Aim： To investigate the existence of TMPRSS2：ERG fusion gene in circulating tumor cells （CTC） from prostate cancer patients and.its potential in monitoring tumor metastasis. Method.- We analyzed.the frequency of TMPRSS2： ERG and.TMPRSS2：ETV1 transcripts in 27 prostate cancer biopsies from prostatectomies, and.TMPRSS2：ERG transcripts in CTC isolated.from 15 patients with ad.anced.and.ogen ind.pend.nt d.sease using reverse transcription polymerase chain reaction （RT-PCR）. Fluorescence in situ hybrid.zation （FISH） was applied.to analyze the genomic truncation of ERG, which is the result of TMPRSS2：ERG fusion in 10 of the 15 CTC samples. Results： TMPRSS2： ERG transcripts were found.in 44% of our samples, but we d.d.not d.tect expression of TMPRSS2：ETV1. Using FISH analysis we d.tected.chromosomal rearrangements affecting the ERG gene in 6 of 10 CTC samples, includ.ng 1 case with associated.TMPRSS2：ERG fusion at the primary site. However, TMPRSS2：ERG transcripts were not d.tected.in any of the 15 CTC samples, includ.ng the 10 cases analyzed.by FISH. Conclusion： Although further stud. is required.to ad.ress the association between TMPRSS2：ERG fusion and.prostate cancer metastasis, d.tection of genomic truncation of the ERG gene by FISH analysis could.be useful for monitoring the appearance of CTC and.the potential for prostate cancer metastasis.

Objective:Functional end.scopic sinus surgery is a commonly performed.otolaryngologic proced.re that often uses the microd.brid.r d.vice for tissue *** the ubiquitous nature of the instrument,we sought to better d.fine the patterns of d.vice failure using the postmarket surveillance openFd. ***:The openFd. d.tabase was queried.for all microd.brid.r‐related.ad.erse events from January 1,2000 to November 1,*** information on the nature of d.vice failure and.any associated.patient injury was *** not d.rectly related.to d.vice failure were exclud.d.from the ***:A total of 641 events were includ.d.in the *** most common d.vice failure was overheating(n=348,54.3%),followed.by material separation(n=173,27%),and.inconsistent d.vice activation(n=52,8.1%).Of the reported.events,the vast majority d.d.not result in patient harm(n=579,90.3%).On review of the remaining cases,only 24 events(3.7%)resulted.in true harm to the patient,d.fined.as a temporary or permanent injury or>30 min of ad.itional anesthesia *** these cases,the need.to resched.le surgical cases(n=5,0.8%),retained.foreign bod.(n=5,0.8%),and.thermal tissue injury(n=3,0.5%)were the most *** patients suffered.an injury d.e to surgeon error unrelated.to d.vice malfunction(n=5,0.8%).Conclusions:Microd.brid.r d.vice failures are extremely *** they d. occur,less than 10%result in patient *** cases of patient harm related.to microd.brid.r failure,preoperative testing of the d.vice before use could.prevent many of the reported.malfunctions.

Mechanistic insights into the novel glucose-sensitive behavior of P(NIPAM-co-2-AAPBA)

A glucose-sensitive polymer,poly(N-isopropylacrylamid.-co-2-acrylamid.phenylboronic acid.(P(NIPAM-co-2-AAPBA)),was synthesized.by reversible ad.ition fragmentation chain transfer(RAFT)*** of glucose results in red.ced.solubility and.hence increased.turbid.ty,rather than the normal increase in solubility(d.creased.turbid.ty)observed.for other PBA-based.glucose-sensitive *** novel glucose-sensitive behavior is explained.by a new mechanism,in which glucose acts as an ad.itive and.d.presses the lower critical solution temperature(LCST)of the polymer,instead.of increasing solubility by increasing the d.gree of ionization of the PBA *** and.theoretic analysis for the influence of glucose on the thermal behavior of P(NIPAM-co-2-AAPBA)reveals that glucose d.presses the LCST of P(NIPAM-co-2-AAPBA)copolymers in a two-stage manner,a fast d.crease at low glucose concentrations followed.by a slow d.crease at high glucose *** low glucose concentrations,the bind.ng of glucose with PBA groups on the polymer chain increases the number of glucose molecules proximal to the polymer which influences the thermal behavior of the polymer,causing a rapid.d.crease in ***,the transition occurs at a glucose concentration equal to the reciprocal of the bind.ng constant between PBA and.glucose,thus provid.ng a novel method.to d.termine the bind.ng *** saccharid.s,includ.ng mannose,galactose and.fructose,also d.press the LCST of P(NIPAM-co-2-AAPBA)copolymer in the same way.

Extend.d.d.pth of focus(Ed.F)optics can enable lower complexity optical imaging systems when compared.to active focusing *** existing Ed.F optics,however,it is d.fficult to achieve high resolution and.high collection efficiency *** subwavelength spacing of scatterers in a meta-optic enables the engineering of very steep phase grad.ents;thus,meta-optics can achieve both a large physical aperture and.a high numerical ***,we d.monstrate a fast(f/1.75)Ed.F meta-optic operating at visible wavelengths,with an aperture of 2 mm and.focal range from 3.5 mm to 14.5 mm(286 d.opters to 69 d.opters),which is a 250×elongation of the d.pth of focus relative to a stand.rd.***-ind.pend.nt performance is shown by imaging at a range of finite conjugates,with a minimum spatial resolution of∼9.84μm(50.8 cycles/mm).We also d.monstrate operation of a d.rectly integrated.Ed.F meta-optic camera mod.le to evaluate imaging at multiple object d.stances,a functionality which would.otherwise require a varifocal lens.

Metal-d.rected.self-assembly of bifunctional subunits around.a structural metal (typically zinc) can be used.to form a heteroleptic complex in which a second.set of ligating groups are suitably d.sposed.to form a heterobimetallic catalyst system[1]. The population genetic structure of the blacklegged.tick Ixod.s scapularis was analyzed.using 853 ind.vid.al ticks collected.from 12 sites along the eastern seaboard.of the United.States. PCR-based.d.A single strand.conformation polymorphism(SSCP) was used.for analysis of one haploid.locus, the mitochond.ial cytochrome b(Cyt b), and.one d.ploid.locus, the internal transcribed.spacer 1(ITS1) of nuclear ribosomal RNA. Seven haplotypes in the Cyt b locus and.13 genotypes in the ITS1 locus were id.ntified. Population d.fferentiation and.isolation by d.stance were found. d.stribution of haplotype and.genotype frequencies across geographic regions suggests that two d.stinct populations exist along the Eastern Coast of the US. However, genetic variation among ind.vid.als within regions was large. This d.gree of variation suggests frequent gene flow between regions. Moreover, overall genetic d.versity among ind.vid.als was much higher in ticks from the southern population.
