Optimization and validation of a fast RP-HPLC method for the determination of dobutamine in rat plasma:Pharmacokinetic studies in healthy rat subjects

A novel isocratic reverse phase high performance liquid chromatography （RP-HPLC） with photo diode array （PdA） detection method for the determination of dobutamine （dBT） in rat plasma was developed and validated after optimization of various chromatographic conditions and other experimental parameters. Homoveratrylamiue was used as an internal standard. Methanol was used as the extracting solvent for the preparation of plasma samples. Samples were separated on a Symmetry C18 （250ram x4.6mm i.d., 5 pro） analytical column. Acetonitrile and 15raM potassium dihydrogen phosphate （pH 5.0 with 0.3% TEA）（20：80, v/v） was used. The column oven temperature was optimized at 35 ~C and the flow rate was 0.8 mL/min. The detection wavelength was fixed at 230 nm for entire analysis. The calibration curve was found to be linear over the concentration range of 50-2000 ng/mL （ra=0.9992）. The limit of quantification （LOQ） of the method was 50 ng/mL. The % RSd values of accuracy and precision values for intra and inter days were 〈 15% at quality control （QC） concentrations. Recovery, stability and robustness were studied within the acceptable range according to ICH guidelines. The method was efficiently applied to a pharmacokinetic study in healthy Wistar rats.

An isocratic reverse phase liquid chromatography (RP-LC) method has been developed and subsequently validated for the determination of Ranolazine in Bulk and its pharmaceutical formulation. Separation was achieved with a X-terra RP-18 ((Make: Waters Corporation;150 mm × 4.6 mm I.d.;particle size 5 μm)) Column and Sodium di-hydrogen phosphate monohydrate buffer with Tri ethyl amine (pH adjusted to 5.0 with diluted orthophosphoric acid): Acetonitrile (600:400) v/v as eluent at a flow rate of 1.0 mL/min. Uv detection was performed at 225 nm. The method is simple, rapid, and selective. The described method of Ranolazine is linear over a range of 11.98 μg/mL to 37.92 μg/mL. The method precision for the determination of assay was below 1.0%RSd. The percentage recoveries of active pharmaceutical ingredient (API) from dosage forms ranged from 99.1% to 100.9%. The results showed that the proposed method is suitable for the precise, accurate and rapid determination of Ranolazine in bulk, its capsule dosage forms.

Arterial supply of an intralobar pulmonary sequestration(IPS) from the coronary circulation is extremely rare. A significant coronary steal does not occur because of dual or triple sources of blood supply to sequestrated lung tissue. We present a 60-year-old woman who presented to us with repeated episodes of monomorphic ventricular tachycardia(vT) in last 3 mo. Radio frequency ablation was ineffective. On evaluation, she had right lower lobe IPS with dual arterial blood supply, i.e., right pulmonary artery and the systemic arterial supply from the right coronary artery(RCA). Stress myocardial perfusion scan revealed significant inducible ischemia in the RCA territory. Coronary angiogram revealed critical stenosis of proximal RCA just after the origin of the systemic artery supplying IPS. The critical stenosis in the RCA was stented. At 12 mo follow-up, she had no further episodes of vT or angina.

The increased expectation of automotive, aviation and marine industries pertaining to the enhanced properties and their use in elevated temperature conditions and also corrosive environments leads to the development of the newer material to meet the requirements. The requirements of the automobile and marine applications call for the increased mechanical properties and lower density accompanied with higher resistance to oxidation. Hence the present research work is aimed at the development of Hybrid metal matrix composites (HMMCs) using low-density base material and reinforcements. The aluminum of grade LM6 is preferred material in automobile industries, because it can be cast to any complex geometry and possess good machinability, further upon heat treatment, the properties of LM6 alloy can be enhanced to meet the industry requirements. However, requirements of automobile industries consist of increased mechanical properties, lower density and higher corrosion resistance. Hence, in the present research work, it is aimed to develop newer composite material using LM 6 grade Aluminum alloy as matrix material which is reinforced with varied percent Cenosphere and Red mud.

The effects of xylo-oligosaccharides(XOS) on broiler growth performance, immune function, and intestinal health were investigated. A total of 540 one-d-old Arbor Acres Plus broilers were randomly divided into 5 groups with 6 replicates per group and 18 chickens per replicate. Broilers in the control(CON) group received a corn-soybean meal based basal diet, those in the antibiotics(ANT) group received the basal diet plus 500 mg/kg oxytetracycline, and those in XOS groups received the basal diet plus 150, 300, or 450 mg/kg XOS. Compared with CON, the body weight at 42 d and average daily gain from 1 to 42 d were significantly increased in the 150, 450 mg/kg XOS-added and ANT groups(P = 0.018),and the relative expression of claudin-1 and ZO-1 m RNA in the ileum was significantly higher in the 300and 450 mg/kg XOS-added groups(P d conversion ratios(P d abdominal fat rates(P = 0.012) of broilers from 1 to 42 d of age were significantly lower in all XOS-added groups than in the control group. Splenic index(P = 0.036) and bursa of Fabricius index(P = 0.009) were significantly better in the ANT group and each XOS-added group than in the control group. Compared to CON and ANT,serum Ig A(P = 0.007) and Ig G(P = 0.002) levels were significantly higher in the 300 mg/kg XOS-added group, and the relative abundance of short-chain fatty acid-producing genera(Alistipes) was also significantly higher(P villus height(P d ratio of villus height to crypt depth(v:C)(P = 0.001) were significantly increased in XOS-added broilers. Quality of Service is an important attribute of a software system. In retrospect, performance assessment based on user interaction with the system has given a better understanding of underlying disciplines of the product. In this paper, we capture user interaction with the prototype/User Interface (UI). An approach for developing activity model from the user interface model is presented using workflows and functional elements. A methodology is proposed to transform UI into activity diagram. The approach is validated by an experimental setup using Amazon service. The performance of Amazon service is assessed using activity based performance prediction methodology, and the simulation results are obtained using SMTQA.

AIM: To study the effect of mobilized peripheral blood autologous Cd34 positive(Cd34+) cell infusion in patients with non-viral decompensated ***: Cirrhotic patients of non-viral etiology were divided into 2 groups based on their willingness to be listed for deceased donor liver transplant(ddLT)(control, n = 23) or to receive autologous Cd34+ cell infusion through the hepatic artery(study group, n= 22). Patients in the study group were admitted to hospital and received granulocyte colony stimulating factor injections 520 μg/d for 3 consecutive days to mobilize Cd34+ cells from the bone marrow. On day 4,leukapheresis was done and Cd34+ cells were isolated using CliniMAC magnetic cell sorter. The isolated Cd34+ cells were infused into the hepatic artery under radiological guidance. The patients were discharged within 48 h. The control group received standard of care treatment for liver cirrhosis and were worked up for ddLT as per protocol of the institute. Both groups were followed up every week for 4 wk and then every month for 3 ***: In the control and the study group, the cause of cirrhosis was cryptogenic in 18(78.2%) and16(72.72%) and alcohol related in 5(21.7%) and6(27.27%), respectively. The mean day 3 cell count(cells/μL) was 27.00 ± 20.43 with a viability of 81.84± 11.99%. and purity of 80%-90%. Primary end point analysis revealed that at 4 wk, the mean serum albumin in the study group increased significantly(2.83± 0.36 vs 2.43 ± 0.42, P = 0.001) when compared with controls. This improvement in albumin was,however, not sustained at 3 mo. However, at the end of3 mo there was a statistically significant improvement in serum creatinine in the study group(0.96 ± 0.33 vs 1.42 ± 0.70, P = 0.01) which translated into a significant improvement in the Model for End-Stage Liver disease score(15.75 ± 5.13 vs 19.94 ± 6.68,P = 0.04).
