Axonal regeneration following surgical nerve repair is slow and often incomplete,resulting in poor functional recovery which sometimes contributes to lifelong ***,there are no FdA-approved therapies available to promote nerve *** accelerates axonal regeneration,but systemic side effects presently outweigh its potential benefits for peripheral nerve *** authors describe herein a biodegradable polyurethane-based drug delivery system for the sustained local release of tacrolimus at the nerve repair site,with suitable properties for scalable production and clinical application,aiming to promote nerve regeneration and functional recovery with minimal systemic drug *** is encapsulated into co-axially electrospun polycarbonate-urethane nanofibers to generate an implantable nerve wrap that releases therapeutic doses of bioactive tacrolimus over 31 *** and drug loading are adjustable for applications in small and large caliber nerves,and the wrap degrades within 120 days into biocompatible *** released from the nerve wrap promotes axon elongation in vitro and accelerates nerve regeneration and functional recovery in preclinical nerve repair models while off-target systemic drug exposure is reduced by 80%compared with systemic *** its surgical suitability and preclinical efficacy and safety,this system may provide a readily translatable approach to support axonal regeneration and recovery in patients undergoing nerve surgery.

A derivative-free frame-based conjugate gradients algorithm is presented. convergence is shown for c^1 functions, and this is verified in numerical trials. The algorithm is tested on a variety of low dimensional problems, some of which are ill-conditioned, and is also tested on problems of high dimension. Numerical results show that the algorithm is effective on both classes of problems. The results are compared with those from a discrete quasiNewton method, showing that the conjugate gradients algorithm is competitive. The algorithm exhibits the conjugate gradients speed-up on problems for which the Hessian at the solution has repeated or clustered eigenvalues. The algorithm is easily parallelizable.

Background Octogenarians constitute an increasing proportion of patients presenting for non-emergency percutaneous coronary intervention （PcI）. Methods This study evaluated the in-hospital procedural characteristics and outcomes, including the bleeding events of 293 octogenarians presenting between january 2010 and december 2012 for non-emergency PcI to a single large volume tertiary care Aus- tralian center. comparisons were made with 293 consecutive patients aged less than or equal to 60 years, whose lesions were matched with the octogenarians. Results Non-ST elevation myocardial infarction was the most frequent indication for non-emergency PcI in octoge- narians. compared to the younger cohort, they had a higher prevalence of co-morbidities and more complex coronary disease, comprising more type c and calcified lesions. Peri-procedural use of low molecular weight heparin （LMWH; 1.0% vs. 5.8%; P 〈 0.001） and glycopro- tein IIb/IIIa inhibitors （2.1% vs. 9.6%; P 〈 0.001） was lower, while femoral arterial access was used more commonly than in younger patients （80.9% vs. 67.6%; P 〈 0.001）. Overall, there was a non-significant trend towards higher incidence of all bleeding events in the elderly （9.2% vs. 5.8%; P = 0.12）. There was no significant difference in access site or non-access site bleeding and major or minor bleeding between the two cohorts. Sub-analysis did not reveal any significant influence on bleeding rates by the use of LMWH, glycoprotein IIb/IIIa inhibitors or femoral arterial access. In addition, there were no significant differences in the rates of in-hospital mortality, stroke or acute stent thrombosis between the two groups. conclusions In this single center study, we did not observe significant increases in adverse in-hospital outcomes including the incidence of bleeding in octogenarians undergoing non-emergency PcI.

Alley cropping of cassia siamea and maize was studied in semi arid Kenya for soil fertility improvement. Katumani composite maize was planted except in the short rains of 1988 (SR88) when a hybrid variety was sown. Therefore the grain yield per row increased differently in the alley cropped maize (cM). Sole maize (SM) and cM yields were higher in SR88 than in the long rains of 1988 (LR88) by 62% and 38%, while yields from the same treatments in LR89 were only 21% and 45% of those in SR88 These differences in relative maize yields are attributed to differences between the two maize varieties in competition under nutrient stress conditions.

Magnetic resonance studies of jets in a gas-solid fluidised bed

Magnetic resonance imaging （MRI） has been used to study the behaviour Of jets at the distributor of a 50 mm diameter fluidised bed of 0.5 mm diameter poppy seeds. Two perforated-plate distributors were examined, containing either 10 or 14 holes, each 1 mm diameter. Ultra-fast MR imaging was able to show the transient nature of the upper parts of the jets, where discrete bubbles are formed. Imaging in 3d showed that the central jets were the longest for flow rates below minimum fluidisation. Above minimum fluidisation, the outer jets, nearest the wall of the fluidised bed, arched inward towards the central axis. In this latter case, interpretation of the time-averaged 3d image required the use of ultra-fast MR imaging to identify the approximate height above the distributor at which discrete bubbles were formed. The apparently continuous void extending along the central axis above this height in the time-averaged 3d image was thus identified, using ultra-fast MR imaging, as representing the averaged paths of released bubbles. Time-averaged MR velocity mapping was also used to identify dead zones of stationary particles resting on the distributor between the jets. The dead zones could be observed when the superficial velocity of the gas approached minimum fluidisation, but they were smaller than those observed at lower gas superficial velocity. comparable images of a single jet through 1.2 mm diameter poppy seeds from MRI and electrical capacitance volume tomography （EcVT） are also demonstrated.

Traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease are three distinct neurological disorders that share common pathophysiological mechanisms involving neuroinflammation. One sequela of neuroinflammation includes the pathologic hyperphosphorylation of tau protein, an endogenous microtubule-associated protein that protects the integrity of neuronal cytoskeletons. Tau hyperphosphorylation results in protein misfolding and subsequent accumulation of tau tangles forming neurotoxic aggregates. These misfolded proteins are characteristic of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease and can lead to downstream neuroinflammatory processes, including assembly and activation of the inflammasome complex. Inflammasomes refer to a family of multimeric protein units that, upon activation, release a cascade of signaling molecules resulting in caspase-induced cell death and inflammation mediated by the release of interleukin-1β cytokine. One specific inflammasome, the NOd-like receptor protein 3, has been proposed to be a key regulator of tau phosphorylation where it has been shown that prolonged NOd-like receptor protein 3 activation acts as a causal factor in pathological tau accumulation and spreading. This review begins by describing the epidemiology and pathophysiology of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease. Next, we highlight neuroinflammation as an overriding theme and discuss the role of the NOd-like receptor protein 3 inflammasome in the formation of tau deposits and how such tauopathic entities spread throughout the brain. We then propose a novel framework linking traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease as inflammasomedependent pathologies that exist along a temporal continuum. Finally, we discuss potential therapeutic targets that may intercept this pathway and ultimately minimize long-term neurological decline.
