Objectivc Cloned animals in several species,including sheep(Schnieke et al.1997),cattle(Bordignon et al. 2003),mice(Wakayama et al.1998),goats(Baguisi et al.1999),cats(Shin et al.2002),horses(galli et al.2003),dogs(Lee et al.2005) and rabbits(Li et al.2006c),have been successfully produced by somatic cell nuclear transfer(NT).Although several groups have conducted NT with pigs(Betthauser et al.2000;Onishi et al.2000;Polejaeva et al.2000;Boquest et al. 2002),the efficiency is extremely low,withd embryos surviving to term(Martinez et al.2003). Therefore,it is particularly important to improve the efficiency of cloning in *** recent years,du et *** the blastocyst rate with the handmade cloning method to be 50-80%with ten piglets delivered(du et al.2007).In the field of medicine,miniature pigs are the preferred animals for pulmonary drugs toxicity tests and metabolic *** is well known that many human diseases can be treated by organ allotransplantation,but the shortage of human organ sources presents a significant barrier for *** of organs from miniature pigs has been considered as a suitable option because it has many advantages,such as compatibility in terms of size and physiology and a potentially large supply(Cozzi et al.2000).Multiple factors render pig cloning *** regard to the technique,pig oocytes are difficult to enucleate because their cytoplasm contains many lipid *** ’blind’ enucleation method involves removal of the nucleus through the first polar body and the adjacent cytoplasm(Onishi et al.2000).Yet,the inner development environment of oocytes is cytoplasm and its main functions are metabolism and maintain structure of oocytes *** to the oocytes occurs by removal of most of the cytoplasm during the ’blind’ method and it cannot be clearly confirmed whether the karyon in the oocytes has been ***,some investigators have used the centrifugal
