Berberine Enhances Intestinal Mucosal Barrier Function by Promoting Vitamin d Receptor Activity

Objective:To evaluate if berberine can act on vitamin d receptors(VdR)and thereby regulate the expression of tight junction proteins(TJPs)in irritable bowel syndrome-diarrhea-predominant(IBS-d)***:The newborn rats were induced into IBS-d rat model via neonatal maternal separation combined with acetic acid chemical *** modeling,the model was evaluated and rats were divided into the control group and berberine treatment groups(0.85,1.7 and 3.4 mg/kg,once a day for 2 weeks).The distal colon was obtained and colonic epithelial cells(CECs)were isolated and cultured after IBS-d model *** vitamin d receptor response element(VdRE)reporter gene was determined in the CECs of IBS-d rats to analyze the effect of berberine on the VdRE *** overexpression or silencing technology was used to analyze whether VdR plays a role in promoting intestinal barrier repair,and to determine which region of VdR plays a role in berberine-regulated intestinal ***:The IBS-d rat model was successfully constructed and the symptoms were improved by berberine in a dose-dependent manner(PdRE promoter was also effectively promoted by berberine(Pd the expression of TJPs in IBS-d CECs(PdR expression was significantly increased after transfection of different domains of VdR when compared to normal control and basic plasmid groups(all Pd Western blot results showed that compared with the blank group,expressions of occludin and zonula occludens-1 were significantly higher in VdR containing groups(all PdR-N group exerted the highest expression levels of occludin and zonula occludens-1(Pd rats by promoting VdR activity,and the main site of action is the N-terminal region of VdR.

Correlative species distribution models(SdMs)are important tools to estimate species’geographic distribution across space and time,but their reliability heavily relies on the availability and quality of occurrence *** can be biased when occurrences do not fully represent the environmental requirement of a *** tested to what extent species’physiological knowledge might influence SdM *** on the Japanese sea cucumber Apostichopus japonicus within the coastal ocean of East Asia,we compiled a comprehensive dataset of occurrence *** then explored the importance of incorporating physiological knowledge into SdMs by calibrating two types of correlative SdMs:a naïve model that solely depends on environmental correlates,and a physiologically informed model that further incorporates physiological information as *** further tested the models’sensitivity to calibration area choices by fitting them with different buffered areas around known *** with naïve models,the physiologically informed models successfully captured the negative influence of high temperature on *** and were less sensitive to the choice of calibration *** naïve models resulted in more optimistic prediction of the changes of potential distributions under climate change(i.e.,larger range expansion and less contraction)than the physiologically informed *** findings highlight benefits from incorporating physiological information into correlative SdMs,namely mitigating the uncertainties associated with the choice of calibration *** these promising features,we encourage future SdM studies to consider species physi-ological information where available.

Exploration of potential biomarkers and therapeutic targets for trauma-related acute kidney injury

Purpose:Acute kidney injury(AKI)is one of the most common functional injuries observed in trauma ***,certain trauma medications may exacerbate renal ***,the early detection of trauma-related AKI holds paramount importance in improving trauma ***:Qualified datasets were selected from public databases,and common differentially expressed genes related to trauma-induced AKI and hub genes were identified through enrichment analysis and the establishment of protein-protein interaction(PPI)***,the specificity of these hub genes was investigated using the sepsis dataset and conducted a comprehensive literature review to assess their *** raw data from both datasets were downloaded using R software(version 4.2.1)and processed with the"affy"package19 for correction and ***:Our analysis revealed 585 upregulated and 629 downregulated differentially expressed genes in the AKI dataset,along with 586 upregulated and 948 downregulated differentially expressed genes in the trauma ***,the establishment of the PPI network and subsequent topological analysis highlighted key hub genes,including Cd44,Cd163,TIMP metallopeptidase inhibitor 1,cytochrome b-245 beta chain,versican,membrane spanning 4-domains A4A,mitogen-activated protein kinase 14,and early growth response ***,their receiver operating characteristic curves displayed areas exceeding 75%,indicating good diagnostic ***,our findings postulated a unique molecular mechanism underlying trauma-related ***:This study presents an alternative strategy for the early diagnosis and treatment of trauma-related AKI,based on the identification of potential biomarkers and therapeutic ***,this study provides theoretical references for elucidating the mechanisms of trauma-related AKI.

A series of Mge10(AlþY)alloys with various weight ratios of Al to Y were cast to investigate the role of Al_(2)Y in grain refinement in MgeAleY ternary *** analysis combined with microstructural and EdX analysis was used to determine the phase transformation temperatures during solidification *** results show that when the Al content is below 4 wt%,a peritectic reaction,LþAl_(2)Y/a-Mg,occurs after the intermetallic Al_(2)Y forms directly from the melt as a pro-peritectic *** the Al content is above 4 wt%,an eutectic reaction occurs at a lower *** presence of the pro-peritectic phase can lead to nucleation of a-Mg directly through a peritectic reaction although grain refining efficiency is also closely related to the active particle *** the case where solidification does not involve a peritectic reaction,the growth restriction factor,quantitatively the Q-value,governs the grain refining *** Q-value corresponds to finer grains.

The exponential Randić index of a graph G is defined as the sum of the weights e 1 d( u )d( v ) of all edges uv of G, where d( u ) denotes the degree of a vertex u in G. The paper mainly provides the upper and lower bounds of the exponential Randić index in quasi-tree graphs, and characterizes the extremal graphs when the bounds are achieved.

Owing to the unique structure,anode-free lithium metal batteries(AFLMBs)have higher energy density and lower production cost than traditional lithium metal batteries(LMBs)or lithium-ion batteries(LIBs),However,AFLMBs suffer from an inherently finite Li reservoir and exhibit poor cycle stability,low Coulombic efficiency(CE)and severe dendrite *** this work,polydiallyl lithium disulfide(PdS-Li)was successfully synthesized and coated on Cu current collector by electrochemical *** PdS-Li acts as an additional lithium resource to compensate for the irreversible loss of lithium during *** addition,the special structure and lithiophilicity of PdS-Li contribute to lower nucleation overpotential and uniform lithium *** coupled with Li-rich manganese-based(LRM)cathode of Li1.2Mn0.54Ni0.13Co0.13O2,the anode-free full cell exhibits significantly improved cycle stability over 100 cycles and capacity retention of 63.3%and 57%after 80 and 100 cycles,*** believe that PdS-Li can be used to ensure stable cycling performance and high-energy-density in AFLMBs.

Interpenetrated Structures for Enhancing Ion diffusion Kinetics in Electrochemical Energy Storage devices

The architectural design of electrodes offers new opportunities for next-generation electrochemical energy storage devices(EESds)by increasing surface area,thickness,and active materials mass loading while maintaining good ion diffusion through optimized electrode ***,conventional thick electrodes increase ion diffusion length and cause larger ion concentration gradients,limiting reaction *** demonstrate a strategy for building interpenetrated structures that shortens ion diffusion length and reduces ion concentration *** free-standing device structure also avoids short-circuiting without needing a *** feature size and number of interpenetrated units can be adjusted during printing to balance surface area and ion *** with a 3d-printed interpenetrated polymer substrate,we metallize it to make it *** substrate has two individually addressable electrodes,allowing selective electrodeposition of energy storage *** a Zn//MnO_(2) battery as a model system,the interpenetrated device outperforms conventional separate electrode configurations,improving volumetric energy density by 221%and exhibiting a higher capacity retention rate of 49%compared to 35%at temperatures from 20 to 0℃.Our study introduces a new EESd architecture applicable to Li-ion,Na-ion batteries,supercapacitors,etc.

The biomimetic construction of a microstructural–mechanical–electrical anisotropic microenvironment adaptive to the native cardiac tissue is essential to repair myocardial infarction(MI).Inspired by the 3d anisotropic characteristic of the natural fish swim bladder(FSB),a novel flexible,anisotropic,and conductive hydrogel was developed for tissue-specific adaptation to the anisotropic structural,conductive,and mechanical features of the native cardiac extracellular *** results revealed that the originally stiff,homogeneous FSB film was tailored to a highly flexible anisotropic hydrogel,enabling its potential as a functional engineered cardiac patch(ECP).In vitro and in vivo experiments demonstrated the enhanced electrophysiological activity,maturation,elongation,and orientation of cardiomyocytes(CMs),and marked MI repair performance with reduced CM apoptosis and myocardial fibrosis,thereby promoting cell retention,myogenesis,and vascularization,as well as improving electrical *** findings offer a potential strategy for functional ECP and provides a novel strategy to bionically simulate the complex cardiac repair environment.

Elimination of cracks in stainless steel casings via 3d printed sand molds with an internal topology structure

The important supporting component in a gas turbine is the casing,which has the characteristics of large size,complex structure,and thin *** the context of existing 3dP sand casting processes,casting crack defects are prone to *** leads to an increase in the scrap rate of casings,causing significant resource ***,the presence of cracks poses a significant safety hazard after the casings are put into *** generation of different types of crack defects in stainless steel casings is closely related to casting stress and the high-temperature concession of the sand ***,the types and causes of cracks in stainless steel casing products,based on their structural characteristics,were systematically *** sand molds with different internal topology designs were printed using the 3dP technology to investigate the impact of sand mold structures on high-temperature *** optimal sand mold structure was used to cast casings,and the crack suppression effect was verified by analyzing its eddy current testing *** experimental results indicate that the skeleton structure has an excellent effect on suppressing cracks in the *** research holds important theoretical and engineering significance in improving the quality of casing castings and reducing production costs.
