Dear Editor,COVID-19 inactivated vaccines have been extensively administered inChina.However,the majority of the Chinese population has experiencedbreakthrough infections from SARS-CoV-2 ancestral,Delta or Omicronvariants over the past three years,particularly during the wave of Omicron BA.5 and BA.7 variants at the end of 2022(Zhu et al.,2023).Subsequently,new Omicron variants,such as BQ.1,BQ.1.1,XBB,XBB.1/XBB.1.9,and XBB.1.5/XBB.1.9.1,are emerging in China(Yueetal.,2023;Zhu et al.,2023).Therefore,it is an urgentneed and a publichealth imperative to assess the extent of the immunoprotection established in this population.

BACKGROUND The number of patients undergoing solid organ transplantation has increased annually.However,infections in solid organ transplant recipients can have a severe effect on patient survival owing to the continued use of immunosuppressants.Carrimycin is a novel macrolide antibiotic produced by genetically engineered streptomyces spiramyceticus harboring a 4’’-O-isovaleryltransferase gene(ist)from streptomyces thermotoleran.Carrimycin has good antibacterial and antiviral effects.However,no relevant studies have been conducted on the efficacy and safety of carrimycin in patients with severe pneumonia(SP)after solid organ transplantation.AIM To explore the efficacy and safety of carrimycin in patients with SP after solid organ transplantation to provide a medication reference for clinical treatment.METHODS In March 2022,ten patients with SP following solid-organ transplantation were treated at our hospital between January 2021 and March 2022.When the condition was critical and difficult to control with other drugs,carrimycin was administered.These ten patients'clinical features and treatment protocols were retrospectively analyzed,and the efficacy and safety of carrimycin for treating SP following solid organ transplantation were evaluated.RESULTS All ten patients were included in the analysis.Regarding etiological agent detection,there were three cases of fungal pneumonia,two cases of bacterial pneumonia,two cases of Pneumocystis pneumonia,and three cases of mixed infections.After treatment with carrimycin,the disease in seven patients significantly improved,the course of the disease was significantly shortened,fever was quickly controlled,chest computed tomography was significantly improved,and oxygenation was significantly improved.Finally,the patients were discharged after curing.One patient died of acute respiratory distress syndrome,and two patients discontinued treatment.CONCLUSION Carrimycin is a safe and effective treatment modality for SP following solid organ transplantation.Carrimycin may have anti

BACKGROUND Colorectal cancer has a low 5-year survival rate and high mortality.Humanβ-defensin-1(hBD-1)may play an integral function in the innate immune system,contributing to the recognition and destruction of cancer cells.Long non-coding RNAs(lncRNAs)are involved in the process of cell differentiation and growth.AIM To investigate the effect of hBD-1 on the mammalian target of rapamycin(mTOR)pathway and autophagy in human colon cancer SW620 cells.METHODS CCK8 assay was utilized for the detection of cell proliferation and determination of the optimal drug concentration.Colony formation assay was employed to assess the effect of hBD-1 on SW620 cell proliferation.Bioinformatics was used to screen potentially biologically significant lncRNAs related to the mTOR pathway.Additionally,p-mTOR(Ser2448),Beclin1,and LC3II/I expression levels in SW620 cells were assessed through Western blot analysis.RESULTS hBD-1 inhibited the proliferative ability of SW620 cells,as evidenced by the reduction in the colony formation capacity of SW620 cells upon exposure to hBD-1.hBD-1 decreased the expression of p-mTOR(Ser2448)protein and increased the expression of Beclin1 and LC3II/I protein.Furthermore,bioinformatics analysis identified seven lncRNAs(2 upregulated and 5 downregulated)related to the mTOR pathway.The lncRNA TCONS_00014506 was ultimately selected.Following the inhibition of the lncRNA TCONS_00014506,exposure to hBD-1 inhibited p-mTOR(Ser2448)and promoted Beclin1 and LC3II/I protein expression.CONCLUSION hBD-1 inhibits the mTOR pathway and promotes autophagy by upregulating the expression of the lncRNA TCONS_00014506 in SW620 cells.

Although inorganic pigments in common spectral tuning materials show good weatherability and heat resistance,the limited color choices,weak coloring power,poor dispersibility,and a possibility of toxicity limit their development.On the basis of organic pigments which possess a wide range of colors,high coloring power,good transparency,and high safety,herein,the modified pigment and biomimetic coating with improved weatherability,especially ultraviolet(UV) resistance(from 2 to 6 days),was achieved by intercalating acid green 25(AG25) pigment into Mg/Al-layered double hydroxides(Mg/Al-LDH).Furthermore,the heat resistance of AG25 was also significantly increased.Moreover,the spectral stability of pigments after heat treatment is superior with almost unchanged spectral profile and green reflection peak.The formation of strong N-H bonds and the S-M(Mg,Al)bonds between Mg/Al-LDH laminates and AG25 molecules contributes to the improvement.This work shows potential for biomimetic leaf materials in respect of reflective spectra stability.

再生水农业利用被认为是解决水资源短缺和减少水环境污染的有效途径.硅肥可提高作物产量及品质,并增强作物抗逆能力.叶面喷施硅肥对叶际微生物群落的影响仍缺少全面研究.通过盆栽试验,探讨了不同种类硅肥叶面喷施对再生水灌溉水稻叶际细菌群落结构组成及多样性和相关功能基因丰度的影响.结果表明,水稻叶际细菌优势菌门主要为厚壁菌门(Firmicutes)、变形菌门(Proteobacteria)、放线菌门(Actinobacteriota)、拟杆菌门(Bacteroidota)和疣微菌门(Verrucomicrobiota).芽孢杆菌属(Bacillus)相对丰度较其他处理在再生水灌溉+叶面喷施那优硅(RIS3)处理中更高,再生水灌溉使潜在致病菌泛菌属(Pantoea)和肠杆菌属(Enterobacter)的相对丰度显著增加,其中未分类的菌属也是水稻叶际细菌群落重要的组成部分.硅肥处理显著富集了芽孢杆菌属(Bacillus)、微小杆菌属(Exiguobacterium)、Aeromonas和柠檬酸杆菌属(Citrobacter).功能预测分析表明,指示物种主要参与代谢与降解功能,并且预测的功能类群主要来自化能异养、好氧化能异养、硝酸盐还原和发酵.定量PCR结果显示,各处理中氨氧化古菌(AOA)、氨氧化细菌(AOB)和固氮酶基因(nifH)均处于较低的丰度水平,亚硝酸还原酶基因(nirK)丰度在各处理间也无显著差异.上述研究结果有助于深层次了解叶面喷施硅肥影响水稻叶际细菌群落结构及多样性差异,为硅肥在再生水灌溉农业中的应用提供理论依据.

Design and assembly of a nested imaging X-ray telescope for the Hot Universe Baryon Surveyor mission

The Hot Universe Baryon Surveyor (HUBS) mission will carry a nested X-ray telescope capable of observing an energy range from 0.5 keV to 2 keV to study hot baryon evolution. In this paper, we report the latest progress in the design and construction of nested X-ray telescopes which were designed to use a three-stage conic-approximation type assembly to simplify the manufacturing process. The mirror substrate is made using the thermal glass slumping method, with mirrors characterized by a root-mean-square roughness of 0.3 nm, with expected high reflectivity and good thermal stability. We also discuss methods of telescope construction and conduct a deformation analysis of the manufactured mirror. The in situ measurement system program is developed to guide the telescope assembly process.

为正确模拟土体涉及剪切带演化的后失效力学响应,需采用包含细观特征长度的高阶连续介质力学模型.笔者利用前期所建立的微极亚塑性模型,对颗粒土中剪切带的发展过程进行了分析推导,得到了剪切带临界状态条件下关键变量所满足的非线性微分方程.该文展示了上述非线性微分方程的简要推导,重点讨论了该非线性微分方程的主要性质、主要参数变化范围和求解途径;通过对剪切带进一步的力学分析补充建立了一个能量方程,使问题具有确定解.在此基础上,应用数值积分求出了剪切带厚度因子和剪切内应力、变形率分布及剪切速度分布的完全解.其中剪切带厚度因子对于微极亚塑性模型细观参数的确定具有重要作用.

锂离子电池健康状态(SOH)的准确估计对于保障电池系统安全运行具有重要意义.针对传统SOH估计方法在可变工况下失效的问题,提出了一种基于等效电路模型和稀疏高斯过程回归的锂离子电池SOH在线估计方法.通过两个在线滤波器,在恒流充电过程中动态地辨识了锂离子电池等效电路模型的各项参数,构建了工况不敏感的健康因子,结合稀疏高斯过程回归实现SOH的间接估计.该方法在多种工况下使用统一的信号处理方法和特征映射模型,兼具鲁棒性强和冗余度低的优点.实验结果表明,该方法在多种工况下的平均绝对误差不超过0.94%,均方根误差不超过1.12%,与现有方法相比,该方法在综合性能上具有显著优势.

Background: Mucosal-associated invariant T(MAIT) cells are systemically depleted in human immunodeficiency virus type 1(HIV-1) infected patients and are not replenished even after successful combined antiretroviral therapy(cART).This study aimed to identify the mechanism underlying MAIT cell depletion.Methods: In the present study, we applied flow cytometry, single-cell RNA sequencing and immunohistochemical staining to evaluate the characteristics of pyroptotic MAIT cells in a total of 127 HIV-1 infected individuals, including 69 treatment-naive patients, 28 complete responders, 15 immunological non-responders, and 15 elite controllers, at the Fifth Medical Center of Chinese PLA General Hospital, Beijing, China.Results: Single-cell transcriptomic profiles revealed that circulating MAIT cells from HIV-1 infected subjects were highly activated, with upregulation of pyroptosis-related genes. Further analysis revealed that increased frequencies of pyroptotic MAIT cells correlated with markers of systemic T-cell activation, microbial translocation, and intestinal damage in cART-naive patients and poor CD4+ T-cell recovery in long-term cART patients. Immunohistochemical staining revealed that MAIT cells in the gut mucosa of HIV-1 infected patients exhibited a strong active gasdermin-D(GSDMD, marker of pyroptosis) signal near the cavity side, suggesting that these MAIT cells underwent active pyroptosis in the colorectal mucosa. Increased levels of the proinflammatory cytokines interleukin-12(IL-12) and IL-18 were observed in HIV-1 infected patients. In addition, activated MAIT cells exhibited an increased pyroptotic phenotype after being triggered by HIV-1 virions, T-cell receptor signals, IL-12 plus IL-18, and combinations of these factors, in vitro.Conclusions: Activation-induced MAIT cell pyroptosis contributes to the loss of MAIT cells in HIV-1 infected patients,which could potentiate disease progression and poor immune reconstitution.

云边端协同架构中数据类型多样,各级存储资源与计算资源存在差异,给数据管理带来新的挑战.现有数据模型或者数据模型的简单叠加,都难以同时满足云边端中多模态数据管理和协同管理需求.因此,研究面向云边端协同的多模态数据建模技术成为重要问题.其核心在于,如何高效地从云边端三层架构中得到满足应用所需的查询结果.从云边端三层数据的数据类型出发,提出了面向云边端协同的多模态数据建模技术,给出了基于元组的多模态数据模型定义,设计了6种基类,解决多模态数据统一表征困难的问题;提出了云边端协同查询的基本数据操作体系,以满足云边端业务场景的查询需求;给出了多模态数据模型的完整性约束,为查询优化奠定了理论基础.最后,给出了面向云边端协同多模态数据模型的示范应用,并从数据存储时间、存储空间和查询时间这3个方面对所提出的数据模型存储方法进行了验证.实验结果表明,所提方案能够有效地表示云边端协同架构中的多模态数据.
