Type 1 diabetes(T1d)results from autoimmune destruction of insulin-producing b-cells in the pancreatic *** is an immediate need to restore both b-cell function and immune tolerance to control disease progression and ultimately cure ***,there is no effective treatment strategy to restore glucose regulation in patients with ***3-expressing Cd4^(+) regulatory T cells(Tregs)are potential candidates to control autoimmunity because they play a central role in maintaining ***,deficiencies in either naturally occurring Tregs(nTregs)themselves and/or their ability to control pathogenic effector T cells have been associated with ***,we hypothesize that nTregs can be replaced by FoxP3^(+) adaptive Tregs(aTregs),which are uniquely equipped to combat autoreactivity in *** nTregs,aTregs are stable and provide long-lived *** this review,we summarize the current understanding of aTregs and their potential for use as an immunological intervention to treat T1d.

Objective: To report atypical morphological features of Trichophyton(T.) tonsurans strains associated with tinea capitis. Methods: Eighty-two T. tonsurans strains isolated in Ceará, Brazil, were analyzed regarding macro and micromorphological features and nutritional patterns. Results: Fifty-two samples presented abundant chlamydoconidia, which were produced in chains. Macroscopically, these strains developed small glabrous colonies that were firmly attached to the surface of the culture medium, with few or no aerial mycelia and intense rusty yellow pigmentation. Seven strains did not grow with stimulus from thiamine. Samples were heterogeneous regarding urease production and none presented in vitro hair perforation. Conclusions: The observation of T. tonsurans strains with distinct phenotypic features indicates the need to revise the taxonomic criteria for routine identification of this dermatophyte.

Growth factor receptors （GFRs） are often aberrantly expressed in tumor cells, and altered GFR expressionand activity contribute to the pathogenesis of many types of cancer. A variety of mechanisms have been identified that result in enhanced GFR expression and activity in cancer cells. defects in the pathways responsible for GFR internalization and intraceilular trafficking are likely to be involved in altered GFR expression in a variety of cancers. The roles of GFR trafficking pathways in the regulation of GFR expression, in the pathogenesis of tumors, and in the response of tumors to treatment have not been fully delineated, but the likely contributions of GFR signaling to the development and progression of various malignancies suggest that therapies that modify GFR trafficking may be effective as anticancer treatments. The intraceHular trafficking of GFRs is regulated by a number of protein complexes and by protein ubiquitination. Many of the proteins required for this trafficking are products of tumor suppressor genes, and the expression and function of the protein machinery utilized for intracellular trafficking is frequently altered in tumor cells, consistent with the likely role of GFR trafficking in tumorigenesis. Many of the proteins involved in GFR trafficking have been identified as potential targets for anticancer treatment, and novel treatments directed against these targets are currently in preclinical development and in clinical trials. Ubiquitin ligases are critical for GFR trafficking and represent potentially important targets for the development of novel therapies. The genes for the ubiquitin ligases c-Cbl and UBE4B are located in chromosome regions commonly altered in a variety of tumors and therefore are likely to be important for tumorigenesis, c-Cbl nbiquitinates a number of GFRs and directs them for degradation. Ribokinase （EC 2.***.7.15） from Leishmania major was cloned, sequenced and overexpressed in Escherichia coli. The gene expressed an active enzyme that had comparable activity to the same enzyme studied in E. coli. It specifically phosphorylated d-ribose. Under defined conditions, the Km for the substrates d-ribose and ATP were 0.3±0.04 mM and 0.2±0.02mM, respectively. The turnover numbers of the enzyme for the substrates were 10.8s-1and 10.2 s-1, respectively. The enzyme product ribose 5-phosphate inhibited the phosphorylation of d-ribose with an apparent Kiof 0.4 mM, which is close to the Km（0.3mM） of dribose, suggesting that it might play a role in regulating flux through the enzyme.

Atomic simulations provide an effective means to understand the underlying physics of structural phase ***,this remains a challenge for certain allotropic metals due to the failure of classical interatomic potentials to represent the multitude of *** on machine-learning(ML)techniques,we develop a hybrid method in which interatomic potentials describing martensitic transformations can be learned with a high degree of fidelity from ab initio molecular dynamics simulations(AIMd).Using zirconium as a model system,for which an adequate semiempirical potential describing the phase transformation process is lacking,we demonstrate the feasibility and effectiveness of our ***,the ML-AIMd interatomic potential correctly captures the energetics and structural transformation properties of zirconium as compared to experimental and density-functional data for phonons,elastic constants,as well as stacking fault *** dynamics simulations successfully reproduce the transformation mechanisms and reasonably map out the pressure–temperature phase diagram of zirconium.

The efficacy of oral treatment with chewable tablets containing afoxolaner 2.27% w/w (NexGard?, Merial, now part of Boehringer-Ingelheim) or 1.875% w/w of afoxolaner and 0.375% w/w milbemycin oxime (NexGard Spectra?, Merial, now part of Boehringer-Ingelheim) was each assessed in eight dogs diagnosed with generalized demodicosis in this parallel group designed, blinded, randomized, single center negative controlled, efficacy study. Afoxolaner at the therapeutic dose (as close as possible to 2.5 mg/kg) was administered to Group 2 dogs, and afoxolaner (as close as possible to 2.5 mg/kg) with milbemycin oxime at 0.5 mg/kg was administered to Group 3 dogs on days 0, 28 and 56. All dogs were observed once daily for general health starting on day -7 until day 84. All dogs were clinically examined on days -7, -2, 14, 28, 42, 56, 70 and 84. dogs were weighed on days -7, -2, 27, 55, and 84. Live mite counts (based on five scrapings per dog and occasion) and clinical assessments, including photographic documentation, were performed on days -2, 28, 56 and 84. Three monthly treatments with NexGard or NexGard Spectra against generalized demodicosis in dogs were highly effective resulting in a 99.9% and 100% efficacy against mites, respectively. After treatment with NexGard, only three live mites were recovered in five scrapings in one dog on day 84. After treatment with NexGard Spectra, no mites were recovered in any scraping on any dogs in this group by day 84. Both treatments resulted in a marked reduction of skin lesions and >90% hair-regrowth at three months after the initial treatment. Reduction of live mite counts was consistent with reduction in the extent and severity of the skin changes. Although the arithmetic mean mite numbers in the negative control group decreased slightly during the study, infection persisted in 7 of the 8 control dogs until day 84. Both treatment groups of dogs showed a marked improvement of the associated dermatologic signs with steady improvement
