Over-expression of VEGF165 in the adipose tissue-derived stem cells via the lentiviral vector

Background Many researchers studied the possibility of using stem cells as gene therapeutic vector. But few related reports on the adipose tissue-derived stem cells （ADSCs） are available. Therefore we intended to construct a lentiviral VEGF165 expression vector and then infect the ADSCs to produce therapeutic seed *** EHS1001-68950485313912 clone was mutated by PCR method to produce consensus fragment of VEGF165 transcript （NM_001025368）. Lentivirus was enveloped with pGC-FU, pHelper 1.0 and pHelper 2.0 plasmids in 293T *** then the ADSCs （multiplicity of infection=20） were transfected with the vectors after titer determination. Stable expression of VEGF165 in ADSCs was confirmed by immunofluorescence staining, enzyme-linked immunosorbent assay （ELISA） and Western blotting *** DNA sequencing and 293T transfection verified VEGF165 was linked to the GFP fused vector. The virus titer is up to 2x10a determined by quantitative PCR. VEGF165 transduced cells could show green fluorescence confirmed by immunofluorescence staining （almost 95%）. ELISA analyses could detect out the density of VEGF was 850.86-1202.13pg/ml （mean （923.00±31.22） pg/ml） in the supernatant of VEGF16s-transduced cells but not detected in the GFP-transduced cells （P 〈0.001） and the Western blotting analyses also confirmed VEGF165 expression in VEGF165-transduced *** The VEGF165 over-expression ADSCs were obtained and may be used as a cell therapeutic tool and may be applied for vascular regeneration, especially in the treatment of erectile dysfunction.

目的探讨基于CT三维重建技术获得的肾结石物理特征对软性输尿管镜碎石术(flexible ureteroscopic lithotripsy,FURL)碎石效能的影响。方法选取四川大学华西空港医院自2021年7月至2022年12月符合入组条件的86例肾结石患者。术前通过CT三维重建技术测定肾结石的各个物理特征,包括结石数量(个)、结石长径(mm)、结石密度(Hu)、结石体积(cm^(3))和结石质量(Hu*cm^(3))。入选患者均接受FURL治疗,FURL术后根据结石清除状态分为结石清除组和结石残留组,采用多因素二元logistic回归分析法分析肾结石患者FURL术后结石清除率的影响因素。绘制受试者工作特征(receiver operating characteristic,ROC)曲线,分析各结石特征预测肾结石患者FURL术后结石清除的价值。采用Spearman相关系数分析各结石特征与手术时间的相关性。结果结石清除组为67例,结石残留组为19例,结石清除率为77.9%。多因素分析结果显示,结石长径、结石密度、结石体积及结石质量均为影响FURL术后结石清除率的独立危险因素(P<0.05)。ROC曲线显示,结石长径、结石密度、结石体积及结石质量的特征因素曲线下面积(area under the curve,AUC)分别为0.891、0.674、0.919、0.957,结石质量预测结石清除率的效能明显优于其余结石特征因素。Spearman相关系数表明,结石长径、结石密度、结石体积、结石质量与手术时间均呈正相关(r=0.521、0.226、0.573、0.611,P<0.05),结石质量与手术时间相关性更高。结论基于CT三维重建技术获取的结石长径、密度、体积及质量等均是影响FURL术治疗肾结石的危险因素。其中,结石质量作为结石负荷指标可以更好地预测FURL术治疗肾结石的碎石效能。

A patient referred to our hospital, diagnosed with left idiopathic chronic orchialgia, was evaluated with a thorough medical and psychiatric history, physical examination, scrotal ultrasound and magnetic resonance imaging. Conservative management failed. The patient had temporary pain relief after undergoing outpatient cord block three times. Microsurgical denervation of the left spermatic cord was operated in March, 2011. A pain questionnaire was used to determine efficacy before and after operation, and complete pain relief was noted at one week after operation. The follow up period was 12 months, at the end of which the pain score was still zero. No complications, including testicular atrophy and hydrocele, occurred. Microsurgical denervation of the spermatic cord can be a minimally invasive, safe and effective management option for treatment of idiopathic chronic orchialgia.

In this paper, the insurance company considers venture capital and risk-free investment in a constant proportion. The surplus process is perturbed by diffusion. At first, the integro-differential equations satisfied by the expected discounted dividend payments and the Gerber-Shiu function are derived. Then, the approximate solutions of the integro-differential equations are obtained through the sinc method. Finally, the numerical examples are given when the claim sizes follow different distributions. Furthermore, the errors between the explicit solution and the numerical solution are discussed in a special case.

: Based on the current application requirements for wideband response photodetectors, , we designed a novel silicon avalanche photodetector (Si APD) ) with high response in a broad spectral range of 250-1100 nm and it could achieve efficient detection of ultraviolet, , visible and near-infrared light without splicing. The enhancement of silicon on ultraviolet and infrared bands was separately analyzed. This was followed by simulation on the device structure de- signs using different methods such as back incidence, , to improve short wavelength absorption while maintaining a high infrared absorption. The Si APD shows a peak wavelength at around 940 nm and a high photoresponse at 250 nm and 1 100 nm which exceeds 15% of the peak responsivity. This type of device is suitable for multispectral applications and future high-precision detection.

This paper studies the optimal portfolio allocation of a fund manager when he bases decisions on both the absolute level of terminal relative performance and the change value of terminal relative performance comparison to a predefined reference point. We find the optimal investment strategy by maximizing a weighted average utility of a concave utility and an Sshaped utility via a concavification technique and the martingale method. Numerical results are carried out to show the impact of the extent to which the manager pays attention to the change of relative performance related to the reference point on the optimal terminal relative performance.

A 2D axisymmetric numerical model was established to investigate the variations of molten pool with different melt rates during the vacuum arc remelting of 8Cr4Mo4V high-strength steel,and the ingot growth was simulated by dynamic mesh *** results show that as the ingot grows,the molten pool profile changes from shallow and flat to V-shaped,and both the molten pool depth and the mushy width ***,the variation of both the molten pool shape and the mushy width melt rate is clarified by the thermal equilibrium *** melt rate increases,both the molten pool depth and the mushy width *** is caused by the increment in sensible heat stored in the ingot due to the limitation of the cooling capacity of the *** nonlinear increment in sensible heat leads to a nonlinear increase in the mushy *** addition,as melt rate increases,the local solidification time(LST)of ingot decreases obviously at first and then *** melt rate is controlled in a suitable range,LST is the lowest and the secondary dendrite arm spacing of the ingot is the smallest,which can effectively improve the compactness degree of 8Cr4Mo4V high-strength steel.
