Isolable Bis-BN-Based Analogues of Müller's Hydrocarbon

Müller's hydrocarbon and its analogues are classical examples of Kekulé diradicaloids and have gained great attention.However,because of their high reactivities,their isolation and structural characterizations are still challenging.Herein,we report the isolation of two bis-BN-based analogues of Müller's hydrocarbon (1 and 2) as crystalline solids in moderate yields.Experimental results and theoretical studies demonstrated that compound 1 possessing the terminal 1,3,2-diazaboryl groups is an open-shell singlet diradicaloid,which is in contrast to the closed-shell singlet ground states of the bis-BN-based analogues of Thiele's and Chichibabin's hydrocarbons with the same terminal groups,illustrating the strong influence of the central linkers on the ground state.Additionally,the replacement of the terminal 1,3,2-diazaboryl groups in 1 with the bis(2,4,6-triisopropylphenyl)boryl groups affords 2 with higher diradical character and a smaller singlet-triplet energy gap.

Quantitative prediction of shale gas sweet spots based on seismic data in Lower Silurian Longmaxi Formation,Weiyuan area,Sichuan Basin,SW China

Sweet spots in the shale reservoirs of the Lower Silurian Longmaxi Formation in Weiyuan 201 Block of Sichuan Basin were predicted quantitatively using seismic data and fuzzy optimization method. First, based on seismic and rock physics analysis, the rock physics characteristics of the reservoirs were determined, and elastic parameters sensitive to shale reservoirs with high gas content were selected. Second, data volumes with high precision of the elastic parameters were obtained from pre-stack simultaneous inversion. The horizontal distribution of key parameters for shale gas evaluation were calculated based on the results of rock physics analysis. Then, the fuzzy evaluation equation was established by fuzzy optimization method with test and logging data of horizontal wells with similar operation conditions. key parameters affecting the productivity of horizontal wells were sorted out and the weights of them in the sweet spots quantitative prediction were worked out by fuzzy optimization to set up a sweet spots evaluation system. Three classes of shale gas reservoirs which including two kinds of sweet spots were predicted with the above procedure, and the sweet spots have been predicted quantitatively by combining the above prediction results with the testing production. The testing results of 7 verification wells proved the reliability of the prediction results.

Radiotherapy is one of the main cancer treatments,but it may damage normal tissue and cause various side effects.At present,radioprotective agents used in clinics have side effects such as nausea,vomiting,diarrhea and hypotension,which limit their clinical application.It has been found that exosomes play an indispensable role in radiation injury.Exosomes are lipid bilayer vesicles that carry various bioactive substances,such as proteins,lipids and microRNA(miRNA),that play a key role in cell-to-cell communication and affect tissue injury and repair.In addition,studies have shown that radiation can increase the uptake of exosomes in cells and affect the composition and secretion of exosomes.Here,we review the existing studies and discuss the effects of radiation on exosomes and the role of exosomes in radiation injury,aiming to provide new insights for the treatment of radiation injury.

Chimeric antigen receptor T(CAR T)cell therapy has demonstrated efficacy in the treatment of haematologic malignancies.However,the accompanying adverse events,the most common of which is cytokine release syndrome(CRS),substantially limit its wide application.Due to its unique physiological characteristics,CRS in CAR T-cell treatment for B-cell non-Hodgkin lymphoma(BNHL)may exhibit some special features.Although existing guidelines had greatly promoted the recognition and management of CRS,many recommendations are not fully applicable to B-NHL.Therefore,it is imperative to identify responses that are specific to CRS observed following CAR T treatment for B-NHL.Based on underlying biological processes and known pathophysiological mechanisms,we tentatively propose a new model to illustrate the occurrence and evolution of CAR T-cell-therapy-related CRS in BNHL.In this model,tumour burden and bone marrow suppression are considered determinants of CRS.Novel phenomena after CAR T-cell infusion(such as local inflammatory response)are further identified.The proposed model will help us better understand the basic biology of CRS and recognize and manage it more rationally.

Microbial consortia are ubiquitous in nature,in which multiple microbial species cooperate to complete some important tasks such as lignocellulose degradation.Because of the advantages such as reduced metabolic burden and robustness to environment disturbances,developing a microbial consortium is a promising approach for valuable product synthesis,lignocellulose utilization,human health care,bioremediation and sustainable energy,etc.Despite the benefits,however,most artificial microbial consortia confront the problems of instability and low efficiency due to growth competition and metabolite incompatibility.To overcome these challenges,multiple strategies to design efficient synthetic microbial consortia have been reported.In this review,the interactions that determine the stability and performance of microbial consortia were described.Progress of artificial microbial consortia research was summarized,and the key strategies i.e.,spatial or temporal segregation,separated utilization of nutrients,nutrient cross-feeding and division of labor,that will be of great importance for achieving a stable and efficient microbial consortium were highlighted.Two novel advanced tools,signaling molecule systems and computational models,were also introduced and discussed.We believed that combining the universal cell–cell signaling molecule systems with computational models will be promising for synthetic microbial consortia construction in the future.

An analytical biomarker for treatment of patients with recurrent B-ALL after remission induced by infusion of anti-CD19 chimeric antigen receptor T(CAR-T) cells

Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. Combined analyses of laboratory biomarkers with their clinical manifestations before and after salvage treatment showed that the persistent immunosurveillance mediated by CAR-T-19 cells would inevitably potentiate the leukemia-killing effectiveness of subsequent chemotherapy in patients who showed relapse after CAR-T-19-induced remission.

针对塔里木盆地库车坳陷克深地区地震成像存在的静校正处理难度大、原始资料品质低、速度建模及叠前深度偏移难度大3大难题,研究盐下高陡构造成像技术。基于误差反向传播神经网络和最小平方QR分解双尺度层析反演方法获取复杂近地表高精度速度模型,解决盐下高陡构造地震成像静校正问题;在应用高精度静校正和均方根速度的基础上,采用十字排列锥体滤波和球面扩散补偿技术提高地震资料信噪比、恢复深层有效反射信号,解决盐下高陡构造原始地震数据品质低问题;在地质、测井、钻井等多信息的约束下,基于实体模型速度更新和网格层析速度反演的双尺度速度建模技术获取复杂地下构造的高精度速度模型,并应用真地表叠前深度偏移技术提高剧烈起伏地表条件下的地震成像效果,解决盐下高陡构造速度建模及叠前深度偏移问题。通过上述技术获得盐下高陡构造高质量地震成像成果,地震成像预测结果与实钻井吻合好,并成功部署3口超深井。
