Effect of Ultraviolet Radiation on Hsp70 Protein Expression in HaCaT Cells

Ultraviolet radiation by its wavelength is divided into: UVA, UVB and UVC. Only UVA and UVB manage to penetrate the ozone layer, but due to anthropological activities, all of them are capable of interacting with humans to a greater or lesser extent, and can generate adverse effects such as cellular stress when interacting with intra-and extracellular biomolecules. The skin is the first organ in contact with UV radiation, and the stress it generates can be analyzed by the expression of a bioindicator of cellular d.m.ge such as Hsp70. Therefore, the objective of the project was: to determine the effect of UVA, UVB and UVC radiation on HaCaT epithelial cells, by analyzing the expression of Hsp70. materials and.m.thods: HaCaT cells were cultured in vitro, which were irradiated with UVA, UVB and UVC light at different doses, to subsequently determine the degree of Hsp70 expression by Immunodetection by PAGE-SdS and Western Blot. Results: Basal expression of Hsp70 was observed in no irradiated HaCaT cells. When HaCaT cells were irradiated with UVA, UVB, UVC, an increase in this Hsp70 protein was observed. With UVA, a higher degree of expression was observed at a time of 30 minutes of irradiation. With UVB the highest expression shifted to a time of 20 minutes. With UVC, overexpression was observed after 10 minutes. Conclusion: UV radiation generates cellular stress on HaCaT cells, evaluated by the stress bioindicator Hsp70. According to the wavelength of UV radiation, those that have a shorter wavelength have a greater potential for cellular d.m.ge, such as UVC.

Background In ambrosia and bark beetles–fungi interaction, volatile organic compounds(VOCs) play a central role in mediating various aspects of community dynamics of beetles and/or fungi. These functions include facilitating beetle habitat location, mate identification, and fungal partner differentiation. However, the understanding on this context remains limited, especially in the globally distributed subfamily Platypodinae, which comprises pred.m.-nantly ambrosia beetles. There is a lack of chemical data on ambrosia fungi from native South American species. This study addresses this gap by characterizing VOCs from twelve fungal species associated with megaplatypus mutatus and assessing species-specific behavioral responses during *** Fungal VOCs were collected by gas chromatography–mass spectrometry combined with solid-phase microextraction and Y-olfactometry assays of males and females were performed at dispersal stage. Statistical analyses involved: non-metric multid.m.nsional scaling multivariate plot and PERmAnOVA test, a cluster analysis through unweighted pair group method with Jaccard index, and finally, a chi-square goodness-of-fit test for beetle behavioral *** We identified 72 VOCs from the fungal species isolated from m. mutatus galleries, exocuticle, and gut. The olfactory behavior of m. mutatus d.m.nstrated its capacity to discriminate between volatile profiles, showing a pref-erence for either the fungus or the control source. directed x-rays produced in the interaction of sub-picosecond laser pulses of moderate relativistic intensity with plasma of near-critical density are investigated. Synchrotron-like (betatron) radiation occurs in the process of direct laser acceleration (dLA) of electrons in a relativisticlaser channel when the electrons undergo transverse betatron oscillations in self-generated quasi-static electric and.m.gnetic fields. In anexperiment at the PHELIX laser system, high-current directed beams of dLA electrons with a mean energy ten times higher than the ponderomotive potential and.m.ximum energy up to 100 meV were measured at 10^(19) W/cm^(2)laser intensity. The spectrum of directed x-raysin the range of 5–60 keV was evaluated using two sets of Ross filters placed at 0°and 10°to the laser pulse propagation axis. The differential x-ray absorption method allowed for absolute measurements of the angular-dependent photon fluence. We report 10^(13) photons/sr withenergies >5 keV measured at 0°to the laser axis and a brilliance of 10^(21) photons s^(−1) mm^(−2) mrad−2(0.1%BW)−1. The angular distributionof the emission has an FWHm of 14°–16°. Thanks to the ultra-high photon fluence, point-like radiation source, and ultra-short emissiontime, dLA-based keV backlighters are promising for various applications in high-energy-density research with kilojoule petawatt-class laserfacilities.

Forecasting of tropical cyclones ASAnI (2022) and.m.CHA (2023) over the Bay of Bengal - real time challenges to forecasters

This study examines the track and intensity forecasts of two typical Bay of Bengal tropical cyclones (TC) ASAnI and.m.CHA. The analysis of various numerical Weather Prediction (nWP) model forecasts [ECmWF (European Centre for medium range Weather Forecast), nCEP (national Centers for Environmental Prediction), nCUm (national Centre for medium Range Weather Forecast-Unified.m.del), Imd (India meteorological department), HWRF (Hurricane Weather Research and Forecasting)], mmE (multi-model Ensemble), SCIP (Statistical Cyclone Intensity Prediction) model, and OFCL (Official) forecasts shows that intensity forecasts of ASAnI and track forecasts of mOCHA were reasonably good, but there were large errors and wide variation in track forecasts of ASAnI and in intensity forecasts of mOCHA. Among all model forecasts, the track forecast errors of Imd.m.del and.m.E were least in general for ASAnI and.m.CHA respectively. Also, the landfall point forecast errors of Imd were least for ASAnI, and the mmE and OFCL forecast errors were least for mOCHA. no model is found to be consistently better for landfall time forecast for ASAnI, and the errors of ECmWF, Imd and HWRF were least and of same order for mOCHA. The intensity forecast errors of OFCL and SCIP were least for ASAnI, and the forecast errors of HWRF, Imd, nCEP, SCIP and OFCL were comparable and least for mOCHA up to 48 h forecast and HWRF errors were least thereafter in general. The ECmWF model forecast errors for intensity were found to be highest for both the TCs. The results also show that although there is significant improvement of track forecasts and limited or no improvement of intensity forecast in previous decades but challenges still persists in real time forecasting of both track and intensity due to wide variation and inconsistency of model forecasts for different TC cases. A multi-shot target assembly and automatic alignment procedure for laser–plasma proton acceleration at high repetition rate are introduced. The assembly is based on a multi-target rotating wheel capable of hosting more than 5000 targets,mounted on a 3d.m.torized stage to allow rapid replenishment and alignment of the target material between laser irradiations. The automatic alignment procedure consists of a detailed.m.pping of the impact positions at the target surface prior to the irradiation that ensures stable operation of the target, which alongside the purpose-built design of the target wheel, enables operation at rates up to 10 Hz. Stable and continuous laser-driven proton acceleration at 10 Hz is d.m.nstrated, with observed cut-off energy stability about 15%.

This work explored possibilities of biodegradable magnesium alloy mg-3Y preparation by two modern powder metallurgy techniques–spark plasma sintering(SPS)and selective laser melting(SLm).The powder material was consolidated by both methods utilising optimised parameters,which led to very low porosity(∼0.3%)in the SLm material and unmeasurably low porosity in the SPS *** main aim of the study was the thorough microstructure characterisation and interrelation between the microstructure and the functional properties,such as mechanical strength,deformability,and corrosion *** materials showed comparable strength of∼110 mPa in tension and compression and relatively good deformability of∼9%and∼21%for the SLm and SPS materials,*** corrosion resistance of the SPS material in 0.1 m naCl solution was superior to the SLm one and comparable to the conventional extruded *** digital image correlation during loading and the cross-section analysis of the corrosion layers revealed that the residual porosity and large strained grains have the d.m.nant negative effect on the functional properties of the SLm *** the other hand,one of the primary outcomes of this study is that the SPS consolidation method is very effective in the preparation of the W3 biodegradable alloy,resulting in material with convenient mechanical and degradation properties that might find practical applications.

Comparison of Treatment Efficacy of different number of Botulinum Toxin Type A Injection Sites for Crow's Feet Lines: A Single-Center Retrospective Clinical Study in Vietnam

Objective: Botulinum toxin type A is frequently used to treat crow's feet lines. However, the optimal dose and injection sites are still controversial. The objective of this study was to compare the efficacy of different botulinum toxin type A injection patterns for the treatment of crow's feet. methods: This single-center, retrospective, clinical study was conducted at the national Hospital of dermatology and Venereology from July 2020 to december 2020. data on 60 patients with crow's feet were collected and divided into 2 groups (3- or 4-point intramuscular injection) according to the intramuscular injection technique the patients received. The treatment efficacy was assessed based on the changes in the Crow's Feet Grading Scale score at 1, 4, and 16 weeks after treatment. Repeated.m.asures analysis of variance was used for the assessment of changes in scores over time between the two groups. Results: After treatment, the average Crow's Feet Grading Scale score was significantly decreased compared with the pretreatment score at all timepoints (1, 4, and 16 weeks) in both states (dynamic: For 3-point intramuscular injection technique, 1 week: 1.90 ± 0.71 vs. 2.97 ± 0.56; 4 weeks: 1.87 ± 0.68 vs. 2.97 ± 0.56; 16 weeks: 2.60 ± 0.67 vs. 2.97 ± 0.56. For 4-point intramuscular injection technique, 1 week: 1.73 ± 0.83 vs. 3.03 ± 0.49; 4 weeks: 1.73 ± 0.74 vs. 3.03 ± 0.49; 16 weeks: 2.57 ± 0.68 vs. 3.03 ± 0.49, all P nd static: For 3-point intramuscular injection technique, 1 week: 1.20 ± 0.89 vs. 2.20 ± 0.85; 4 weeks: 1.20 ± 0.89 vs. 2.20 ± 0.85; 16 weeks: 1.87 ± 0.97 vs. 2.20 ± 0.85; For 4-point intramuscular injection technique, 1 week: 1.50 ± 0.86 vs. 2.30 ± 0.84; 4 weeks: 1.33 ± 0.84 vs. 2.30 ± 0.84; 16 weeks: 1.87 ± 0.97 vs. 2.30 ± 0.84.
