Multi-alleles predict primary non-response to infliximab therapy in Crohn’s disease

Background:Infliximab(IFX)is the first-line treatment for patients with Crohn’s disease(CD)and is noted for its relatively high *** therapeutic efficacy of IFX has noticeable individual *** single-gene polymorphisms(SNPs)are inadequate for predicting non-response to *** this study,we aimed to identify new genetic factors associated with IFX-therapy failure and to predict non-response to IFX by developing a multivariate predictive ***:In this retrospective study,we collected and analysed the data of Chinese patients with CD who received IFX therapy at one hospital between June 2013 and June *** non-response(PNR)and non-durable response(NDR)were evaluated using a simple endoscopic score for CD(SES-CD).A total of 125 SNPs within 44 genes were genotyped.A multivariate logistic-regression model was established to predict non-response to *** area-under-the-receiver-operatingcharacteristics curve(AUROC)was applied to evaluate the predictive model ***:Forty-two of 206(20.4%)patients experienced PNR and 15 of 159(9.4%)patients experienced *** SNPs were associated with PNR(P<0.05).A PNR predictive model was established,incorporating 2-week high-sensitivity C-reactive protein(hs-CRP),rs61886887,rs61740234,rs357291,rs2269330,and rs111504845,and the AUROC on training and testing data sets were 0.818(P<0.001)and 0.888(P<0.001),*** week 14,hs-CRP levels≥2.25 mg/L were significantly associated with NDR(AUROC=0.815,P<0.001).PNR-associated SNPs were not mutually associated with NDR,suggesting distinct mechanisms between PNR and ***:Genetic polymorphisms are significantly associated with response to IFX among Chinese CD patients.

Novel SNP of EPAS1 gene associated with higher hemoglobin concentration revealed the hypoxia adaptation of yak(Bos grunniens)

Endothelial PAS domain protein 1 gene （EPAS1） is a key transcription factor that activates the expression of oxygen-regu- lated genes. In this study, in order to better understand the effects of EPAS1 gene on hematologic parameters in yak, we firstly quantified the tissue expression patterns for EPASl mRNA of yak, identified polymorphism in this gene and evaluated its association with hematologic parameters. Expression of EPAS1 mRNA was detected in all eight tissues （heart, liver, lung, spleen, pancreas, kidney, muscles and ovary）. The expressions of EPAS1 in lung and pancreas were extremely higher than other tissues examined. Three novel single nucleotide polymorphisms （SNPs）（g.83052 C〉T, g.83065 G〉A and g.83067 C〉A） within the EPAS1 were identified and genotyped in Pali （PL）, Gannan （GN） and Tianzhu White （TZW） yak breeds. Significant higher frequencies of the AA and GA genotypes and A allele of the g.83065 G〉A were observed in the PL and GN breeds than that in the TZW breed （P〈0.01）. Association analysis of the PL breed indicated that the g.83065 G〉A polymorphism was significantly associated with hemoglobin （HGB） concentration in yaks （P〈0.05）. Individuals with genotype AA had significantly higher HGB concentration （P〈0.05） than those with genotype GA and GG. All these results will help our further understanding of biological functional of yak EPAS1 gene in responding to hypoxia and also indicate EPAS1 might contribute to the hypoxia adaptation of the yak.

Simulation and Experimentation for Low Density Drain AlGaN/GaN HEMT

In order to improve the breakdown voltage of AlGaN/GaN high electron mobility transistors （HEMTs）, we report a feasible method of low density drain （LDD） HEMT. The fluoride-based plasma treatment using CF4 gas is performed on the drain-side of the gate edge. The channel two-dimensional electron gas （2DEG） concentrations are modulated by fluoride plasma treatment, and the peak electric field at the gate edge is effectively reduced, so the breakdown voltage is improved. The electric field distributions of the LDD-HEMTs are simulated using the Silvaco software, and the peak of the electric field on the gate edge is effectively reduced. Experimental results show that, compared with the conventional HEMT, LDD-HEMTs have a lower reverse leakage current of the gate, and the breakdown voltage is increased by 36%. The current collapse characteristics of the LDD-HEMTs are confirmed by dual-pulse measurement, and an obvious pulse current reduction is due to the surface states by implanting F ions between the gate and the drain.

Implementing an“Impracticable”Copolymerization to Fabricate a Desired Polymer Precursor for N-doped Porous Carbons

It is common that a proof-of-concept of a desired reaction,which might generate materials with new functions or application potential,is eventually proved impracticable or commercially *** efforts have been made but wasted in searching for unknown reaction conditions in solvent environments because it was believed that the activity of reactants can be enhanced to facilitate reactions by dissolving them in ***,an abnormal case was discovered in this study.A desired copolymerization reaction between 1,3,5-tris(chloromethyl)-2,4,6-trimethylbenzene and melamine was confirmed to be impracticable under various solvent conditions;however,it was successfully implemented using a solvent-free *** first-principle calculations and molecular dynamics simulations,two decisive factors that the reaction in solvents cannot possess,namely the reaction equilibrium being pushed by the timely release of by-products and the confined thermal motions of the activated monomer molecules in the solid phase,were demonstrated to make the copolymerization successful in the solvent-free *** to the high aromaticity and azacyclo-content,the as-synthetic copolymer exhibited good application potential as a precursor to fabricate N-doped porous carbons with satisfactory carbon yields,ideal N contents,desired textural properties,and competitive CO_(2)capture abilities compared to other representative counterparts reported recently.
