Simvastatin Action Is Not Related to HdAC2 Expression in Non-Small Cell Lung Cancer (NSCLC)

In this study, we lowered the expression of HdAC2 protein, to evaluate the effects of simvastatin on the biochemical pathways involved in inflammatory and metastatic response. The model used is the non-small cell lung cancer line (GLC-82). Trypan blue staining for assessing vital cell number to be seed and MTT assay was used as cell proliferation test. Lentivus for HdAC2 was used to silence its mRNA. Western blotting analysis was used for protein extracts, and ELISA was done on culture media for cytokines (IL-6, IL-8 and TNF-alpha) release. Hydrogen peroxide (H2O2) was used to induce oxidative stress. Our results have shown that Lentivirus containing the shHdAC2 in GLC-82 cells was able to reduce protein expression of HdAC2. In the GLCshHdAC2 cell line obtained, H2O2 induced a significant increase in cytokines release and ERK1/2 phosphorylation (P 0.01);a significant decrease of RECK activation (P 0.01);a significant increased activation (P 0.01) of both MMP-2 and MMP-9 and an increased activation of NF-κB, Myd88, TRAF-6, TRAdd, TRAF-2. In GLCshHdAC2 cell, the treatment with simvastatin (30 μM), significantly affected all the biochemical markers examinated (P 0.01). In conclusion, from our report emerge, that simvastatin is able per se to inhibit oxidative stress in lung cancer cells, overcoming HdAC2 expression.

Self-assembled bright luminescent hierarchical materials from a tripodal benzoate antenna and heptadentate Eu(III) and Tb(III) cyclen complexes

The europium heptadentate coordinatively unsaturated (Eu(III)) and the terbium (Tb(III)) 1,4,7,10-tetraazacyclododecane (cyclen) complexes 1 and 2 were used in conjunction with ligand 3 (1,3,5-benzene-trisethynylbenzoate) to form the supramolecular self-assembly structures 4 and 5;this being investigated in both the solid and the solution state. The resulting self-assemblies gave rise to metal centered emission (both in the solid and solution) upon excitation of 3, confirming its role as a sensitizing antenna. drop-cased examples of ligand 3, and the solid forms of 4 and 5, formed from both organic and mixture of organic-aqueous solutions, were analyzed using Scanning Electron Microscopy, which showed significant changes in morphology;the ligand giving rise to one dimensional structures, while both 4 and 5 formed amorphous materials that were highly dense solid networks containing nanoporous features. The surface area (216 and 119 m2·g^-1 for 4 and 5 respectively) and the ability of these porous materials to capture and store gases such as N2 investigated at 77 K. The self-assembly formation was also investigated in diluted solution by monitoring the various photophysical properties of 3–5. This demonstrated that the most stable structures were that consisting of a single antennae 3 and three complexes of 1 or 2 (e.g., 4 and 5) in solution. By monitoring the excited state lifetimes of the Eu(III) and Tb(III) ions in H2O and d2O respectively, we showed that their hydration states (the q-value) changed from -2 to 0, upon formation of the assemblies, indicating that the three benzoates of 3 coordinated directly to the each of the three lanthanide centers. Finally we demonstrate that this hierarchically porous materials can be used for the sensing of organic solvents as the emission is highly depended on the solvent environment;the lanthanide emission being quenched in the presence of acetonitrile and THF, but greatly enhanced in the presence of methanol.

Grafting affects yield and phenolic prof ile of Solanum melongena L. landraces

The influence of grafting on agronomical and qualitative characteristics of four Sicilian eggplant landraces was investigated. Grafted and ungrafted plants were compared in the open field in the northern coast of Sicily. Solanum torvum seedlings were used as rootstock. Regardless of genotypes tested, grafting significantly increased total fruit production, marketable production, and number of marketable fruits, but did not affect weight of marketable fruits and waste production. Landrace 2（Sciacca）, with black epidermal tissue and pyriform fruit shape, when grafted onto S. torvum not only gave a higher yield performance than ungrafted plants, but also showed a higher phenolic antioxidant content. Landrace 4（Sicilia）, with black epidermal tissue and small cylindrical fruits also benefited, when grafted onto S. torvum, from a substantial increase in antioxidant fruit content. As consumers＇ demand for fruits and vegetables rich in compounds important for human health is steadily increasing, these landrace/rootstock combinations should deserve more attention by plant nurseries involved in grafted seedling production and interested in the valorization and conservation of eggplant biodiversity.

Venous thrombosis and prothrombotic factors in inflammatory bowel disease

Patients with inflammatory bowel disease (IBd) may have an increased risk of venous thrombosis (VTE). PubMed, ISI Web of Knowledge and Scopus were searched to identify studies investigating the risk of VTE and the prevalence of acquired and genetic VTE risk factors and prothrombotic abnormalities in IBd. Overall, IBd patients have a two- to fourfold increased risk of VTE compared with healthy controls, with an overall incidence rate of 1%-8%. The majority of studies did not show significant differences in the risk of VTE between Crohn’s disease and ulcerative colitis. Several acquired factors are responsible for the increased risk of VTE in IBd: inflammatory activity, hospitalisation, surgery, pregnancy, disease phenotype (e.g., fistulising disease, colonic involvement and extensive involvement) and drug therapy (mainly steroids). There is also convincing evidence from basic science and from clinical and epidemiological studies that IBd is associated with several prothrombotic abnormalities, including initiation of the coagulation system, downregulation of natural anticoagulant mechanisms, impairment of fibrinolysis, increased platelet count and reactivity and dysfunction of the endothelium. Classical genetic alterations are not generally found more often in IBd patients than in non-IBd patients, suggesting that genetics does not explain the greater risk of VTE in these patients. IBd VTE may have clinical specificities, namely an earlier first episode of VTE in life, high recurrence rate, decreased efficacy of some drugs in preventing further episodes and poor prognosis. Clinicians should be aware of these risks, and adequate prophylactic actions should be taken in patients who have disease activity, are hospitalised, are submitted to surgery or are undergoing treatment.

The prolongation of life expectancy and the drastic reduction of fertility rate are the primary cause of an agingworld. It is projected that the elderly (above 65) will increase within the next 25 years by 82% , whereas the new bornonly by 3% . despite the enormous medical progress during the past few decades, the last years of life are still accom-panied by increasing ill health and disability. The ability to maintain active and independent living for as long as possi-ble is a crucial factor for aging in health and dignity. Therefore, the promotion of healthy aging and the prevention ofdisability in men, must assume a central role in medical research and medical practice as well as in the formulation ofnational health and social policies. Effective programs promoting health and aging will ensure a more efficient use ofhealth and social services and improve the quality of life in older persons by enabling them to remain independent andproductive. The most important and drastic gender differences in aging are related to organs and or systems dependantor influenced by reproductive hormones. In distinction to the course of reproductive aging in women, with the rapid de-cline in sex hormones and expressed by the cessation of menses, aging men experience a slow and continuous decline ofhormones. This decline in endocrine function involves; A decrease of testosterone, dehydroepiandrosterone (dHEA),oestrogens, thyroid stimulating hormone (TSH), growth hormone (GH), insulin-like growth factor-1 (IGF-1), andmelatonin. This decrease is concomitant with an increase of LH and FSH. In addition sex hormone binding globulin's(SHBG) increase with age resulting in further lowering the concentrations of free biologically active androgens. Inter-ventions such as hormone replacement therapy may prevent, delay or alleviate the debilitating conditions which may re-sult from secondary partial endocrine deficiency. The advantages of Low Pressure Carburizing in the Heat Treatment Subcontracting Business

Low Pressure Carburizing (LPC) was introduced in the 90’s in the western Europe Heat Treatment business, mainly for in-house applications where it was especially appreciated for Carburizing of transmission parts. However the success of the LPC units installed for gears Carburizing in the automotive industry has hindered the development of the process in other fields - like subcontracting business -, where its advantages deserve to be enlightened. After a brief review of the principle of the process, the interest of its classical application to transmission parts is described, underlining peculiarly the reduction of the distortion observed when LPC is associated with high pressure gas quenching. Then the less-known advantages of the LPC process, like the high accuracy and reproducibility of the results, the modeling possibility and the simulation easiness, the case-depth uniformity and the full flexibility of the units are considered, showing how they can be beneficial to subcontracting business.

Birds caught in spider webs in Asia

A recent global review of birds caught in spider webs reported only three Asian cases. Given this surprisingly low number, I made a concerted effort to obtain additional Asian cases from the literature, the internet, and field workers. I present a total of 56 Asian cases which pertain to 33 bird species. As in the global dataset, mostly small bird species were caught in spider webs, with a mean body mass of 17.5 g and a mean wing chord length of 73.1 mm. Conse?quently, birds with a body mass >30 g were very rarely caught. This Asian review corroborates the global review that smaller birds are more likely to be caught and that Nephila spiders are most likely to be the predators. Continuous monitoring of spider webs is recommended to ascertain the frequency of these events.

Stage migration vs immunology: The lymph node count story in colon cancer

Lymph node staging is of crucial importance for the therapy stratification and prognosis estimation in colon cancer. Beside the detection of metastases,the number of harvested lymph nodes itself has prognostic relevance in stage Ⅱ/Ⅲ cancers. A stage migration effect caused by missed lymph node metastases has been postulated as most likely explanation for that. In order to avoid false negative node staging reporting of at least 12 lymph nodes is recommended. However,this threshold is met only in a minority of cases in daily practice. due to quality initiatives the situation has improved in the past. This,however,had no influence on staging in several studies. While the numbers of evaluated lymph nodes increased continuously during the last decades the rate of node positive cases remained relatively constant. This fact together with other indications raised doubts that understaging is indeed the correct explanation for the prognostic impact of lymph node harvest. Several authors assume that immune response could play a major role in this context influencing both the lymph node detectability and the tumor's behavior. Further studies addressing this issue are need. Based on the findings the recommendations concerning minimal lymph node numbers and adjuvant chemotherapy should be reconsidered.
