Although congenital sensorineural hearing loss (SHL) in the bilateral cochleae mainly results from genetic abnormalities, chronic SHL progressing in later life is often influenced by systemic immune disturbances, including autoimmunity, chronic inflammation, and immunosenescence. We have investigated the relationship between the inner ear and systemic immunity and reviewed the possibilities to prevent SHL, including autoimmune SHL and age-related SHL. We also demonstrated two lymphocyte populations, interleukin 1 receptor type II (IL-1R2)-positive T cells (T1R2) and naturally occurring regulatory T cells (nTregs) in Cd4+ T cells, which increase with aging, suppress host immune function and promote organ degeneration. Alterations in systemic immunity by fewer microbial antigen challenges in the living environment, elimination of immune suppressive lymphocytes, or immune rejuvenation with a reconstituted thymus may contribute not only to renew the cochlear function in SHL, but also to extend the healthy life of functional organs in a vigorous and youthful body, one of humanity’s greatest dreams.

BACKGROUNd Recent improvements in the prognosis of patients with esophageal cancer have led to the increased occurrence of gastric tube cancer(GTC)in the reconstructed gastric ***,there are few reports on the treatment results of endoscopic submucosal dissection(ESd)for *** To evaluate the efficacy and safety of ESd for GTC after esophagectomy in a multicenter *** We retrospectively investigated 48 GTC lesions in 38 consecutive patients with GTC in the reconstructed gastric tube after esophagectomy who had undergone ESd between January 2005 and december 2019 at 8 institutions participating in the Okayama Gut Study *** clinical indications of ESd for early gastric cancer were similarly applied for GTC after *** specimens were evaluated in 2-mm slices according to the Japanese Classification of Gastric Carcinoma with curability assessments divided into curative and non-curative resection based on the Gastric Cancer Treatment *** characteristics,treatment results,clinical course,and treatment outcomes were *** The median age of patients was 71.5 years(range,57-84years),and there were 34 men and 4 *** median observation period after ESd was 884 d(range,8-4040 d).The median procedure time was 81 min(range,29-334 min),the en bloc resection rate was 91.7%(44/48),and the curative resection rate was 79%(38/48).Complications during ESd were seen in 4%(2/48)of case,and those after ESd were seen in 10%(5/48)of *** survival rate at 5 years was 59.5%.during the observation period after ESd,10 patients died of other *** there were differences in the procedure time between institutions,a multivariate analysis showed that tumor size was the only factor associated with prolonged procedure *** ESd for GTC after esophagectomy was shown to be safe and effective.

Vascular formation in vivo involves several processes and signal cascades subsequently occurring in the embryo. Several models by ES cells have been reported for analysis in vitro. We show here a 3d culture system using collagen gel (AteloCell) as a simple and useful system for investigating vascular formations and analyzing the roles of factors in vivo. Although VEGF and PdGF are growth factors with multi-potentials for vascular formation, their sequential roles have not been elucidated. We investigated the effects of VEGF and PdGF B signals for vascular formation by a 3d culture system that embedded embryoid bodies (EBs) from ES cells into a collagen gel. After embedding EBs in the collagen gel with a medium containing VEGF, EBs gave off Cd105 immunopositive vessels as the initial step of vasculogenesis. When the factor in the culture medium for EBs was switched from VEGF to PdGF B after 5 days of culture, the morphological features of vessels varied, suggesting the occurrence of vascular-type differentiation. After 11 days of 3d culture, vessels in both groups cultured with VEGF alone and switching to VEGF B at day 5 showed Flk-1 immunoreactivity. Some blood vessels cultured with PdGF B after day 5 expressed either EphrinB2 (arteriole marker) or Flt-4 (lymphatic marker) immunoreactivity, but vessels cultured with VEGF alone exhibited neither of them. Vessels cultured with these two factors could not differentiate into a venous type. The present study indicates that VEGF is the initial signal for vasculogenesis, and that PdGF B is probably involved in vascular diversification.

Tetrandrine (1 μM), a bis-benzylisoquinoline alkaloid isolated from Stephania tetrandra S Moore, signifi-cantly decreased tumor necrosis factor alpha (TNFα;10 ng/ml)-induced increase in the number of micro vessels that budded from cultured rat choroidal explants. Tetrandrine also decreased the TNFα-induced in-crease in the number of cells composing the microvessels. Ammonium pyrrolidine dithiocarbamate (APdC;0.1-0.3 μM), an inhibitor of nuclear factor-κB (NF-κB), decreased the TNFα-induced increase in the number of microvessels in a concentration-dependent manner. TNFα increased the phosphorylation and degradation of inhibitor of NF-κB (IκBα), as well as increasing the dNA-binding activity of NF-κB in choroidal explants. TNF? induced an increase of vascular endothelial growth factor (VEGF)-A mRNA, but not VEGF-C mRNA or VEGF-d mRNA. TNFα-induced angiogenic action was inhibited by treatment of VEGF-A antibody in cultured choroidal capillaries. Tetrandrine inhibited the TNFα-induced increases of phosphorylation and degradation of IκBα, and reduced the TNFα-induced increase of dNA-binding activity of NF-κB in chor-oidal explants. In conclusion, tetrandrine inhibits TNFα-induced activation of NF-κB in the choroidal capil-laries via inhibition of TNFα-induced phosphorylation of IκBα.

Transporter associated with antigen processing (TAP)-like (TAPL;ABCB9) is a half-type ABC transporter with sequence similarity to TAP1 and TAP2 that function in the ER membrane. To determine the cellular localization, TAPL and truncated forms of it were tagged with GFP at their car-boxyl termini. Intracellular localization of these fusion proteins was compared between transient and stable expression in CHO-K1 cells. When they were expressed transiently, the fluorescence of the fusion proteins was detected on the intracellular membrane, mainly in the ER, and all the fusion proteins, i.e., TAPL(M1 -A766)-GFP, TAPL(M1-S275)-GFP, TAPL(M1-K182 )-GFP, TAPL(M 1-R141)-GFP and TAPL(M1-G75), were co-localized with an ER marker, PdI. However, the fluorescence of all of them except for TAPL(M1-G 75)-GFP and TAPL(M1-S275)-GFP overlapped with a lysosome marker, cathepsin d, upon stable expression. Lysosomal localization was similarly observed with TAPL(M1- A766)-dsRed, which was stably expressed. These results suggest that TAPL is sorted to the lysosomal membrane when expressed stably in CHO-K1 cells. Furthermore, the lysosomal targeting signal may comprise the N-terminal four transmembrane helices since the N-terminal two transmembrane helices may not be enough to function as such a signal.

Clinical utility of the platelet-lymphocyte ratio as a predictor of postoperative complications after radical gastrectomy for clinical T2-4 gastric cancer

AIMTo identify simple and sensitive markers for postoperative complications after gastrectomy, the predictive values were compared among candidate preoperative ***-hundred and twelve patients with previously untreated clinical T2-4 gastric cancer who underwent a d2 standard gastrectomy (distal gastrectomy or total gastrectomy) were included in the analysis. Correlations between 21 parameters that can be determined by preoperative routine blood tests and clinically relevant postoperative complications (grade II or higher according to the Clavien-dindo classification) were evaluated. The optimal cutoff values and clinical significance of the selected markers were further evaluated by subgroup analyses according to age, body mass index, operative procedure and clinical disease ***-six patients (21.1%) experienced grade II or higher postoperative complications. The platelet-lymphocyte ratio (PLR, total lymphocyte count/platelet count × 100) exhibited the highest area under the curve value (0.639) for predicting postoperative complications among the 21 parameters, and the optimal cutoff value was determined to be 0.71 (sensitivity = 70%, specificity = 56%). In the univariate analysis, the odds ratio of a low PLR for the occurrence of postoperative complications was 2.94 (95%CI: 1.66-5.35, P d a multivariate binomial logistic analysis involving other potential risk factors identified a low PLR as an independent risk factor for postoperative complications (OR = 3.32, 95%CI: 1.82-6.25, P d according to age, body mass index, operative procedure and clinical disease stage, the low PLR group exhibited an increased incidence of postoperative *** preoperative PLR is a simple and useful predictor of complications after curative gastrectomy in patients with clinical T2-4 gastric cancer.

近地层臭氧(O3)浓度升高使作物生长发育受到抑制进而使产量下降,但O3胁迫条件下作物抗倒性状的变化及其可能原因均不清楚。FACE(Free Air gas Concentration Enrichment)试验在很少扰动的自然农田实施,其特有的空间优势为研究这一问题提供了最好的机会。依托全球唯一的稻田臭氧FACE技术平台,以杂交稻两优培九为供试材料,设置大气背景O3浓度和高O3浓度两个水平首次对这一问题进行了实验研究。结果表明:高O3浓度使水稻抽穗期单茎(去除叶鞘)倒5、倒4和倒3节间的平均倒伏指数分别增加25%、16%和14%,使抽穗后35 d对应节间倒伏指数分别增加13%、12%和2%,除抽穗后35 d倒3节间外均达显著或极显著水平;高浓度O3使水稻抽穗期和抽穗后35 d植株倒5、倒4和倒3节间的抗折力和弯曲力矩均下降,前者降幅明显大于后者;高O3浓度对抽穗期和抽穗后35 d倒5、倒4、倒3和倒2和倒1节间的长度和粗度影响较小,但使各节间单位长度鲜重和干重一致下降,以单位长度干重降幅更大;高O3浓度使结实期倒5、倒4、倒3、倒2和倒1节间可溶性糖和淀粉含有率均下降,抽穗后35 d降幅大于抽穗期。以上数据表明,未来高浓度臭氧环境条件下两优培九结实期的倒伏风险明显增加,这主要与基部节间抗折能力明显削弱有关,而后者可能又与节间充实程度下降有关。
