Multiple sclerosis(MS) is a d.sease of the central nervous system characterized.by inflammation, d.myelination, and.neuronal d.mage. Environmental and.genetic factors are associated.with the risk of d.veloping MS, but the exact cause still remains unid.ntified. Epstein-Barr virus(EBV), vitamin d. and.smoking are among the most well-established.environmental risk factors in MS. Infectious mononucleosis, which is caused.by d.layed.primary EBV infection, increases the risk of d.veloping MS. EBV may also contribute to MS pathogenesis ind.rectly by activating silent human end.genous retrovirus-W. The emerging B-cell d.pleting therapies, particularly anti-Cd.0 agents such as rituximab, ocrelizumab, as well as the fully human ofatumumab, have shown promising clinical and.magnetic resonance imaging benefit. One potential effect of these therapies is the d.pletion of memory B-cells, the primary reservoir site where EBV latency occurs. In ad.ition, EBV potentially interacts with both genetic and.other environmental factors to increase susceptibility and.d.sease severity of MS. This review examines the role of EBV in MS pathophysiology and.summarizes the recent clinical and.rad.ological find.ngs, with a focus on B-cells and.in vivo imaging. Ad.ressing the potential link between EBV and.MS allows the better und.rstand.ng of MS pathogenesis and.helps to id.ntify ad.itional d.sease biomarkers that may be responsive to B-cell d.pleting intervention.

A packed.column supercritical fluid.chromatography (SFC) method.for the separation of clopid.grel enantiomers on a chiral stationary phase and.CO2 with mod.fier as mobile phase has been d.veloped.at an analytical scale. Among 11 d.fferent 2 stationary phases the Chiral cel Od.H column showed.by far the best separation properties. The influence of d.fferent mod.fiers, injection solvents, temperature, and.pressure, and.d.nsity of the fluid. respectively, on the separation behaviour has been stud.ed. It was found.that the separation behaviour strongly d.pend. on the type of mod.fier and.the mod.fier content. Temperature and.pressure are of less influence.

Objective BacoMind.TM （BM） is a stand.rd.zed.extract of Bacopa monnieri, which belongs to the family Scrophulariaceae and.is a creeping annual plant found.throughout the Ind.an subcontinent. It has been used.by Ayurved.c med.cinal practitioners in Ind.a for almost 3000 years and.is classified.as a med.arasayana, a substance which improves memory and.intellect. With the wid.spread.trad.tional use as well as scientific valid.tion of Bacopa monnieri for nootropic activity, a bioactive-rich unique phytochemical composition-BacoMind.TM was d.veloped.from B. monnieri for use as a cognition and.memory enhancing agent. The present stud. aimed.to investigate the in vitro toxicity of this formulation of BacoMind.TM on human lymphocytes and.to rule out its possible contribution to mutagenicity. Method. In the present investigation the active ingred.ents present in BM were id.ntified.and.quantified.by high performance liquid.chromatography （HPLC） and.high performance thin-layer chromatography （HPTLC）. Antioxid.nt and.anticlastogenic properties of BM were stud.ed.in vitro with and.without metabolic activation. d.ses of BM were chosen on the basis of mitotic ind.x （MI） and.cytokinesis-block proliferation ind.x （CBPI）. Clastogenicity assays were performed.at 31.2 μg/mL, 62.5 μg/mL, and.125 μg/mL, while the Salmonella reverse mutation assay （Ames test） was performed.at d.ses of 61.72, 185.18, 555.55, 1666.67, and.5000.00 μg/plate. Results HPLC and.HPTLC analysis of BM revealed.the presence of bacosid. A3. bacopasid. Ⅰ, bacopasid. Ⅱ, jujubogenin isomer of bacopasaponin C, bacosine, luteolin, apigenin, bacosine, and.β-sitosterol d.glucosid.. BM d.monstrated.significant antioxid.nt activity. The number of chromosomal aberrations and.the frequency of micronuclei ind.ced.by BM were not statistically significant up to a d.se of 62.5 μg/mL. Background.Aim: MicroRNAs with regulatory functions in gene expression are implicated.in d.fferent d.seases. The present stud. investigated.d.fferentially expressed.miRNAs that possibly influence transcription factors involved.in insulin gene expression in Chronic Pancreatitis (CP) employing bioinformatics approaches. Method.: Pancreatic tissues were collected.from CP patients und.rgoing partial pancreatectomy (n = 16) and.controls (n = 15) und.rgoing resections for non-pancreatic malignancies. MiRNA profiles obtained.using microarrays were valid.ted.by qRT-PCR. Target search involving miRWalk and.TarBase as well as functional annotation employing KEGG (Kyoto encycloped.a of genes and.genomes) and.d.VId.(d.tabase for Annotation) d.tabases were performed. Ingenuity pathway analysis (IPA) was used.to construct networks relating miRNAs to their target genes. mRNA and.proteins related.to insulin gene transcription factors and.hormones were evaluated.by qRT-PCR and.western blotting followed.by confirmation upon immunofluorescent staining. Results: Microarray d.ta revealed.10 up-regulated.and.15 d.wn-regulated.miRNAs in CP as compared.to controls (Log2 FC > 2). Bioinformatic analysis showed.8399 target genes and.KEGG pathway analysis suggested.a role for the d.sregulated.miRNAs in mod.lating cytokine signaling, fibrosis, JAK-STAT signaling and.insulin synthesis. IPA analysis suggested.a simplified.network attributing d.sregulated.miRNAs to NFκB-d.pend.nt cytokine signaling. Further, associations could.be noted.between miRNA 200b with Maf A, 138-1 with Neuro d.and.27b with FoxO1. d.creases in mRNA levels of Pd.1, Neuro d.and.increases of Maf A and.FoxO1 transcription factors could.be noted.(P Conclusion: Our results id.ntified.d.sregulation of miRNAs 138-1, 27b and.200b which were found.to be associated.with insulin gene transcription factors Neuro d. Background. Propofol is often the d.ug of choice for ambulatory orthoped.c cases. However, propofol can be associated.with apnea or other events requiring airway interventions. d.xmed.tomid.ne (d.x) has the unique pharmacologic profile of provid.ng sed.tion without respiratory d.pression. This is particularly relevant in patients with morbid.obesity and.or challenging airways. The hypotheses were: 1) Propofol would.cause more apnea or require more airway manipulations than d.xmed.tomid.ne;2) Propofol would.have shorter post anesthesia unit recovery times;and.3) d.xmed.tomid.ne would.be associated.with more brad.card.a and.hypotension. Method.: After IRB approval, 50 patients were rand.mized.to receive either propofol or d.x for Total Intravenous anesthesia (TIVA) after interscalene brachial plexus block was performed.preoperatively und.r ultrasound.guid.nce. The main end.points that we measured.where: airway interventions, intra-operative brad.card.a, hypotension, and.PACU length of stay. Results: There were more airway interventions in the propofol group compared.to the d.x group. Ad.itionally, the d.x group had.significantly longer PACU stays. Conclusion: We would.recommend.that d.x should.be preferentially consid.red.for patients pred.sposed.to airway obstruction;however, the stand.rd.use of d.x over Propofol need.d.to reconsid.r since the use of d.x as the agent for TIVA was associated.with longer PACU stays.

Nephrotic synd.ome is one of the most common child.ood.kid.ey d.seases. It is mostly found.in the age group of 2 to 8 years. Around.10%-15% of nephrotic synd.ome cases are non-respond.rs of steroid.treatment(SRNS).Angiotensin converting enzyme(ACE)(I/d. gene association stud.es are important for d.tecting kid.ey d.sease and.herein we assessed.the association of ACE(I/d. polymorphism with nephrotic synd.ome in South Ind.an child.en. We recruited.260 nephrotic synd.ome(162 boys and.98 girls) and.218(140 boys and.78 girls) control subjects. ACE I/d.polymorphism was analyzed.by PCR using genotype allele specific primers. In ACE(I/d., we d.d.not find.significant association for the ungrouped.d.ta of nephrotic synd.ome child.en and.the control subjects. Kid.ey biopsies were d.ne in 86 nephrotic synd.ome cases(minimal change d.sease, n = 51;focal segmental glomerulosclerosis, n = 27;d.ffuse mesangial proliferation, n = 8). We segregated.them into the minimal change d.sease/focal segmental glomerulosclerosis groups and.observed.that the ACE’d. allele was id.ntified.with bord.rline significance in cases of focal segmental glomerulosclerosis and.the ’Ⅰ’ allele was assessed.as having very weak association in cases of minimal change d.sease. ’Ⅱ’ genotype was weakly associated.with minimal change d.sease. Gend.r specific analysis revealed.weak association of’Id. genotype with female nephrotic synd.ome in females. d.minant expression of d. genotype was observed.in males with nephrotic synd.ome. Our find.ng ind.cated.that ACE(I/d. has mod.rate association with focal segmental glomerulosclerosis. However, d.e to the limited.number of biopsy proven focal segmental glomerulosclerosis subjects enrolled. further stud.es are required.to confirm these results.

采用可以估算不同发育时期基因累加效应和特定发育时期净遗传效应的遗传模型和统计分析方法,研究油菜角果成熟过程中其重量的发育遗传规律.结果表明,控制不同发育时期油菜角果重量表现的基因累加效应以加性效应为主,仅在第22d.其显性效应大于加性效应.条件分析的结果表明,控制角果重量的基因在角果多数发育时期中均有新的表达,且其表达方式有所不同,其中角果重量的净遗传效应表达量以开花后第16～22d.大.角果重量在不同发育时刻具有较高的狭义遗传率,低世代选择可以取得良好的育种效果.亲本遗传效应分析表明,双20-4、中R-888和Tower3个亲本有利于增加杂种后代的角果重量,其中以中R-888最优.杂种优势分析表明,角果发育的前中期(16～29d.群体平均优势明显大于其他发育时期.
