent implantation.Considering the patient's condition,an ultrasound-guided fascia iliaca compartment block combined with general anesthesia was implemented for amputation.The fascia iliaca compartment block provided analgesia for the operation,and reduced the dosage of general anesthetics.It also alleviated adverse cardiovascular effects caused by pain stress,and ensured the safety of the patient during the perioperative period.This block also provided postoperative analgesia.The patient had a good prognosis,and was subsequently discharged from hospital.CONCLUSION Fascia iliaca compartment block provides surgical analgesia.It also alleviates adverse cardiovascular effects,and ensures patient safety during the perioperative period.

AIM: To evaluate the treatment selections and outcomes of keratoconus and discuss the grading treatment of keratoconus.METHODS: Medical records of 1162 patients(1863 eyes) with keratoconus treated with rigid gas permeable(RGP), corneal collagen crosslinking, and keratoplasty were reviewed. The patients were grouped according to the CLEK Study. The advanced group was further divided into a 60 D group. The best-corrected visual acuity(BCVA) and topographic data before and after treatment were recorded.RESULTS: In the 761 eyes with steep K60 D, surgical management accounted for 90.6%. A total of 618 eyes had improved BCVA at the final fol ow-up point(>18 mo, P52 D, the BCVA and topographic astigmatism outcomes showed no differences among the treatment groups. When steep K was >60 D, the BCVA in eyes treated with LKP was worse than those with steep K<60 D(P=0.025). The incidence of steep K progression in the RGP group was higher in advanced group(20.0% vs 10.8%, P=0.019). The probability of future keratoplasty in RGP was higher in advanced group(14.8% vs 7.0%, P=0.027). The incidence of steep K progression in the corneal collagen crosslinking(CXL) group was higher in advanced group(32.3% vs 8.5%, P=0.007). Multivariate logistic regression revealed the following related factors for treatment options: steep K [odds ratio(OR)=1.208, 95%CI: 1.052-1.387], TA(OR=1.171, 95%CI: 1.079-1.270), and TCT(OR=0.978, 95%CI: 0.971-0.984). The level of steep K, TA, and TCT all relates to the treatment choices of both keratoplasty and non-keratoplasty, while steep K provided the highest diagnostic accuracy(AUC=0.947, P<0.001).CONCLUSION: Steep K is an important grading treatment indicator. When steep K is <52 D, RGP lenses should be recommended. It is the best time for LKP when the steep K ranges

数字贸易是经济增长的新引擎,数据跨境流动则是国际数字贸易开展的新动能和加速器。作为全球首份数字经济区域协定,DEPA特设单独模块规定了数据跨境流动的规则。对接DEPA的高水平规则切合我国建设开放型经济新体制和对外贸易高质量发展的战略规划,也有助于完善我国数据治理体系和打造国际化数据市场宜商环境。在当前全面推进中国加入DEPA谈判的重要阶段,剖析我国数据跨境治理规则与DEPA及其所涉成员国之间的差异与差距,并针对我国当前面临的基础规则完备度不足、规则适用灵活度不够、数据流动自由度不高等问题,提出DEPA视域下我国数据跨境治理规则的应对方案。为顺利推进中国加入DEPA的进程,我国需要进一步提高数据要素的流动型开放水平,并在准确把握数据流动和安全发展平衡点的基础上,提升跨境合作水平,完善数据跨境合作运行机制与管理模式。

Comparison of long-term results after manual and femtosecond assisted corneal trephination in deep anterior lamellar keratoplasty for keratoconus

AIM: To compare long-term postoperative outcomes of manual and femtosecond assisted corneal trephination in deep anterior lamellar keratoplasty(FS-DALK) for keratoconus.METHODS: In the retrospective study, 17 consecutive eyes that underwent vertical side cut incision FS-DALK and 22 eyes that underwent trephine incision DALK were collected over a 2-year period. Main measurements included postoperative uncorrected-visual acuity(UCVA), corrected distance visual acuity(CDVA), refractive sphere and cylinder, manifest refraction spherical equivalent(MRSE), flat and steep corneal keratometry(K1 and K2), endothelial cell density(ECD), and time of epithelium healing and suture removal.RESULTS: Groups were comparable for diagnosis and preoperative visual acuity. Follow-up averaged 23 mo(range, 12-36 mo). At 12 mo, the mean UCVA was better in the manual-DALK group(P=0.039), and the refractive sphere was lower in the FS-DALK group(P=0.040). MRSE between groups differed at 1, 6, and 12 mo postoperatively(P=0.047, 0.025, 0.042, respectively). Mean CDVA, cylinder, K1, K2,corneal astigmatism, ECD, and time of epithelium healing were similar between groups. Stability of MRSE, ECD, and K1 returned sooner after FS-DALK. Initial loosened suture removal time was earlier in the manual-DALK group(P=0.042) while complete suture removal time was similar(P=0.122).CONCLUSION: Manual and femtosecond assisted corneal trephination in DALK are options for advanced keratoconus. FS-DALK do not result in improved visual acuity but it is more stable during the follow-up period. FSDALK in the present form show limited benefit, so surgical design and parameters still need to be optimized and explored.
