dysregulation of G9a,a histone-lysine N-methyltransferase,has been observed in Alzheimer’s disease and has been correlated with increased levels of chronic inflammation and oxidative ***,microRNAs are involved in many biological processes and diseases playing a key role in pathogenesis,especially in multifactorial diseases such as Alzheimer’s ***,our aim has been to provide partial insights into the interconnection between G9a,microRNAs,oxidative stress,and *** better understand the biology of G9a,we compared the global microRNA expression between senescence-accelerated mouse-prone 8(sAMP8)control mice and sAMP8 treated with G9a inhibitor *** found a downregulation of miR-128 after a G9a inhibition treatment,which interestingly binds to the 3′untranslated region(3′-UTR)of peroxisome-proliferator activator receptor γ(PPARG)***,Pparg gene expression levels were higher in the sAMP8 group treated with G9a inhibitor than in the sAMP8 control *** also observed modulation of oxidative stress responses might be mainly driven Pparg after G9a *** confirm these antioxidant effects,we treated primary neuron cell cultures with hydrogen peroxide as an oxidative *** this setting,treatment with G9a inhibitor increases both cell survival and antioxidant ***,up-regulation of PPARγby G9a inhibitor could also increase the expression of genes involved in dNA damage responses and *** addition,we also described that the PPARγ/AMPK axis partially explains the regulation of autophagy markers ***,PPARγ/GAdd45αpotentially contributes to enhancing synaptic plasticity and neurogenesis after G9a ***,we propose that pharmacological inhibition of G9a leads to a neuroprotective effect that could be due,at least in part,by the modulation of PPARγ-dependent pathways by miR-128.

Generalized polynomial chaos expansion by reanalysis using static condensation based on substructuring

This paper presents a new computational method for forward uncertainty quantification(UQ)analyses on large-scale structural systems in the presence of arbitrary and dependent random *** method consists of a generalized polynomial chaos expansion(GPCE)for statistical moment and reliability analyses associated with the stochastic output and a static reanalysis method to generate the input-output data *** the reanalysis,we employ substructuring for a structure to isolate its local regions that vary due to random *** allows for avoiding repeated computations of invariant substructures while generating the input-output data *** substructuring with static condensation further improves the computational efficiency of the reanalysis without losing ***,the GPCE with the static reanalysis method can achieve significant computational saving,thus mitigating the curse of dimensionality to some degree for UQ under high-dimensional *** numerical results obtained from a simple structure indicate that the proposed method for UQ produces accurate solutions more efficiently than the GPCE using full finite element analyses(FEAs).We also demonstrate the efficiency and scalability of the proposed method by executing UQ for a large-scale wing-box structure under ten-dimensional(all-dependent)random inputs.

Ion heat transport in electron cyclotron resonance heated L-mode plasma on the T-10 tokamak

Anomalous ion heat transport is analyzed in the T-10 tokamak plasma heated with electron cyclotron resonance heating(ECRH) in second-harmonic extra-ordinary mode. Predictive modeling with empirical scaling for Ohmical heat conductivity shows that in ECRH plasmas the calculated ion temperature could be overestimated, so an increase of anomalous ion heat transport is required. To study this effect two scans are presented: over the EC resonance position and over the ECRH power. The EC resonance position varies from the high-field side to the low-field side by variation of the toroidal magnetic field. The scan over the heating power is presented with on-axis and mixed ECRH regimes. discharges with high anomalous ion heat transport are obtained in all considered regimes. In these discharges the power balance ion heat conductivity exceeds the neoclassical level by up to 10 times. The high ion heat transport regimes are distinguished by three parameters: the ratio Te/Ti, the normalized electron density gradient R/■, and the ion–ion collisionality νii~*. The combination of high Te/Ti, high νii~*, and R/■=6-10 results in values of normalized anomalous ion heat fluxes up to 10 times higher than in the low transport scenario.

While we eagerly anticipate sMILE’s(solar wind Magnetosphere Ionosphere Link Explorer)unprecedented X-ray observations of the Earth’s magnetosheath and the initiation of a new era of magnetospheric research,it seems appropriate to look ahead to the abilities of the next generation of astrophysics *** these,the Line Emission Mapper(LEM),a large aperture micro-calorimeter based mission,is currently planned to be able to observe the magnetosheath at high spectral resolution(~2 eV).With a field of view of~30′,LEM will allow higher spatial resolution and higher cadence measurement of the motion of a very small portion of the magnetopause over relatively short periods of time(multiple hours),complementing sMILE’s global ***’s strength is its spectral *** will be able to measure the abundance of a broad range of elements and ionization states,many of which are inaccessible to current in situ instruments,and will be able to separate the emission from the magnetosheath from the emission from the cosmic X-ray background using the difference in their relative velocities.

This paper details the development and testing of the first working prototype of the s-band high-power klystron,accomplished at the Budker Institute of Nuclear Physics,siberian Branch,Russian Academy of sciences(BINP sB RAs).Upon testing,the klystron demonstrated the following parameters:an operating frequency of 2856 MHz and a peak power output of 50 *** paper presents the klystron's design,its constituent units,and pertinent processing procedures,along with discussions on the measurement of its parameters.

Neuropathic pain is a severe and chronic condition widely found in the general *** reason for this is the extensive variety of damage or diseases that can spark this unpleasant constant feeling in *** the processing of pain,the dorsal root ganglia constitute an important region where dorsal root ganglion neurons play a crucial role in the transmission and propagation of sensory electrical ***,the dorsal root ganglia have recently exhibited a regenerative capacity that should not be neglected in the understanding of the development and resolution of neuropathic pain and in the elucidation of innovative ***,we will review the complex interplay between cells(satellite glial cells and inflammatory cells)and factors(cytokines,neurotrophic factors and genetic factors)that takes place within the dorsal root ganglia and accounts for the generation of the aberrant excitation of primary sensory neurons occurring in neuropathic *** importantly,we will summarize an updated view of the current pharmacologic and nonpharmacologic therapies targeting the dorsal root ganglia for the treatment of neuropathic pain.

The stockholm Convention on Persistent Organic Pollutants(POPs)is a legally binding instrument for 186 Parties(status:April 2023).Accordingly,among other responsibilities,countries are obliged to report the production,import,or export of the POPs listed in Annexes A,B,or C;provide information to registers;maintain inventories;and monitor the presence of POPs in the *** the broader context of international chemicals and waste management,producer responsibilities,harmonized reporting,and compliance with national and international regulations,Ecuador has addressed the newly listed group of perfluorinated alkyl substances(PFAs)in its national implementation plan and sent selected products from its national market for PFAs *** products analyzed came from the initially listed fields of specific exemptions and acceptable purposes,including:fire-fighting foams;photographic aids;greasers/degreasers;various kinds of paper/packaging;textiles;and leather,coatings,cleaners,metal plating,and *** results showed that the three PFAs presently listed in the stockholm Convention could be quantified in only a few samples;additional PFAs,not yet listed in the Convention also had low detection *** the number of samples was limited,the samples covered a large spectrum of sample matrices,making it possible to conclude that—once these products become waste and are regulated under the Basel Convention—they would not constitute a disposal ***,verification of the presence of PFAs in products on the market is expected to pose an analytical challenge for both,developed and developing countries.

A novel method for geometric quality assurance of rock joint replicas in direct shear testing-Part 2:Validation and mechanical replicability

Each rock joint is unique by nature which means that utilization of replicas in direct shear tests is required in experimental parameter ***,a method to acquire knowledge about the ability of the replicas to imitate the shear mechanical behavior of the rock joint and their dispersion in direct shear testing is *** this study,a novel method is presented for geometric quality assurance of *** aim is to facilitate generation of high-quality direct shear testing data as a prerequisite for reliable subsequent analyses of the *** Part 1 of this study,two quality assurance parameters,smf and V_(Hp100),are derived and their usefulness for evaluation of geometric deviations,*** reproducibility,is *** Part 2,the parameters are validated by showing a correlation between the parameters and the shear mechanical behavior,which qualifies the parameters for usage in the quality assurance *** results from direct shear tests presenting comparisons between replicas and the rock joint show that replicas fulfilling proposed threshold values of σ_(mf)d|V_(Hp100)|dispersion and imitate the shear mechanical behavior of the rock joint in all aspects apart from having a slightly lower peak shear *** wear in these replicas,which have similar morphology as the rock joint,is in the same areas as in the rock *** wear is slightly larger in the rock joint and therefore the discrepancy in peak shear strength derives from differences in material properties,possibly from differences in *** is shown by application of the suggested method that the quality assured replicas manufactured following the process employed in this study phenomenologically capture the shear strength characteristics,which makes them useful in parameter studies.

In this study,a calcium-phosphate coating was formed on a Mg-Mn-Ce alloy by the plasma electrolytic oxidation(PEO).The antibiotic vancomycin,widely used in the treatment of infections caused by staphylococcus aureus(***),was impregnated into the *** with vancomycin showed high bactericidal activity against *** mechanical and electrochemical properties of the formed coatings were studied,as well as in vitro cytotoxicity tests and in vivo tests on mature male rats were *** to sEM,Eds,XRd and XPs data,coatings had a developed morphology and contained hydroxyapatite,which indicates high *** analysis of roughness of coatings without and with vancomycin did not reveal any differences,confirming the high roughness of the *** electrochemical tests,an increase in corrosion resistance by more than two times after the application of PEO coatings was *** to the results of an in vivo study,after 28 days of the implantation of samples with calcium phosphate PEO coating and vancomycin,no signs of inflammation were observed,while an inflammatory reaction developed in the area of implantation of bare alloy,followed by *** release tests from the coatings show a sharp decrease in the concentration of the released antibiotic on day 7 and then a gradual decrease until day *** the experiment,no significant deviations in the condition and behavior of the animals were observed;clinical tests did not reveal a systemic toxic reaction.
