SARS-CoV-2 continues to threaten human society by generating novel variants via mutation and recombination.The high number of mutations that appeared in emerging variants not only enhanced their immune-escaping ability but also made it difficult to predict the pathogenicity and virulence based on viral nucleotide sequences.Molecular markers for evaluating the pathogenicity of new variants are therefore needed.By comparing host responses to wild-type and variants with attenuated pathogenicity at proteome and metabolome levels,six key molecules on the polyamine biosynthesis pathway including putrescine,SAM,dc-SAM,ODC1,SAMS,and SAMDC were found to be differentially upregulated and associated with pathogenicity of variants.To validate our discovery,human airway organoids were subsequently used which recapitulates SARS-CoV-2 replication in the airway epithelial cells of COVID-19 patients.Using ODC1 as a proof-ofconcept,differential activation of polyamine biosynthesis was found to be modulated by the renin-angiotensin system(RAS)and positively associated with ACE2 activity.Further experiments demonstrated that ODC1 expression could be differentially activated upon a panel of SARS-CoV-2 variants of concern(VOCs)and was found to be correlated with each VOCs’pathogenic properties.Particularly,the presented study revealed the discriminative ability of key molecules on polyamine biosynthesis as a predictive marker for virulence evaluation and assessment of SARS-CoV-2 variants in cell or organoid models.Our work,therefore,presented a practical strategy that could be potentially applied as an evaluation tool for the pathogenicity of current and emerging SARS-CoV-2 variants.

Increasing the electrode thickness is a significant method to decrease the weight and volume ratio of the inactive components for high energy density of the devices.In this contribution,we extracted a repeating unit in the configurations and establish the empirical energy density model based on some assumptions.In this model,the effects of the electrode thickness on the energy density for lithium-ion batteries(LIBs),lithium metal batteries(LMBs),and anode-free lithium batteries(ALBs)are evaluated quantitively with the current parameters of the batteries.The results demonstrate that the structure evolutions from LIBs,LMBs to ALBs with the reduction of the anode weight contribution,the energy density can be well improved exactly.While the increase of the thickness of the electrode provide another route to furthe r enhance the energydensity by decreasing the weight contribution of inactive materials;meanwhile the effects for ALBs are higher than LMBs and LIBs due to the higher weight ratio of inactive materials.This empirical energy density model is also applied into the practical system and provide intuitional results to guide the battery design for higher energy density.

The Li-Mg-B-H composite(2LiBH_(4)+MgH_(2))has a high capacity of 11.4 wt%as a hydrogen storage material.However,the slow kinetics and poor cycling stability severely restrict its practical applications.In this work,a layered Nb_(2)C MXene was first synthesized and then introduced to tailor the kinetics and cycling stability of the Li-Mg-B-H composite.The milled 2LiH+MgB_(2)composites were initially hydrogenated to obtain the 2LiBH_(4)+MgH_(2)composites.The 2LiBH_(4)+MgH_(2)+5wt%Nb_(2)C can release 9.0 wt%H_(2)in 30 min at 400℃,while it is only 2.7 wt%for the undoped 2LiBH_(4)+MgH_(2).The dehydrogenation activation energies of MgH_(2)and LiBH_(4)are 123 and 154 kJ·mol^(-1)respectively for the 5 wt%Nb_(2)C-doped composite,lower than the undoped composite(164 and 165 kJ·mol^(-1)).The 2LiBH_(4)+MgH_(2)+5 wt%Nb_(2)C possesses excellent cycling stability,with the reversible capacity only slightly reduced from 9.4 wt%for the 1st cycle to 9.3 wt%for the 20th cycle.Nb_(2)C keeps stable in the composite and acts as an efficient catalyst for the Li-Mg-B-H composite.It is believed that both the layered structure and the active Nb element contribu te to the enhanced hydrogen storage performances of the Li-Mg-B-H composite.This work confirms that the Nb_(2)C MXene with layered stru cture has a significant enhancing impact on the Li-Mg-B-H hydrogen storage materials,which is different from the bulk NbC.

在单一颗粒系中,消光光谱法颗粒粒径测量模型通常基于Mie散射理论和Lambert-Beer(LB)定律。但多种颗粒物构成的混合颗粒系的消光特性更为复杂,颗粒粒径与混合比均会影响消光谱,需要发展新的理论模型。作者设计一种蒙特卡罗(Monte Carlo,MC)原理的混合颗粒系消光模型,将入射光束离散化成光子,通过追踪其从发射至接收/逃逸过程中所有事件,统计光子去向并研究混合颗粒系的消光特性。为了验证蒙特卡罗模型的准确性,分别对聚苯乙烯和玻璃微珠单一颗粒系中消光谱进行数值计算,结果与Lambert-Beer定律预测误差小于2%。进一步将模型扩展至由聚苯乙烯和玻璃微珠构成的混合颗粒系,其消光谱随玻璃微珠占比(混合比)增大而依次递增,直至混合比为1时退化为单一颗粒系,而波长减小时,消光随混合比变化趋势由线性向非线性转变,两种颗粒自身消光特性差异越大则非线性趋势越明显,说明混合系消光值由颗粒类型、混合比、颗粒粒径和光波长共同决定且相互耦合。根据预测消光谱,采用三种全局最优算法对混合颗粒粒径和混合比参数反演。结果表明:混合比单参数反演结果的误差最小,均在1.5%以内;对两种颗粒粒径进行双参数反演时,误差均在3%以内;对两种粒径和混合比进行三参数同步反演时,误差增大,但均在10%以内。综合分析三种反演算法,PSO算法耗费时间最长,是其他算法的数倍,IGA算法效果较好且更加稳定。初步验证了设计的蒙特卡罗模型可应用于混合颗粒系的消光谱预测,实现了两种颗粒粒径与颗粒系混合比的同步反演。

Patients with diabetes mellitus have a higher risk of developing Parkinson’s disease(PD). However, the molecular links between PD and diabetes remain unclear.In this study, we investigated the roles of thioredoxininteracting protein(TXNIP) in Parkin/PINK1-mediated mitophagy in dopaminergic(DA) cells under high-glucose(HG) conditions. In streptozotocin-induced diabetic mice,TXNIP was upregulated and autophagy was inhibited in the midbrain, while the loss of DA neurons was accelerated by hyperglycemia. In cultured PC12 cells under HG, TXNIP expression was upregulated and the intracellular reactive oxygen species(ROS) levels increased, leading to cell death. Autophagic flux was further blocked and PINK1 expression was decreased under HG conditions. Parkin expression in the mitochondrial fraction and carbonyl cyanide 3-chlorophenylhydrazone(CCCP)-induced co-localization of COX IV(marker for mitochondria) and LAMP1(marker for lysosomes) were also significantly decreased by HG. Overexpression of TXNIP was sufficient to decrease the expression of both PINK1 and Parkin in PC12 cells, while knockdown of the expression of TXNIP by si RNA decreased intracellular ROS and attenuated cellular injury under HG. Moreover, inhibition of TXNIP improved the CCCP-induced co-localization of COX IV and LAMP1 in PC12 cells under HG. Together, these results suggest that TXNIP regulates Parkin/PINK1-mediated mitophagy under HG conditions, and targeting TXNIP may be a promising therapeutic strategy for reducing the risk of PD under hyperglycemic conditions.

The present study was designed to examine the therapeutic effects of Botulinum neurotoxin A(BoNT/A)on depression-like behaviors in mice and to explore the potential mechanisms.These results revealed that a single facial injection of BoNT/A induced a rapid and prolonged improvement of depression-like behaviors in naive and space-restriction-stressed(SRS)mice,reflected by a decreased duration of immobility in behavioral despair tests.BoNT/A significantly increased the 5-hydroxytryptamine(5-HT)levels in several brain regions,including the hippocampus and hypothalamus,in SRS mice.BoNT/A increased the expression of the N-methyl-Daspartate receptor subunits NR1 and NR2 B in the hippocampus,which were significantly decreased in SRS mice.Furthermore,BoNT/A significantly increased the expression of brain-derived neurotrophic factor(BDNF)in the hippocampus,hypothalamus,prefrontal cortex,and amygdala,which were decreased in SRS mice.Finally,BoNT/A transiently increased the levels of phosphorylated extracellular signal-regulated kinase(p-ERK)and cAMPresponse element binding protein(p-CREB),which were suppressed in the hippocampus of SRS mice.Collectively,these results demonstrated that BoNT/A treatment has antidepressant-like activity in mice,and this is associated with increased 5-HT levels and the activation of BDNF/ERK/CREB pathways in the hippocampus,supporting further investigation of BoNT/A therapy in depression.

目的:评价通督整脊法治疗Tourette综合征临床疗效。方法:选取46例Tourette综合征儿童,随机分为对照组和试验组各23例。对照组予口服盐酸硫必利片规律治疗,试验组予通督整脊法治疗,通过耶鲁综合抽动严重程度量表(YGTSS)、中医证候积分表观察Tourette综合征儿童治疗前后抽动总积分、耶鲁总积分和中医证候积分改善状况。结果:与本组治疗前比较,两组治疗后4周、8周YGTSS评分及中医证候积分均显著下降(P<0.01)。治疗后4周及8周试验组YGTSS改善率及中医证候积分疗效均较对照组显著升高(P<0.01),试验组不良反应更少。结论:通督整脊法治疗Tourette综合征临床疗效可靠、安全性高,值得临床推广应用。
