Acupotomy alleviates knee osteoarthritis in rabbit by regulating chondrocyte mitophagy via Pink1-Parkin pathway

OBJECTIVE: To investigate the effect of acupotomy, on mitophagy and the Pink1-Parkin pathway in chondrocytes from rabbits with knee osteoarthritis(KOA).METHODS: A KOA model was established via the modified Videman method. Rabbits were randomly divided into a control group(CON), KOA group and KOA + acupotomy group(Acu). Rabbits in the acupotomy group were subjected to acupotomy for 4 weeks after model establishment. The behavior of the rabbits before and after intervention was recorded. Cartilage degeneration was evaluated by optical microscopy and fluorescence microscopy. The level of mitophagy was evaluated by transmission electron microscopy,immunofluorescence and enzyme-linked immunosorbent assay(ELISA). The expression of phosphatase and tensin homolog(PTEN)-induced kinase 1(Pink1)-Parkin mitophagy pathway components was evaluated by immunofluorescence, Western blotting and real-time polymerase chain ***: In rabbits with KOA, joint pain, mobility disorders and cartilage degeneration were observed, the Mankin score was increased, collagen type Ⅱ(Col-Ⅱ)expression was significantly decreased, mitophagy was inhibited, mitochondrial function was impaired, and factors associated with the Pink1-Parkin pathway were inhibited. Acupotomy regulated the expression of Pink1-Parkin pathway-related proteins, the mitophagy-related protein microtubule-associated protein-1 light chain-3,the translocase of the outer membrane, and the inner mitochondrial membrane 23; increased the colocalization of mitochondria and autophagosomes; promoted the removal of damaged mitochondria;restored mitochondrial adenosine-triphosphate(ATP) production;and alleviated cartilage degeneration in rabbits with ***: Acupotomy played a role in alleviating KOA in rabbits by activating mitophagy in chondrocytes via the regulation of proteins that are related to the Pink1-Parkin pathway.

Body size induced changes in metabolic carbon of soil nematodes under N deposition and precipitation regime change in a temperate grassland

Background Global climate change has resulted in precipitation regimes exhibiting an increasing trend in rainfall intensity but a reduction in its frequency. Nitrogen(N) deposition is a crucial component of the global N cycling. Nematode body size is a trait that responds to climate change and is used as a standard trait-based indicator in soil community analysis. Variations in body size influence metabolic carbon(C). We examined the ways by which body size and metabolic C of nematodes respond to changing precipitation regimes and how N deposition regulates these responses by an 8-year manipulative *** Nematode body size was indicated by the community-weighted mean(CWM) mass. We quantified C metabolism components of soil nematodes including production C, respiration C, and corresponding C use efficiency(CUE) under different precipitation intensities and N addition in a semi-arid steppe on the Mongolian Plateau. The Mantel test was used to determine the correlations between CWM, CUE and environmental factors. The partial least squares path modeling(PLS-PM) was conducted to quantify direct or indirect contributions among latent *** We found that heavy precipitation intensity increased the CWM mass of total nematodes and omnivorespredators without N addition. N addition decreased CWM mass of bacterivores across all the precipitation intensity treatments. Stronger precipitation intensities might be favorable for nematode production and respiration C. Small cell lung cancer(SCLC)is a highly malignant tumor with a very poor prognosis;therefore,more effective treatments are urgently needed for patients afflicted with the *** recent years,emerging molecular classifications based on key transcription factors of SCLC have provided more information on the tumor pathophysiology,metastasis,immune microenvironment,and acquired therapeutic resistance and reflected the intertumoral heterogeneity of the various SCLC ***,advances in genomics and single-cell sequencing analysis have further revealed the high intratumoral heterogeneity and plasticity of the ***,we review and summarize these recent lines of evidence and discuss the possible pathogenesis of SCLC.

Manufacturing CAR-NK against tumors: Who is the ideal supplier?

Chimeric antigen receptor-natural killer(CAR-NK) cells have emerged as another prominent player in the realm of tumor immunotherapy following CAR-T cells. The unique features of CAR-NK cells make it possible to compensate for deficiencies in CAR-T therapy, such as the complexity of the manufacturing process, clinical adverse events, and solid tumor challenges. To date, CAR-NK products from different allogeneic sources have exhibited remarkable anti-tumor effects on preclinical studies and have gradually been applied in clinical ***, each source has advantages and disadvantages. Selecting a suitable source may help maximize CAR-NK cell efficacy and increase the feasibility of clinical transformation. Therefore, this review discusses the development and challenges of CAR-NK cells from different sources to provide a reference for future exploration.

Resonant four-photon photoemission from SnSe_(2)(001)

High-order nonlinear multiphoton absorption is usually inefficient,but can be enhanced by designing resonant excitations between occupied and unoccupied energy *** conducted angle-resolved multi-photon photoemission(mPPE)studies on the SnSe_(2)(001)surfaces excited by ultrashort laser *** tuning photon energy and light polarization,we demonstrate the presence of a resonant four-photon photoemission(4PPE)process involving the occupied valence band(VB),the unoccupied second conduction band(CB2)and the unoccupied image-potential state(IPs)of SnSe_(2).In this 4PPE process,VB electrons of SnSe_(2) are resonantly excited into CB2 by adsorbing two photons,followed by the adsorption of another photon to populate the n=1 IPs before being emitted out to the vacuum by adsorbing one more *** results in a double-resonant 4PPE process,which exhibits approximately a 40 times enhancement in photoemission yields compared to cases where one of the resonant pathways,CB2→IPs,is inhibited by involving a virtual state instead of the IPs in the *** double-resonant 4PPE process efficiently excite the bulk VB electrons outside the vacuum,like taking advantage of resonant“ladders”through two real empty electronic states of SnSe_(2).Our results highlight the important applications of mPPE in probing the band-structure,particularly the unoccupied states,of recently emerging main group dichalcogenide ***,the discovered resonant mPPE process contributes to the exploration of their promising optoelectronic applications.

Cosmological interpretation for the stochastic signal in pulsar timing arrays

The pulsar timing array(PTA)collaborations have recently reported compelling evidence for a stochastic signal consistent with a gravitational-wave *** this paper,we combine the latest data sets from NANOGrav,PPTA,and EPTA to explore cosmological interpretations for the PTA signal from first-order phase transitions,domain walls,and cosmic strings,*** find that the domain wall model is strongly disfavored with the Bayes factors compared with the first-order phase transitions and cosmic strings being 1/90 and 1/189,respectively,breaking the degeneracy among these models in individual data *** also find that:(1)a strong phase transition at temperatures below the electroweak scale is favored,and the bubble collisions make the dominant contribution to the energy density spectrum;(2)a small reconnection probability p<1.55×10^(-1)allowed by strings in(super)string theory is favored at the 95%confidence level,and ground-based detectors can further constrain the parameter space.

Facile synthesis of composite polyferric magnesium-silicate-sulfate coagulant with enhanced performance in water and wastewater

The coagulation process is a widely applied technology in water and wastewater *** composite polyferric mag-nesium-silicate-sulfate(PFMS)coagulants were synthesized using Na_(2)SiO_(3)·9H_(2)O,Fe_(2)(SO_(4))_(3),and MgSO_(4) as raw materials in this *** effects of aging time,Fe:Si:Mg,and OH:M molar ratios(M represents the metal ions)on the coagulation performance of the as-pre-pared PFMS were systematically investigated to obtain optimum *** results showed that PFMS coagulant exhibited good co-agulation properties in the treatment of simulated humic acid-kaolin surface water and reactive dye *** the molar ratio was controlled at Fe:Si:Mg=2:2:1 and OH:M=0.32,the obtained PFMS presented excellent stability and a high coagulation *** removal efficiency of ultraviolet UV254 was 99.81%,and the residual turbidity of the surface water reached 0.56 NTU at a dosage of 30 mg·L^(-1).After standing the coagulant for 120 d in the laboratory,the removal efficiency of UV254 and residual turbidity of the surface wa-ter were 88.12%and 0.68 NTU,respectively,which accord with the surface water treatment *** addition,the coagulation performance in the treatment of reactive dye wastewater was greatly improved by combining the advantages of magnesium and iron *** with polyferric silicate-sulfate(PFS)and polymagnesium silicate-sulfate(PMS),the PFMS coagulant played a better decolor-ization role within the pH range of 7-13.
