Treatment with NAdPH oxidase inhibitor apocynin alleviates diabetic neuropathic pain in rats

Increased reactive oxygen species by the activation of NAdPH oxidase（NOX） contributes to the development of diabetic ***,a NOX inhibitor,increases sciatic nerve conductance and blood flow in diabetic *** investigated potential protective effect of apocynin in rat diabetic neuropathy and its precise mechanism of action at molecular *** models of streptozotocin-induced diabetes were treated with apocynin（30 and 100 mg/kg per day,intragastrically） for 4 *** hyperalgesia and allodynia were determined weekly using analgesimeter and dynamic plantar *** blot analysis and histochemistry/immunohistochemistry were performed in the lumbar spinal cord and sciatic nerve *** injection reduced pain threshold in analgesimeter,but not in *** treatment increased pain threshold *** blot analysis showed an increase in catalase and NOX-p47 phox protein expression in the spinal ***,protein expressions of neuronal and inducible nitric oxide synthase（n NOS,i NOS）,superoxide dismutase,glutathion peroxidase,nitrotyrosine,tumor necrosis factor-α,interleukin-6,interleukin-1β,aldose reductase,cyclooxygenase-2 or MAC-1（marker for increased microgliosis） in the spinal cord remained *** blot analysis results also demonstrated that apocynin decreased NOX-p47 phox expression at both doses and catalase expression at 100 mg/kg per *** of diabetic sciatic nerve revealed marked degeneration.n NOS and i NOS immunoreactivities were increased,while S-100 immunoreactivity（Schwann cell marker） was decreased in sciatic *** treatment reversed these changes *** conclusion,decreased pain threshold of diabetic rats was accompanied by increased NOX and catalase expression in the spinal cord and increased degeneration in the sciatic nerve characterized by increased NOS expression and Schwann cell *** treatment attenua

This study investigates the biological effects on a 3d scaffold based on hydroxyapatite cultured with MC3T3 osteoblasts in response to flow-induced shear stress(FSS).The scaffold adopted here(B-HA)derives from the biomorphic transformation of natural wood and its peculiar channel geometry mimics the porous structure of the *** the point of view of fluid dynamics,B-HA can be considered a network of micro-channels,intrinsically offering the advantages of a microfluidic *** work,for the first time,offers a description of the fluid dynamic properties of the B-HA scaffold,which are strongly connected to its *** features are necessary to determine the FSS ranges to be applied during in vitro studies to get physiologically relevant *** selected ranges of FSS promoted the elongation of the attached cells along the flow direction and early osteogenic cell *** data confirmed the ability of B-HA to promote the differentiation process along osteogenic ***,such a bioactive and naturally derived scaffold can be considered as a promising tool for bone regeneration applications.

MOdELING OF STATIC RECRYSTALLIZATION KINETICS OF Nb-BEARING STEELS

By using isothermal double hit compression tests and applying the 2% offset method, a new model was developed to predict the microstructural evolution of Nb-bearing steels at temperatures above and below the start temperature of strain-induced precipitation （Tp）. The Tp was developed as a function of true strain, initial austenite grain size and the Nb content. The activation energy of static recrystallization （Qrex） was expressed as a function of the content of different alloy elements. It was found that Nb played the most important role in increasing the value of Qrex, The microstructural observations and measurements confirmed the validity of the model developed in the present investigation.

Multifactorial nature of hepatocellular carcinoma drug resistance: Could plant polyphenols be helpful?

Primary hepatocellular carcinoma （HCC） is a quite frequent tumor which results in high mortality and most often exhibits a poor response to present drug therapies. Clearly, a thorough understanding of the biological bases of this malignancy might suggest new strategies for its treatment. Here we examine the evidences that both ＂pharmacological＂ mechanisms （e.g. drug transporter or detoxification enzyme over-expression） and alterations in other critical factors, including the IAPs （Inhibitory of Apoptosis Proteins）, involved in enhancement of cell survival and proliferation may determine the therapeutic resistance of HCC; we also underline the possible role in the process of the activation of transcription factors, like NF-κB, capable of contemporaneously up-regulating the mechanisms discussed. On this basis, we finally comment on the possible use of natural multi-targeted antitumoral agents like plant polyphenols to achieve sensitization to treatments in HCC.

Relationships between testate amoeba communities and water quality in Lake donghu, a large alkaline lake in Wuhan, China

The middle Yangtze Reach is one of the most developed regions of China. As a result, most lakes in this area have suffered from eutrophication and serious environmental pollution during recent decades. The aquatic biodiversity in the lakes of the area is thus currently under significant threat from continuous human activities. Testate amoebae （TA） are benthic （rarely planktonic） microorganisms characterized by an agglutinated or autogenous shell. Owing to their high abundance, preservation potential in lacustrine sediments, and distinct response to environmental stress, they are increasingly used as indicators for monitoring water quality and reconstructing palaeoenvironmental changes. However this approach has not yet been developed in China. This study presents an initial assessment of benthic TA assemblages in eight lakes of Lake donghu in the region of Wuhan, China. Testate amoeba community structure was most strongly correlated to water pH. In more alkaline conditions, communities were dominated by Centropyxis aculeata, difflugia oblonga, Pontigulasia compressa, Pon. elisa and Lesquereusia modesta. These results are consistent with previous studies and show that TA could be useful for reconstructing past water pH fluctuations in China. To achieve this, the next step will be to expand the database and build transfer function models.

Unusual case of insitu(intracystic) papillary carcinoma of breast

The term "intracystic papillary ductal carcinoma in situ ", has recently changed and is now more appropriately referred to as "intracystic papillary carcinoma'' constituting only 0.5% to 1% of all breast cancers. Herein, we discuss an unusual case of intracystic insitu papillary carcinoma of breast in a postmenopausal woman, the diagnosis of which was made on histopathology and confirmed by immunohistochemistry. Patient responded well to postoperative adjuvant radiotherapy without any recurrence, thereby preventing further morbidity and mortality related to invasion or tumor progression. So careful histopathological evaluation is the mainstay to arrive at the correct diagnosis to avoid untoward complications related to under diagnosis and /over diagnosis.

A simple method to model the reduced environment of lake bottom sapropel formation

The Kambala and Barchin brackish lakes （Baraba steppe, southern West Siberia） contain an organic-rich sapropel layer that was formed in oxygen-depleted waters. We measured the bulk sediment elemental composition, the water chemistry and determined the mineralogical composition and predominant biota species （diatoms and Cyanobacteria in phytoplankton community respectively） in the lakes. The result indicates that the first lake has a siliceous type of sapropel and the second a carbonaceous one. A computer thermodynamic model was developed for chemical interaction in water-bottom sediment systems of the Kambala and Barchin Lakes. The surface sodium bicarbonate waters are supersaturated with respect to calcite, magnesite （or low Mg-calcite）, quartz and chlorite with minor strontianite, apatite and goethite （pH 8.9-9.3, Eh 0.3 V）. Nevertheless, it is shown that during sapropel deposition, deep silt waters should be anoxic （Eh〈0 V）. The virtual component CH20 has been used to create an anoxic environment suitable for pyrite formation due to the biotic community impact and abiotic reduction. Thermodynamic calculation has shown that silt water is not necessarily euxinic （anoxic and sulfidic）. depending on Eh, sulfate sulfur can dominate in solution, causing the formation of gypsum together with pyrite. An attempt was made to find a reason for solution supersaturation with respect to Ca and Mg ions due to their complexation with humic acids.
