The precise mechanism of bone regeneration in different bone graft substitutes has been well studied in recent researches. However, miRNAs regulation of the bone formation has been always mysterious. We developed the anterior lumbar interbody fusion （AliF） model in pigs using equine bone protein extract （BPE）, recombinant human bone morphogenetic protein-2 （rhBMP-2） on an absorbable collagen sponge （ACS）, and autograft as bone graft substitute, respectively. The miRNA and gene expression profiles of different bone graft materials were examined using microarray technology and data analysis, including self-organizing maps, KEGG pathway and Biological process GO analyses. We then jointly analyzed miRNA and mRNA profiles of the bone fusion tissue at different time points respectively. Results showed that miRNAs, including let-7, miR-1.9, m iR-21. miR-1.3, miR-1.0, miR-1.6, miR-1.4, and miR-224, were involved in the regulation of the immune and inflammation response, which provided suitable inflammatory microenvironment for bone formation. At late stage, several miRNAs directly regulate SMAD4, Estrogen receptor 1.and 5-hydroxytryptamine （serotonin） receptor 2C for bone formation. It can be concluded that miRNAs play important roles in balancing the inflammation and bone formation.

Percent free prostatic-specific antigen （%fPSA） has been introduced as a tool to avoid unnecessary biopsies in patients with a serum PSA level of 4.0-1..0 ng ml^-1. however, it remains controversial whether %fPSA is effective in PSA range of 1..1.20.0 ng ml^-1.in both Chinese and Western population. In this study, the diagnostic performance of %fPSA and serum PSA in predicting prostate cancer （PCa） and high-grade PCa （HGPCa） was analyzed in a multi-center biopsy cohort of 591. consecutive Chinese patients who underwent prostate biopsy in 22 hospitals across china from January 1. 201. to December 31. 201.. The indication for biopsy was PSA〉4.0 ng ml^-1.or/and suspicious digital rectal examination. Total and free serum PSA determinations were performed by three types of electrochemiluminescence immunoassays with recalibration to the World Health Organization standards. The diagnostics accuracy of PSA, %fPSA and %fPSA in combination with PSA （%fPSA ＋ PSA） was determined by the area under the receivers operating characteristic curve （AUC）. %fPSA was more effective than PSA in men aged ≥60 years old. The AUC was 0.584 and 0.635 in men aged ≥60 years old with a PSA of 4.0-1..0 ng ml^-1.and 1..1.20.0 ng ml^-1. respectively. The AUC of %fPSA was superior to that of PSA in predicting HGPCa in patients ≥60 years old in these two PSA range. Our results indicated that %fPSA is both statistically effective and clinical applicable to predict prostate biopsy outcome in Chinese patients aged ≥60 years old with a PSA of 4.0-1..0 ng ml^-1.and 1..1.20.0 ng ml^-1.

Hypobaric hypoxia (HH) exposure can cause serious brain injury as well as life-threatening cerebral edema in severe cases. Previous studies on the mechanisms of HH-induced brain injury have been conducted primarily using non-primate animal models that are genetically distant to humans, thus hindering the development of disease treatment. Here, we report that cynomolgus mon{1.s (Macaca fascicularis) exposed to acute HH developed human-like HH syndrome involving severe brain injury and abnormal behavior. Transcriptome profiling of white blood cells and brain tissue from mon{1.s exposed to increasing altitude revealed the central role of the HIF-1.and other novel signaling pathways, such as the vitamin D receptor (VDR) signaling pathway, in co-regulating HH-induced inflammation processes. We also observed profound transcriptomic alterations in brains after exposure to acute HH, including the activation of angiogenesis and impairment of aerobic respiration and protein folding processes, which likely underlie the pathological effects of HH-induced brain injury. Administration of progesterone (PROG) and steroid neuroprotectant 5α-androst-3β,5,6β-triol (TRIOL) significantly attenuated brain injuries and rescued the transcriptomic changes induced by acute HH. Functional investigation of the affected genes suggested that these two neuroprotectants protect the brain by targeting different pathways, with PROG enhancing erythropoiesis and TRIOL suppressing glutamate-induced excitotoxicity. Thus, this study advances our understanding of the pathology induced by acute HH and provides potential compounds for the development of neuroprotectant drugs for therapeutic treatment.

We present the observational results from a detailed timing analysis of the black hole candidate EXO 1.46-031.during its outburst in 201. with the observations of Insight-HXMT,NICER and *** outburst can be classified roughly into four different ***-C quasi-periodic oscillations(QPOs)observed by NICER(about 0.1.6 Hz)and Insight-HXMT(about 0.7-8 Hz)are also reported in this ***,we study various physical quantities related to QPO *** QPO rms-frequency relationship in the energy band 1.1. keV indicates that there is a turning pointing in frequency around2 Hz,which is similar to that of GRS 1.1.+1.5.A possible hypothesis for the relationship above may be related to the inclination of the source,which may require a high inclination to explain *** relationships between QPO frequency and QPO rms,hardness,total fractional rms and count rate have also been found in other transient sources,which can indicate that the origin of type-C QPOs is non-thermal.

Jujube witches’broom(JWB)phytoplasmas parasitize the sieve tubes of diseased phloem and cause an excessive proliferation of axillary shoots from dormant lateral buds to favour their *** previous research,two JWB effectors,SJP1.and SJP2,were identified to induce lateral bud outgrowth by disrupting ZjBRC1.mediated auxin ***,the pathogenesis of JWB disease remains largely ***,tissue-specific transcriptional reprogramming was examined to gain insight into the genetic mechanisms acting inside jujube lateral buds under JWB phytoplasma *** phytoplasmas modulated a series of plant signalling networks involved in lateral bud development and defence,including auxin,abscisic acid(ABA),ethylene,jasmonic acid,and salicylic ***-induced bud outgrowth was accompanied by downregulation of ABA synthesis within lateral *** application rescued the bushy appearances of transgenic Arabidopsis overexpressing SJP1.and SJP2 in Col-0 and ZjBRC1.in the brc1.2 ***,the expression of ZjBRC1.and ABA-related genes ZjHB40 and ZjNCED3 was negatively correlated with lateral main bud outgrowth in decapitated healthy *** evidence showed that ZjBRC1.interacted with ZjBRC2 via its N-terminus to activate ZjHB40 and ZjNCED3 expression and ABA accumulation in transgenic jujube *** addition,ZjBRC1.widely regulated differentially expressed genes related to ABA homeostasis and ABA signalling,especially by binding to and suppressing ABA ***,these results suggest that JWB phytoplasmas hijack the ZjBRC1.mediated ABA pathways to stimulate lateral bud outgrowth and expansion,providing a strategy to engineer plants resistant to JWB phytoplasma disease and regulate woody plant architecture to promote crop yield and quality.

CFTR,a chloride channel and ion channel regulator studied mostly in epithelial cells,has been reported to participate in immune regulation and likely affect the risk of cancer ***,little is known about the effects of CFTR on the differentiation and function ofγδT *** this study,we observed that CFTR was functionally expressed on the cell surface ofγδT *** deletion and pharmacological inhibition of CFTR both increased IFN-γrelease by peripheralγδT cells and potentiated the cytolytic activity of these cells against tumor cells both in vitro and in ***,the molecular mechanisms underlying the regulation ofγδT cell IFN-γproduction by CFTR were either TCR dependent or related to Ca^(2+)*** was recruited to TCR immunological synapses and attenuated Lck-P38 MAPK-c-jun *** addition,CFTR was found to modulate TCR-induced Ca^(2+)influx and membrane potential(Vm)-induced Ca^(2+)influx and subsequently regulate the calcineurin-NFATc1.signaling pathway inγδT ***,CFTR serves as a negative regulator of IFN-γproduction inγδT cells and the function of these cells in antitumor *** investigation suggests that modification of the CFTR activity ofγδT cells may be a potential immunotherapeutic strategy for cancer.

Lepton scattering is an established ideal tool for studying inner structure of small particles such as nucleons as well as *** a future high energy nuclear physics project,an Electron-ion collider in china(EicC)has been *** will be constructed based on an upgraded heavy-ion accelerator,High Intensity heavy-ion Accelerator Facility(HIAF)which is currently under construction,together with a new electron *** proposed collider will provide highly polarized electrons(with a po-larization of 80%)and protons(with a polarization of 70%)with variable center of mass energies from 1. to 20 GeV and the luminosity of(2–3)×1.33 cm^(−2)·s^(−1..Polarized deuterons and Helium-3,as well as unpolarized ion beams from Carbon to Uranium,will be also available at the *** main foci of the EicC will be precision measurements of the structure of the nucleon in the sea quark region,including 3D tomography of nucleon;the partonic structure of nuclei and the parton interaction with the nuclear environment;the exotic states,especially those with heavy flavor quark *** addition,issues fundamental to understanding the origin of mass could be addressed by measurements of heavy quarkonia near-threshold production at the *** order to achieve the above-mentioned physics goals,a hermetical detector system will be constructed with cutting-edge *** document is the result of collective contributions and valuable inputs from experts across the *** EicC physics program complements the ongoing scientific programs at the Jefferson laboratory and the future EIC project in the United *** success of this project will also advance both nuclear and particle physics as well as accelerator and detector technology in china.
