Using a modified cDNA representational difference analysis(RDA)method,a LEA gene fragment was isolated from the regulated carrot somatic embryo,which was used as the probe to screen the cDNA library of the regulated carrot somatic embryo and the genomic library constructed by the method of altering osmotic *** analysis showed that it is homologous to LEA gene family and designated as DcLEA1(GenBank number:AF308739),a new member of the carrot LEA gene *** transcription re-gion contains5′UTR,two exons,one intron and3′UTR region;its coding region is480bp long,coding for159amino acids and one stop *** hybridization indicated that DcLEA1gene was not expressed in the adult carrot but expressed at high levels in the regulated carrot somatic *** carrot somatic embryo which had been deregulated for12.ours,the expression levels dropped rapid-ly;with the prolongation of deregulation,the radicle of carrot somatic embryo began to stretch,and the expression level of DcLEA1gene *** phenomenon is similar to the expression pattern of LEA gene in the course of dormancy and germination of the seed;thus suggesting that the sucrose regulation-deregulation system of the carrot somatic embryo can be used to mimic plant seed dormancy and ger-mination and can also be used to study the molecular mechanisms of these two biological processes.

The characteristics of drought in Xinjiang Uygur Autonomous Region(Xinjiang),china have changed due to changes in the spatiotemporal patterns of temperature and precipitation,however,the effects of temperature and precipitation—the two most important factors influencing drought—have not yet been thoroughly explored in this *** this study,we first calculated the standard precipitation evapotranspiration index(SPEI)in Xinjiang from 1980 to 2.2. based on the monthly precipitation and monthly average *** the spatiotemporal characteristics of temperature,precipitation,and drought in Xinjiang from 1980 to 2.2. were analyzed using the Theil-sen median trend analysis method and Mann-Kendall test.A series of SPEI-based scenario-setting experiments by combining the observed and detrended c{2.matic factors were uti{2.zed to quantify the effects of individual c{2.matic factor(i.e.,temperature and precipitation).The results revealed that both temperature and precipitation had experienced increasing trends at most meteorological stations in Xinjiang from 1980 to 2.2.,especially the spring temperature and winter *** to the influence of temperature,trends of intensifying drought have been observed at spring,summer,autumn,and annual *** addition,the drought trends in southern Xinjiang were more notable than those in northern *** 1980 to 2.2.,temperature trends exacerbated drought trends,but precipitation trends alleviated drought trends in *** meteorological stations in Xinjiang exhibited temperature-dominated drought trend except in winter;in winter,most stations exhibited precipitation-dominated wetting *** findings of this study high{2.ght the importance of the impact of temperature on drought in Xinjiang and deepen the understanding of the factors influencing drought.

Aging has a profound impact on the gingiva and significantly increases its susceptibility to periodontitis,a worldwide prevalent inflammatory ***,a systematic characterization and comprehensive understanding of the regulatory mechanism underlying gingival aging is still ***,we systematically dissected the phenotypic characteristics of gingiva during aging in primates and constructed the first single-nucleus transcriptomic landscape of gingival aging,by which a panel of cell type-specific signatures were *** cells were identified as the most affected cell types by aging in the *** analyses pinpointed the crucial role of YAP in epithelial self-renew and homeostasis,which declined during aging in epithelial cells,especially in basal *** decline of YAP activity during aging was confrmed in the human gingival tissues,and downregulation of YAP in human primary gingival keratinocytes recapitulated the major phenotypic defects observed in the aged primate gingiva while overexpression of YAP showed rejuvenation *** work provides an in-depth understanding of gingival aging and serves as a rich resource for developing novel strategies to combat aging-associated gingival diseases,with the ultimate goal of advancing periodontal health and promoting healthy aging.

As the co-chaperone of DnaK/Hsp70 protein, DnaJ/Hsp40 protein influences the synthesis and assembly of the protein complex by regulating ATPase activity of DnaK/Hsp70 protein. By employing the modified method of cDNA representational difference analysis, a homologous fragment of DnaJ was isolated from the deregulated carrot somatic embryos, and it was further used as the probe to screen the cDNA library of carrot somatic embryo deregulated for 12.h. As the result, DcJ1 gene, the homologous gene of DnaJ, was isolated from carrot. Sequence analysis showed that its coding region is 12.7 bp, which codes 418 amino acids and comprises 3 highly-conserved characteristic domains. Southern blot analysis suggested that the DcJ1 gene seems to be a single copy in the genome, while Northern blot result indicated that DcJ1 expresses only in roots and its degree of expression changes obviously with the regulation-deregulation process. BACKGROUND Research on gastrointestinal mucosal adenocarcinoma(GMA)is limited and controversial,and there is no reference tool for predicting postoperative *** To investigate the prognosis of GMA and develop predictive *** From the Surveillance,Epidemiology,and End Results database,we collected clinical information on patients with *** random sampling,the patients were divided into the discovery(70%of the total,for model training),validation(2.%,for model evaluation),and completely blind test cohorts(10%,for further model evaluation).The main assessment metric was the area under the receiver operating characteristic curve(AUC).All collected clinical features were used for Cox proportional hazard regression analysis to determine factors influencing GMA’s *** This model had an AUC of 0.7433[95% confidence intervals(95%CI):0.742.-0.7442.in the discovery cohort,0.72.4(GMA:0.72.4-0.72.4)in the validation cohort,and 0.7388(95%CI:0.7378-0.7398)in the test *** packaged it into Windows software for doctors’use and uploaded *** gastric adenocarcinoma had the worst prognosis,and these were protective factors of GMA:Regional nodes examined[hazard ratio(HR):0.98,95%CI:0.97-0.98,P1]and chemotherapy(HR:0.62.95%CI:0.58-0.66,P1).CONCLUSION The deep learning-based tool developed can accurately predict the overall survival of patients with GMA *** surgery,chemotherapy,and adequate lymph node dissection during surgery can improve patient outcomes.

Rechargeable batteries currently hold the largest share of the electrochemical energy storage market,and they play a major role in the sustainable energy transition and industrial decarbonization to respond to global climate *** to the increased popularity of consumer electronics and electric vehicles,lithium-ion batteries have quickly become the most successful rechargeable batteries in the past three decades,yet growing demands in diversified application scenarios call for new types of rechargeable *** efforts are made to developing the next-generation post-li-ion rechargeable batteries,which include,but are not limited to solid-state batteries,lithium–sulfur batteries,sodium-/potassium-ion batteries,organic batteries,magnesium-/zinc-ion batteries,aqueous batteries and flow *** the great achievements,challenges persist in precise understandings about the electrochemical reaction and charge transfer process,and optimal design of key materials and interfaces in a *** roadmap tends to provide an overview about the current research progress,key challenges and future prospects of various types of rechargeable *** computational methods for materials development,and characterization techniques will also be discussed as they play an important role in battery research.
