MKT-077 normalizes mitochondrial function and mitigates cardiac pathology in mdx mice

Objectives:Duchenne muscular dystrophy(DMD)is characterized by the development of cardiac pathology secondarily expressed in mitochondrial *** treatment includes support for mitochondrial *** purpose of this work was to evaluate the effects of the lipophilic cation MKT-077,capable of modulating mitochondrial activity,on the structure and function of cardiac mitochondria in mdx and wild-type mice,as well as the state of this ***:Animals were divided into 4 groups:wild type(WT),WT+5 mg/kg MKT-077,mdx,mdx+5 mg/kg ***-077 was administered intraperitoneally daily for 28 ***,we assessed the parameters of the functioning of the cardiac mitochondria of mice,the expression of genes encoding proteins involved in mitochondrial communication,as well as the histology and ultrastructure of the myocardium and heart ***:MKT-077 was shown to reverse mitochondrial hyperfunctionalization in mdx mice,reducing respiratory parameters to WT ***-077 also inhibited mitochondrial respiration in the hearts of WT ***-077 administration was accompanied by a tendency to normalize calcium retention capacity in the cardiac mitochondria of mdx *** ultrastructure and gene signature of mdx+MKT077 animals demonstrated increased *** noted changes in the sarcoplasmic reticulum(SR)/mitochondria contacts,accompanied by differential changes in the level of genes encoding proteins involved in the communication of these *** effect of MKT-077 was accompanied by normalization of the relative heart weight of mdx mice,a decrease in the level of fibrosis,and a tendency toward normalization of heart ***:MKT-077-induced reversal of cardiac mitochondrial hyperfunctionalization in mdx mice may promote a healthy-like state of the myocardium in these animals.

Numerical optimisation of a classical stochastic system for targeted energy transfer

The paper studies stochastic dynamics of a two-degree-of-freedom system,where a primary linear system is connected to a nonlinear energy sink with cubic stiffness nonlinearity and viscous *** the primary mass is subjected to a zero-mean Gaussian white noise excitation,the main objective of this study is to maximise the efficiency of the targeted energy transfer in the system.A surrogate optimisation algorithm is proposed for this purpose and adopted for the stochastic *** optimisations are conducted separately for the nonlinear stiffness coefficient alone as well as for both the nonlinear stiffness and damping coefficients *** different optimisation cost functions,based on either energy of the system’s components or the dissipated energy,are *** results demonstrate some clear trends in values of the nonlinear energy sink coefficients and show the effect of different cost functions on the optimal values of the nonlinear system’s coefficients.

Nanoparticles(NPs)can be transported via the nose-to-brain(N_(2)B)***,quantitative data on their spatiotemporal dynamics and regulation of the N_(2)B transport are largely *** surveyed metal oxide/hydroxide NPs as magnetic resonance imaging(MRI)contrasts for quantitative N_(2)B *** containing divalent transition metals were the only ones capable of N_(2)B *** T1-weighted(T1W)MRI,we showed that Mn_(3)O_(4)-NPs were readily engulfed by olfactory receptor neurons(ORNs)without disrupting olfactory sensing,we mapped their N_(2)B *** neurons,the Mn_(3)O_(4)-NPs were localized to the cytosol,mitochondria,vesicles,moved at mixed fast and slow axonal transport velocities intraand extra-vesicularly through *** NPs’axonal transport is dependent on neuronal activity and microtubule *** Mn_(3)O_(4)-NPs were trans-synaptically transmitted through at least four synapses across the olfactory ***-synaptic transmission of the NPs was dependent on N-type Ca^(2+)channels and NMDA receptors but blocked by GABAB receptor activation.A five-parameter Weibull signal increase/decrease model fitted to the T1W MRI data allowed for estimating kinetic parameters of Mn_(3)O_(4)-NP accumulation/*** and relative accumulation rates,but not elimination,correlated negatively with the number of synapses from ORNs,indicating a coupling of the NPs’N_(2)B transport with spontaneous neuronal ***,olfactory stimuli(2,5-dimethylpyrazine and acetophenone)significantly modulated and rerouted the Mn_(3)O_(4)-NP N_(2)B transport odor ***,the NPs’trans-synaptic transmission was impaired by aging and the onset of Parkinson’s *** data suggest new approaches to diagnostics,functional neuroimaging,controlling N_(2)B drug delivery.

Ultrafast supercontinuum generation in gas-filled waveguides is an enabling technology for many intriguing applications ranging from attosecond metrology towards biophotonics,with the amount of spectral broadening crucially depending on the pulse dispersion of the propagating *** this study,we show that structural resonances in a gas-filled antiresonant hollow core optical fiber provide an additional degree of freedom in dispersion engineering,which enables the generation of more than three octaves of broadband light that ranges from deep Uv wavelengths to near *** observation relies on the introduction of a geometric-induced resonance in the spectral vicinity of the ultrafast pump laser,outperforming gas dispersion and yielding a unique dispersion profile independent of core size,which is highly relevant for scaling input *** a krypton-filled fiber,we observe spectral broadening from 200 nm to 1.7μm at an output energy of B 23μJ within a single optical mode across the entire spectral *** show that the frequency generation results from an accelerated fission process of solitonlike waveforms in a non-adiabatic dispersion regime associated with the emission of multiple phase-matched Cherenkov radiations on both sides of the *** effect,along with the dispersion tuning and scaling capabilities of the fiber geometry,enables coherent ultra-broadband and high-energy sources,which range from the Uv to the mid‐infrared spectral range.
