A novel long non-coding RNA, DIR, increases drought tolerance in cassava by modifying stress-related gene expression

Cassava is an important tropical cash crop. Severe drought stresses affect cassava productivity and quality, and cause great economic losses in agricultural production. Enhancing the drought tolerance of cassava can effectively improve its yield. Long non-coding RNAs(lncRNAs) are present in a wide variety of eukaryotes. Recently, increasing evidence has shown that lncRNAs play a critical role in the responses to abiotic stresses. However, the function of cassava lncRNAs in the drought response remains largely unknown. In this study, we identified a novel lncRNA, DROUGHT-INDUCED INTERGENIC lncRNA(DIR). Gene expression analysis showed that DIR was significantly induced by drought stress treatment, but did not respond to abscisic acid(ABA) or jasmonic acid(JA) treatments. In addition, overexpression of the DIR gene enhanced proline accumulation and drought tolerance in transgenic cassava. RNA-seq analysis revealed that DIR preferentially affected drought-related genes that were linked to transcription and metabolism. Moreover, RNA pull-down mass spectrometry analysis showed that DIR interacted with 325 proteins. A protein–protein interaction(PPI)analysis found a marked enrichment in proteins associated with the mRNA export and protein quality control pathways.Collectively, these results suggest that DIR and its interacting proteins that regulate mRNA or protein metabolism are involved in mediating the drought stress response. Thus, regulating DIR expression has potential for improving cassava yield under drought conditions.

Exceptional points enhance sensing in silicon micromechanical resonators

Exceptional points(EPs)have recently emerged as a new method for engineering the response of open physical systems,that is,systems that interact with the environment.The systems at the EPs exhibit a strong response to a small perturbation.Here,we show a method by which the sensitivity of silicon resonant sensors can be enhanced when operated at EPs.In our experiments,we use a pair of mechanically coupled silicon micromechanical resonators constituting a parity-time(PT)-symmetric dimer.Small perturbations introduced on the mechanically coupled spring cause the frequency to split from the EPs into the PT-symmetric regime without broadening the two spectrum linewidths,and this frequency splitting scales with the square root of the perturbation strength.The overall signal-tonoise ratio is still greatly enhanced,although the measured noise spectral density of the EP sensing scheme has a slight increase comparable to the traditional counterpart.Our results pave the way for resonant sensors with ultrahigh sensitivity.

Projections of summer light rain frequency in typical terrain over eastern China under wind speed constraint

The variation in near-surface wind speed is a key dynamic parameter in the orographic effect of precipitation over eastern China.In this study,we used the latest high-resolution outputs from six GCMs in CMIP6-HighResMIP to evaluate the performance of high-resolution models in simulating the orographic precipitation characteristics of typical mountainous areas in summer over eastern China.The orographic precipitation under warming scenarios was projected and constrained according to observational data.The results indicated that during the contemporary climate reference period(1979-2009),although the relationship between model-simulated near-surface wind speed and orographic light rain frequency was consistently stable,the sensitivity of the orographic light rain frequency to surface wind variability was generally underestimated,with a deviation approximately 24.1% lower than the observational values.The estimated orographic light rain frequency corrected based on the observed near-surface wind speed under a 1.5℃ warming scenario,was 36.1% lower than that of the contemporary period;this reduction was 8.6 times that without the wind speed constraint(4.2%).The MRI-AGCM3-2-S model,with a longer dataset,demonstrated relatively stable reductions in orographic light rain frequency under different warming scenarios(1.5℃,2℃,3℃,and 4℃)after the application of wind speed constraints.In all cases,the reductions exceeded those for the predictions made without the wind speed constraint.

The roles of RACK1 in the pathogenesis of Alzheimer's disease

The receptor for activated C kinase 1(RACK1)is a protein that plays a crucial role in various signaling pathways and is involved in the pathogenesis of Alzheimer's disease(AD),a prevalent neurodegenerative disease.RACK1 is highly expressed in neuronal cells of the central nervous system and regulates the pathogenesis of AD.Specifically,RACK1 is involved in regulation of the amyloid-β precursor protein processing through α-or β-secretase by binding to different protein kinase C isoforms.Additionally,RACK1 promotes synaptogenesis and synaptic plasticity by inhibiting N-methyl-D-aspartate receptors and activating gamma-aminobutyric acid A receptors,thereby preventing neuronal excitotoxicity.RACK1 also assembles inflammasomes that are involved in various neuroinflammatory pathways,such as nuclear factor-kappa B,tumor necrosis factor-alpha,and NOD-like receptor family pyrin domain-containing 3 pathways.The potential to design therapeutics that block amyloid-β accumulation and inflammation or precisely regulate synaptic plasticity represents an attractive therapeutic strategy,in which RACK1 is a potential target.In this review,we summarize the contribution of RACK1 to the pathogenesis of AD and its potential as a therapeutic target.

Dual metal ions/BNQDs boost PMS activation over copper tungstate photocatalyst for antibiotic removal:Intermediate,toxicity assessment and mechanism

In the metal-based peroxymonosulfate(PMS)activation process,the sluggish surface redox cycle of metal ions generally hampered the efficiency of PMS activation for pollutant removal.Herein,Codoped CuWO 4/BN quantum dots(CW/Co/BNQDs)photocatalysts were developed to realize Cu^(2+)/Cu+and Co^(2+)/Co^(3+)dual ions redox cycles for PMS activation,which would facilitate the tetracycline(TC)removal.CW/4Co/2BNQDs could degrade 94.8%TC within 30 min in PMS/Vis system,and the apparent rate constant of CW/4Co/2BNQDs was 2.7 times and 1.2 times higher than those of CW and CW/4Co,respectively.The improved TC degradation performance could be attributed to the synergetic effect between BNQDs and dual redox cycles.X-ray photoelectron spectroscopy(XPS)spectra of samples before and after the reaction demonstrated that BNQDs were beneficial for accelerating the Cu^(2+)/Cu+and Co^(2+)/Co^(3+)redox cycles in CW/4Co/2BNQDs,further boosting the activation of PMS in TC degradation.Experiments of different radical scavengers revealed that the SO_(4)^(·−)/·OH/h+/^(1)O_(2)reactive species participated in the PMS activation for the TC degradation process.The possible TC degradation pathway and intermediate toxicity were detailed investigated.In addition,CW/4Co/2BNQDs exhibited outstanding photocatalytic activity over five consecutive cycles,which illustrated that it was supposed to be a reliable PMS activator over antibiotic elimination for practical application.And this work shed new light on constructing dual redox cycles for efficient PMS activation.

Combining TDLAS and multi-fusion algorithms for methane gas concentration detection

High-precision methane gas detection is of great importance in industrial safety, energy production and environmental protection, etc. However, in the existing measurement techniques, the methane gas concentration information is susceptible to noise, which leads to its useful signal being drowned by noise. A fusion algorithm of variational modal decomposition(VMD) and improved wavelet threshold filtering is proposed, which is used in combination with tunable diode laser absorption spectroscopy(TDLAS) to implement a non-contact, high-resolution methane gas concentration detection. The fusion algorithm can perform noise reduction and further segmentation of the methane gas detection signal. And the simulation and experiment verify the effectiveness of the fusion algorithm, and the experimental results show that for the detection of air containing 10 ppm, 30 ppm, 60 ppm, 80 ppm, and 99 ppm methane, the errors are 12.75%, 8.18%, 3.37%, 2.46%, and 1.78%, respectively.

Visible light cross-linking and bioactive peptides loaded integrated hydrogel with sequential release to accelerate wound healing complicated by bacterial infection

The effective management of bacterial infections that are resistant to multiple drugs remains a substantial clinical challenge.The eradication of drug-resistant bacteria and subsequent promotion of angiogenesis are imperative for the regeneration of the infected wounds.Here,a novel and facile peptide containing injectable hydrogel with sustained antibacterial and angiogenic capabilities is developed.The antibacterial peptide that consists of 11 residues(CM11,WKLFKKILKVL)is loaded onto acrylate-modified gelatin through charge interactions.A vascular endothelial growth factor mimetic peptide KLT(KLTWQELYQLKYKGI)with a GCG(Gly-Cys-Gly)modification at the N-terminal is covalently coupled through a visible light-induced thiol-ene reaction.In this reaction,the acrylate gelatin undergoes cross-linkage within seconds.Based on the physical/chemical double crosslinking strategy,the bioactive peptides achieve sustained and sequential release.The results show that the hydrogel significantly inhibits methicillin-resistant Staphylococcus aureus(MRSA)growth through the rapid release of CM11 peptides at early stage;it forms obvious growth inhibition zones against pathogenic bacterial strains.Moreover,cell counting kit-8 assay and scratch test confirm that the CM11/KLT-functionalized hydrogels promote cell proliferation and migration through the later release of KLT peptides.In a mouse skin wound infected with self-luminous MRSA,the CM11/KLT-functionalized hydrogels enhance wound healing,with rapidly bacterial infection reduction,lower expression of inflammatory factors,and neovascularization promotion.These results suggest that the rationally designed,sustained and sequential release CM11/KLT-functionalized hydrogels have huge potential in promoting the healing of multi-drug resistant bacterial infected wounds.

Increased retinal venule diameter as a prognostic indicator for recurrent cerebrovascular events:a prospective observational study

Microvasculature of the retina is considered an alternative marker of cerebral vascular risk in healthy populations.However,the ability of retinal vasculature changes,specifically focusing on retinal vessel diameter,to predict the recurrence of cerebrovascular events in patients with ischemic stroke has not been determined comprehensively.While previous studies have shown a link between retinal vessel diameter and recurrent cerebrovascular events,they have not incorporated this information into a predictive model.Therefore,this study aimed to investigate the relationship between retinal vessel diameter and subsequent cerebrovascular events in patients with acute ischemic stroke.Additionally,we sought to establish a predictive model by combining retinal veessel diameter with traditional risk factors.We performed a prospective observational study of 141 patients with acute ischemic stroke who were admitted to the First Affiliated Hospital of Jinan University.All of these patients underwent digital retinal imaging within 72 hours of admission and were followed up for 3 years.We found that,after adjusting for related risk factors,patients with acute ischemic stroke with mean arteriolar diameter within 0.5-1.0 disc diameters of the disc margin(MAD_(0.5-1.0DD))of≥74.14μm and mean venular diameter within 0.5-1.0 disc diameters of the disc margin(MVD_(0.5-1.0DD))of≥83.91μm tended to experience recurrent cerebrovascular events.We established three multivariate Cox proportional hazard regression models:model 1 included traditional risk factors,model 2 added MAD_(0.5-1.0DD)to model 1,and model 3 added MVD0.5-1.0DD to model 1.Model 3 had the greatest potential to predict subsequent cerebrovascular events,followed by model 2,and finally model 1.These findings indicate that combining retinal venular or arteriolar diameter with traditional risk factors could improve the prediction of recurrent cerebrovascular events in patients with acute ischemic stroke,and that retinal imaging could be a useful and non-invasive method for ide

Light-sheet fluorescence microscopy(LSFM)has played an important role in bio-imaging due to its advantages of high photon efficiency,fast speed,and long-term imaging capabilities.The perpendicular layout between LSFM excitation and detection often limits the 3D resolutions as well as their isotropy.Here,we report on a reflective type light-sheet microscope with a mini-prism used as an optical path reflector.The conventional high NA objectives can be used both in excitation and detection with this design.Isotropic resolutions in 3D down to300 nm could be achieved without deconvolution.The proposed method also enables easy transform of a conventional fluorescence microscope to high performance light-sheet microscopy.

An approximate analytical model for unconventional reservoir considering variable matrix blocks and simultaneous matrix depletion

In regard to unconventional oil reservoirs,the transient dual-porosity and triple-porosity models have been adopted to describe the fluid flow in the complex fracture network.It has been proven to cause inaccurate production evaluations because of the absence of matrix-macrofracture communication.In addition,most of the existing models are solved analytically based on Laplace transform and numerical inversion.Hence,an approximate analytical solution is derived directly in real-time space considering variable matrix blocks and simultaneous matrix depletion.To simplify the derivation,the simultaneous matrix depletion is divided into two parts:one part feeding the macrofractures and the other part feeding the microfractures.Then,a series of partial differential equations(PDEs)describing the transient flow and boundary conditions are constructed and solved analytically by integration.Finally,a relationship between oil rate and production time in real-time space is obtained.The new model is verified against classical analytical models.When the microfracture system and matrix-macrofracture communication is neglected,the result of the new model agrees with those obtained with the dual-porosity and triple-porosity model,respectively.Certainly,the new model also has an excellent agreement with the numerical model.The model is then applied to two actual tight oil wells completed in western Canada sedimentary basin.After identifying the flow regime,the solution suitably matches the field production data,and the model parameters are determined.Through these output parameters,we can accurately forecast the production and even estimate the petrophysical properties.
